Cardiovascular diseases, such as heart attack and stroke, are the leading causes of death in North America. According to data provided by the World Health Organization in 2000, heart diseases are responsible for 29% of the total mortality rate in the world. Since cardiovascular diseases have a major impact on an individual’s quality of life, a tremendous amount of research has been done in the area of prevention and the treatment of the diseases. Fibrinolytic enzymes are agents that dissolve fibrin clots. Recently many food derived fibrinolytic enzymes have been found in various traditional Asian foods. Fibrinolytic enzymes can be found in a variety of foods, such as Japanese Natto, Tofuyo, Korean Chungkook-Jang soy sauce, and edible honey mushroom. Enzymes have been purified from these foods, and their physiochemical properties have been characterized. Fermented shrimp paste, a popular Asian seasoning, was shown to have strong fibrinolytic activity. These novel fibrinolytic enzymes derived from traditional Asian foods are useful for thrombolytic therapy. They will provide an adjunct to the costly fibrinolytic enzymes that are currently used in managing heart disease, since large quantities of enzyme can be conveniently and efficiently produced. In addition, these enzymes have significant potential for food fortification and nutraceutical applications, such that their use could effectively prevent cardiovascular diseases. 相似文献

Through a mechanism called quorum sensing, bacteria are able to express specific genes in response to population density. Cell-to-cell communication in bacteria is mediated by signal molecules such as acylated homoserine lactones (AHLs). This work aimed to detect AHL production in Gram-negative psychrotrophic bacteria isolated from raw milk. A total of 84.9% of the bacteria were identified as AHL producers eliciting a diversity of responses in the AHL-monitor systems. These results demonstrate that AHL-production is common among psychrotrophic bacteria isolated from milk, and indicate that quorum sensing may play an important role in the spoilage of this product. 相似文献

A facile approach has been developed to synthesize Fe3O4@PAM (polyacrylamide) nanoparticles (NPs) with carboxyl groups on the surfaces by copolymerization with acrylamide and acrylic acid in Fe3O4 NPs aqueous suspension. Nitrilotriacetic acid (NTA) was conjugated to the magnetic NPs via well-known carboniimide chemistry using EDC and NHS. The Ni2+ ions loaded on the surface of NPs provide abundant docking sites for immobilization of His-tagged green fluorescent proteins (His-tagged GFP). The high magnetic property of Fe3O4@PAM@NTA-Ni2+ allows an easy separation of the NPs from solution under an external magnetic field, with high His-tagged protein binding capacity (42 μg protein/mg of NPs). The NPs can be recycled for at least four times without significant loss of binding capacity to proteins. A TaqMan real-time polymerase chain reaction (PCR) method was developed for specific detection of bovine, ovine and caprine processed animal protein (PAP) in industrial feedstuffs. The method uses species-specific primers and probes targeting short mitochondrial D-loop sequences, and a positive amplification control based on 18S rRNA gene. The applicability of the real-time PCR protocol was assessed through analysis of 126 industrial feed samples that were manufactured to reproduce rendering processes of commercial feeds destined for farmed animals. The assay successfully classified samples as positive or negative according to the ruminant composition, enabling qualitative detection of banned material in feeds at levels as low as 0.1%. Although the method provides quantitative potential, results suggest that the real quantitative capability of the assay is limited by the existing variability in terms of composition and processing treatments of the feeds, which affect the amount and quality of amplifiable DNA. 相似文献

The aim of the present study was to evaluate the applicability of Quillaja saponaria extract (QSE) and Nα-lauroyl-l-arginine ethyl ester (LAE) as antimicrobial wash water additives in fresh-cut lettuce processing. Antibacterial activities of LAE and QSE against selected strains of the foodborne pathogens Salmonella enterica, Bacillus cereus, Staphylococcus aureus, Pseudomonas aeruginosa, and Listeria monocytogenes were examined in vitro. Minimum inhibitory concentrations determined by broth microdilution assay demonstrated that LAE exhibited a strong antimicrobial activity with MICs between 4 and 32 μg/mL against all tested strains, whereas QSE showed a weaker antimicrobial activity with MICs >512 μg/mL. On a pilot-plant scale, the effects of warm water washing at 45 °C for 120 s with and without 40 mg/L QSE or 100 mg/L LAE as well as cold water washing at 4 °C for 120 s with QSE or LAE, respectively, of shredded endive (Cichorium endivia L.) were investigated regarding microbiological and sensory quality as well as physiological properties. Samples were analyzed for headspace O2 and CO2 levels, phenylalanine-ammonia-lyase activity and contents of nitrite and nitrate during nine days of cold storage at 4 °C. By analogy to its antimicrobial effect against the foodborne pathogens in vitro, LAE allowed up to 4 log10 cfu/mL reduction of the microbial load in the washing water of the pilot plant, and might therefore reduce cross-contamination while saving water. The addition of LAE to warm washing water impaired sensory properties of fresh-cut endive during storage, which was predicted by chlorophyll fluorescence imaging analyses. QSE treatment combined with warm water washing best retained sensory appearance throughout our study, being possibly suitable for the production of premium products. 相似文献

This study evaluates the aflatoxin M1 (AFM1) contamination in 804 samples of raw milk from cow and buffalo, collected randomly in Campania and Calabria regions of Southern Italy over a two years period.The competitive enzyme linked immunosorbent assay (ELISA) method was used to analyze AFM1 in the samples. AFM1 levels result above the CCβ value of 0.004 μg/kg in 51 (12.3%) cow milk samples and in 28 (7.2%) buffalo milk samples. Positive results from screening analysis were confirmed by high performance liquid chromatography with fluorimetric detection (HPLC-FLD) after a procedure of centrifugation, extraction and immunoaffinity column clean-up of milk. Only one cow milk sample exceeded the maximum limit (0.05 μg/kg) set by the European Regulation.The occurrence of AFM1 contamination was significantly (p < 0.05) higher in cold season, particularly fall, than in warm season, principally spring.Our results indicate that feedstuff used in the buffalo and cow farms were not highly contaminated with aflatoxins, determining a good quality of the analyzed milk.
