Pancreatic ductal adenocarcinoma (PDAC), a highly aggressive malignancy with a poor prognosis is usually detected at the advanced stage of the disease. The only US Food and Drug Administration-approved biomarker that is available for PDAC, CA 19-9, is most useful in monitoring treatment response among PDAC patients rather than for early detection. Moreover, when CA 19-9 is solely used for diagnostic purposes, it has only a recorded sensitivity of 79% and specificity of 82% in symptomatic individuals. Therefore, there is an urgent need to identify reliable biomarkers for diagnosis (specifically for the early diagnosis), ascertain prognosis as well as to monitor treatment response and tumour recurrence of PDAC. In recent years, proteomic technologies are growing exponentially at an accelerated rate for a wide range of applications in cancer research. In this review, we discussed the current status of biomarker research for PDAC using various proteomic technologies. This review will explore the potential perspective for understanding and identifying the unique alterations in protein expressions that could prove beneficial in discovering new robust biomarkers to detect PDAC at an early stage, ascertain prognosis of patients with the disease in addition to monitoring treatment response and tumour recurrence of patients. 相似文献

The study of numerical abilities, and how they are acquired, is being used to explore the continuity between ontogenesis and environmental learning. One technique that proves useful in this exploration is the artificial simulation of numerical abilities with neural networks, using different learning paradigms to explore development. A neural network simulation of subitization, sometimes referred to as visual enumeration, and of counting, a recurrent operation, has been developed using the so-called multi-net architecture. Our numerical ability simulations use two or more neural networks combining supervised and unsupervised learning techniques to model subitization and counting. Subitization has been simulated using networks employing unsupervised self-organizing learning, the results of which agree with infant subitization experiments and are comparable with supervised neural network simulations of subitization reported in the literature. Counting has been simulated using a multi-net system of supervised static and recurrent backpropagation networks that learn their individual tasks within an unsupervised, competitive framework. The developmental profile of the counting simulation shows similarities to that of children learning to count and demonstrates how neural networks can learn how to be combined together in a process modelling development. 相似文献

Using the high-speed camera the time sequences of the classical flow patterns of horizontal gas-liquid pipe flow are recorded, from which the average gray-scale values of single-frame images are extracted. Thus obtained gray-scale time series is decomposed by the Empirical Mode Decomposition (EMD) method, the various scales of the signals are processed by Hurst exponent method, and then the dual-fractal characteristics are obtained. The scattered bubble and the bubble cluster theories are applied to the evolution analysis of two-phase flow patterns. At the same time the various signals are checked in the chaotic recursion chart by which the two typical characteristics (diagonal average length and Shannon entropy) are obtained. Resulting term of these properties, the dynamic characteristics of gas-liquid two-phase flow patterns are quantitatively analyzed. The results show that the evolution paths of gas-liquid two-phase flow patterns can be well characterized by the integrated analysis on the basis of the gray-scale time series of flowing images from EMD, Hurst exponents and Recurrence Plot (RP). In the middle frequency section (2nd, 3rd, 4th scales), three flow patterns decomposed by the EMD exhibit dual fractal characteristics which represent the dynamic features of bubble cluster, single bubble, slug bubble and scattered bubble. According to the change of diagonal average lengths and recursive Shannon entropy characteristic value, the structure deterministic of the slug flow is better than the other two patterns. After the decomposition by EMD the slug flow and the mist flow in the high frequency section have obvious peaks. Anyway, it is an effective way to understand and characterize the dynamic characteristics of two-phase flow patterns using the multi-scale non-linear analysis method based on image gray-scale fluctuation signals. 相似文献

引入递归图分析方法对具有混沌与分形特性的气-固循环流化床压力脉动信号进行分析,针对Logisitc模型比较了递归图特征量递归率、确定性、平均对角线长度、分叉性与最大Lyapunov指数之间的关系,得出了递归图特征量对于混沌特性具有表征作用的结论。研究了递归图特征量在气-固循环流化床不同床型下的变化规律,递归图特征量在固定床型、湍流床型、气力输送床型下保持稳定,在鼓泡床型、快速流化床型下分别迅速递增或递减,因此递归图特征量同样可以对循环流化床不同床型进行辨识。 相似文献

Cancer-associated fibroblasts (CAFs) are one of the most abundant and critical components of the tumor stroma. CAFs can impact many important steps of cancerogenesis and may also influence treatment resistance. Some of these effects need the direct contact of CAFs and cancer cells, while some involve paracrine signals. In this study, we investigated the ability of head and neck squamous cell carcinomas (HNSCC) patient-derived CAFs to promote or inhibit the colony-forming ability of HNSCC cells. The effect of cisplatin on this promoting or inhibiting influence was also studied. The subsequent analysis focused on changes in the expression of genes associated with cancer progression. We found that cisplatin response in model HNSCC cancer cells was modified by coculture with CAFs, was CAF-specific, and different patient-derived CAFs had a different “sensitizing ratio”. Increased expression of VEGFA, PGE2S, COX2, EGFR, and NANOG in cancer cells was characteristic for the increase of resistance. On the other hand, CCL2 expression was associated with sensitizing effect. Significantly higher amounts of cisplatin were found in CAFs derived from patients who subsequently experienced a recurrence. In conclusion, our results showed that CAFs could promote and/or inhibit colony-forming capability and cisplatin resistance in HNSCC cells via paracrine effects and subsequent changes in gene expression of cancer-associated genes in cancer cells. 相似文献

Lipofilling (LF) is a largely employed technique in reconstructive and esthetic breast surgery. Over the years, it has demonstrated to be extremely useful for treatment of soft tissue defects after demolitive or conservative breast cancer surgery and different procedures have been developed to improve the survival of transplanted fat graft. The regenerative potential of LF is attributed to the multipotent stem cells found in large quantity in adipose tissue. However, a growing body of pre-clinical evidence shows that adipocytes and adipose-derived stromal cells may have pro-tumorigenic potential. Despite no clear indication from clinical studies has demonstrated an increased risk of cancer recurrence upon LF, these observations challenge the oncologic safety of the procedure. This review aims to provide an updated overview of both the clinical and the pre-clinical indications to the suitability and safety of LF in breast oncological surgery. Cellular and molecular players in the crosstalk between adipose tissue and cancer are described, and heterogeneous contradictory results are discussed, highlighting that important issues still remain to be solved to get a clear understanding of LF safety in breast cancer patients. 相似文献

For non-small cell lung cancer (NSCLC), radiotherapy (RT) and platinum-based chemotherapy (CHT) are among the main treatment options. On the other hand, radioresistance and cytotoxic drug resistance are common causes of failure. The epidermal growth factor receptor (EGFR) plays an important role in radioresponse and therapy resistance. We hypothesized that single nucleotide polymorphisms (SNPs) in the EGFR gene might affect individual sensitivity to these treatments, and thus, therapy outcome and prognosis. The association between functional EGFR SNPs and overall (OS), locoregional recurrence-free (LFRS), and metastasis-free (MFS) survival was examined in 436 patients with unresectable NSCLC receiving RT and platinum-based CHTRT. In a multivariate analysis, the rs712830 CC homozygotes showed reduced OS in the whole group (p = 0.039) and in the curative treatment subset (p = 0.047). The rs712829 TT genotype was strongly associated with decreased LRFS (p = 0.006), and the T-C haplotype was a risk factor for locoregional recurrence in our patients (p = 0.003). The rs2227983 GG alone and in combination with rs712829 T was an indicator of unfavorable LRFS (p = 0.028 and 0.002, respectively). Moreover, significant independent effects of these SNPs on OS, LRFS, and MFS were observed. Our results demonstrate that inherited EGFR gene variants may predict clinical outcomes in NSCLC treated with DNA damage-inducing therapy. 相似文献
