When the different parts of a structure are modelled independently by BEM or FEM methods, it is sometimes necessary to put the parts together without remeshing of the nodes along the part interfaces. Frequently the nodes do not match along the interface. In this work, the symmetric Galerkin multi‐zone curved boundary element is a fully symmetric formulation and is the method used for the boundary element part. For BEM–FEM coupling it is then necessary to interpolate the tractions in‐between the non‐matching nodes for the FEM part. Finally, the coupling is achieved by transforming the finite element domains to equivalent boundary element domains in a block symmetric formulation. A typical quadruple node (QN) of triple junctions (TJs) of grain boundaries (GBs) in a Eu2+‐doped KCl0.52Br0.48 solid solution is investigated from the geometrical point of view by epifluorescence microscopy using the doping ion as a fluorochrome. The excitation and fluorescence optical properties of the fluorochrome were previously characterised by spectrophotometry whereas the structural nature of the studied material as well as its Bravais lattice type, unit cell size and long‐range translational order degree was determined by X‐ray diffraction. A three‐dimensional reconstruction was built from the microscopy images of different optical cross‐sections of the studied arrangement of crystal defects. In the close vicinity of the QN, the studied arrangement of crystal defects adopts the geometry of a collapsed tristetrahedron which, centred at the QN, has its legs along the TJs and, hence, has its faces as collapsed in pairs into the GBs. The angles defined by different TJ couples as well as the dihedral angles defined by the different GB couples meeting in every TJ were measured at the QN site. All, the image recording and stacking as well as the measuring procedures are carefully described. The measured TJ angles (97°, 117°, 95°, 117°, 99° and 130° ± 2°) depart from the characteristic angle (109.47°) of a tetrahedron whereas the measured GB angles (101°, 119°, 140°; 125°, 127°, 108°; 133°, 109°, 119°; 129°, 99° and 132° ± 2°) depart from the angular argument (120°) of a 3‐fold symmetry rotation indicating that, in the close neighbourhood of the QN, the studied arrangement of crystal defects is structurally unstable. Such an instability is associated with an observed mismatch in orientation (by angles of 20°, 15°, 33° and 30° ± 2°) between the TJs and some <111> zone axis matrix lattice crystallographic directions ([], [11], [11] and [11]), respectively). Local variations in anionic composition, existing within the solid solution matrix, are discussed to be responsible for this mismatching and, therefore, for the observed structural instability. 相似文献

Recently, we identified 6 genomic loci affecting daughter yield deviations (DYD) for somatic cell score (SCS) in a genome-wide association study (GWAS) performed with German Holstein bulls. In the current study, we tested if these loci were associated with SCS in cows using their own performance data. The study was performed with 1,412 German Holstein cows, of which 483 were daughters of 71 bulls that had been used in the GWAS. We tested 10 single nucleotide polymorphisms (SNP) representing 6 genomic regions that were associated with DYD for SCS in bulls. All tested SNP were significant in cows. Seven of them, located on Bos taurus autosomes (BTA) 6, 13, and 19, had the same direction of effect as those previously reported in the bull population. The most significant associations were detected on BTA6 and BTA19, accounting for 1.8% of the total genetic variance. The major allele of the 2 SNP on BTA6 and the minor allele of the 2 SNP on BTA19 were favorable for lower SCS. The differences between the homozygous genotype classes were up to 15,000 cells/mL. The verification of SNP associated with SCS in this study provides further evidence for the functional role of the linked genomic regions for immune response and contributes to identification of causative mutations. In particular, SNP with minor frequency of the favorable allele possess high potential to reduce SCS in German Holstein cattle by selection. 相似文献

The aim of this study was to integrate multi omics data to characterize underlying functional pathways and candidate genes for drip loss in pigs. The consideration of different omics levels allows elucidating the black box of phenotype expression. Metabolite and protein profiling was applied in Musculus longissimus dorsi samples of 97 Duroc × Pietrain pigs. In total, 126 and 35 annotated metabolites and proteins were quantified, respectively. In addition, all animals were genotyped with the porcine 60 k Illumina beadchip. An enrichment analysis resulted in 10 pathways, amongst others, sphingolipid metabolism and glycolysis/gluconeogenesis, with significant influence on drip loss. Drip loss and 22 metabolic components were analyzed as intermediate phenotypes within a genome-wide association study (GWAS). We detected significantly associated genetic markers and candidate genes for drip loss and for most of the metabolic components. On chromosome 18, a region with promising candidate genes was identified based on SNPs associated with drip loss, the protein “phosphoglycerate mutase 2” and the metabolite glycine. We hypothesize that association studies based on intermediate phenotypes are able to provide comprehensive insights in the genetic variation of genes directly involved in the metabolism of performance traits. The problem of finding the expected shortest path in stochastic networks, where the presence of each node is probabilistic and the arc lengths are random variables, have numerous applications, especially in communication networks. The problem being NP-hard we use an ant colony system (ACS) to propose a metaheuristic algorithm for finding the expected shortest path. A new local heuristic is formulated for the proposed algorithm to consider the probabilistic nodes. The arc lengths are randomly generated based on the arc length distribution functions. Background: The assessment of human epidermal growth factor receptor 2 (HER2) gene amplification is essential in order to identify those patients affected by advanced gastric cancer who may benefit from Trastuzumab targeted therapy. Materials and Methods: With the aim to investigate the concordance rate in HER2 status between primary gastric carcinoma (GC) and synchronous lymphnode metastases, we investigated HER2 status in a cohort of 108 surgical formalin-fixed paraffin-embedded specimens of GC and matched synchronous metastatic lymph nodes collected from three different units of Anatomic Pathology in southern of Italy. Fleiss-Cohen weighted k statistics were used to assess the concordance rate of HER2 status. Results: HER2 amplification was observed in 17% of primary GCs and the overall concordance rate with corresponding nodal metastases was 90.74%. Changes in HER2 status between primary GC and matched synchronous metastases were evidenced in 10 (9.26%) cases. Of these, 6 cases were HER2 amplified in the primary GC and not amplified in the metastases, while 4 were HER2 not amplified in the primary tumour and amplified in the lymph node metastases. Conclusions: Although at present the simultaneous determination of HER2 in advanced gastric cancer and corresponding metastatic lymph nodes is not mandatory, the possibility that the synchronous metastases of GC have a different HER2 status from that of the primary tumour is of remarkable significance; Indeed this may have influence on the therapeutic management and prognosis of the patients. 相似文献

Listeria monocytogenes (Lm) bacterial ghosts (LMGs) were produced by the minimum inhibitory concentration (MIC) of HCl, H2SO4, and NaOH. Acid and alkali effects on the LMGs were compared by in vitro and in vivo analyses. Scanning electron microscope showed that all chemicals form lysis pores on the Lm cell envelopes. Real-time qPCR revealed a complete absence of genomic DNA in HCl- and H2SO4-induced LMGs but not in NaOH-induced LMGs. HCl-, H2SO4- and NaOH-induced LMGs showed weaker or missing protein bands on SDS-PAGE gel when compared to wild-type Lm. Murine macrophages exposed to the HCl-induced LMGs showed higher cell viability than those exposed to NaOH-induced LMGs or wild-type Lm. The maximum level of cytokine expression (TNF-α, iNOS, IFN-γ, and IL-10 mRNA) was observed in the macrophages exposed to NaOH-induced LMGs, while that of IL-1β mRNA was observed in the macrophages exposed to HCl-induced LMGs. To investigate LMGs as a vaccine candidate, mice were divided into PBS buffer-injected, HCl- and NaOH-induced LMGs immunized groups. Mice vaccinated with HCl- and NOH-induced LMGs, respectively, significantly increased in specific IgG antibodies, bactericidal activities of serum, and CD4+ and CD8+ T-cell population. Antigenic Lm proteins reacted with antisera against HCl- and NOH-induced LMGs, respectively. Bacterial loads in HCl- and NaOH-induced LMGs immunized mice were significantly lower than PBS-injected mice after virulent Lm challenges. It suggested that vaccination with LMGs induces both humoral and cell-mediated immune responses and protects against virulent challenges. 相似文献
