In men with gonococcal urethritis, the urethral epithelial cell is a site of infection. To study the pathogenesis of gonorrhea in this cell type, we have developed a method to culture primary human urethral epithelial cells obtained at the time of urologic surgery. Fluorescent analysis demonstrated that 100% of the cells stained for keratin. Microscopic analyses indicated that these epithelial cells arrayed in a pattern similar to that seen in urethral epithelium. Using immunoelectron and confocal microscopy, we compared the infection process seen in primary cells with events occurring during natural infection of the same cell type in men with gonococcal urethritis. Immunoelectron microscopy studies of cells infected with Neisseria gonorrhoeae 1291 Opa+ P+ showed adherence of organisms to the epithelial cell membrane, pedestal formation with evidence of intimate association between the gonococcal and the epithelial cell membranes, and intracellular gonococci present in vacuoles. Confocal studies of primary urethral epithelial cells showed actin polymerization upon infection. Polyclonal antibodies to the asialoglycoprotein receptor (ASGP-R) demonstrated the presence of this receptor on infected cells in the primary urethral cell culture. In situ hybridization using a fluorescent-labeled probe specific to the ASGP-R mRNA demonstrated this message in uninfected and infected cells. These features were identical to those seen in urethral epithelial cells in exudates from males with gonorrhea. Infection of primary urethral cells in culture mimics events seen in natural infection and will allow detailed molecular analysis of gonococcal pathogenesis in a human epithelial cell which is commonly infected.  相似文献

Several studies have suggested that primates react differently to spatial reduction. In this article, the authors tested some general hypotheses on primate response to spatial reduction by studying the Apenheul lowland gorillas (Gorilla gorilla gorilla; Apeldoorn, the Netherlands). The frequency of conflicts did not greatly change between the 2 housing settings, thus not supporting the density- aggression model. Indoor, gorillas performed touching behavior more often and increased their level of reconciliation. These findings support the coping model. Indoor, the gorillas also maintained broader interindividual distances by increasing the levels of sitting alone, avoidance, and dismissing behaviors. In conclusion, the Apenheul gorillas modified selectively the distribution of some patterns typical of their behavioral repertoire in response to a high-density condition. Diabetes mellitus is a group of heterogeneous metabolic disorders characterized by chronic hyperglycaemia mainly due to pancreatic β cell death and/or dysfunction, caused by several types of stress such as glucotoxicity, lipotoxicity and inflammation. Different patho-physiological mechanisms driving β cell response to these stresses are tightly regulated by microRNAs (miRNAs), a class of negative regulators of gene expression, involved in pathogenic mechanisms occurring in diabetes and in its complications. In this review, we aim to shed light on the most important miRNAs regulating the maintenance and the robustness of β cell identity, as well as on those miRNAs involved in the pathogenesis of the two main forms of diabetes mellitus, i.e., type 1 and type 2 diabetes. Additionally, we acknowledge that the understanding of miRNAs-regulated molecular mechanisms is fundamental in order to develop specific and effective strategies based on miRNAs as therapeutic targets, employing innovative molecules.  相似文献

The kinetics of diffusion of water from different activity vapors and liquid phase have been investigated in glassy amorphous poly(aryl ether ether ketcne) (PEEK) films at the temperature of 60°C and in glassy semicrystalline PEEK sheets at different temperatures, respectively. In the case of the amorphous PEEK films (250 μm thick) the data at low activity levels were interpreted by means of a purely Fickian mechanism. At higher activity levels the material has shown the presence of a relaxation process; in this case the data have been interpreted using a model proposed by Berens and Hopfenberg. Equilibrium sorption isotherm is also reported. Liquid water sorption in semicrystalline (30%) PEEK sheets (2 mm thick) has been determined to follow the classical Fickian mechanism. The water uptake values obtained for both amorphous and semicrystalline PEEK, confirm the good moisture and liquid water resistance of this kind of high performance thermoplastic polymer.  相似文献

One of the main aims in agriculture is to guarantee soil wellness, which is a fundamental requirement to produce high-quality crops with high yields. Focused on this aim, periodical administrations of nutrients or phytostrengtheners are often necessary. The most relevant disadvantages of these administrations are the high dosage number required and the low availability of the substance within the soil. For these reasons, a crucial goal to increase the economic and environmental sustainability of the cultivation process is to reduce the dosage number, which can be obtained increasing the active substance availability in the soil. A granular hydroxypropyl methylcellulose (HPMC) matrix, produced using the wet granulation process, was used to encapsulate a phytostrengthener and to guarantee its controlled release. The granular product was characterized in terms of granules properties and phytostrengtheners leaching within the soil. The results showed good flowability and mechanical properties of the granules as well as the possibility to reduce the product leaching with the phytostrengtheners encapsulation in the HPMC matrices.  相似文献

The viscoelastic properties of the amorphous and semicrystalline PEEK and of a PEEK-based composite (APC2) have been analyzed as a function of the aging time and related free volume relaxation, at temperatures close to the glass transition temperature. The effect of thermal treatments (physical aging) on the polymer amorphous phase of the three systems is presented. Creep tests approaching the “zero stress” were performed on samples subjected to different thermal histories. The presence of a second phase, namely crystallites and fibers, determines a loss of the mobility of the polymeric amorphous chains and consequently a decrease of the rate of free volume relaxation.  相似文献

Pancreatic islet transplantation consists of replacing insulin-producing cells to restore normal glycemia in diabetic patients. This is a minimal invasive procedure that has been proved successful. Unfortunately unpredictability of islet transplant outcome remains a frustrating and costly issue limiting the clinical implementation of this procedure. Multiple variables are involved in the procedure and assessment is subjective to individual operators. The aim of this study was to generate a system expressing the probability of transplant success in relation to four classes of identified variables (donor, organ, isolation and recipient). We have proposed the utilization of Multi-Criteria Decision Making methods (MCDM) as a powerful tool for evaluating pancreatic islet transplant-related information with the goal to achieve optimal decision. Technique Ordered Preference by Similarity to the Ideal Solution (TOPSIS), one of the most widely used MCDM methods in decision support systems, was here utilized with modification to fit better in a medical system. In our modified method, we have utilized fuzzy logic in order to consider uncertain and vague data.  相似文献

Carbon fiber posts (CFP) are widely used in the restoration of endodontically treated teeth to enhance the mechanical behavior in spite of metallic posts and to prevent vertical fractures of the tooth under chewing loads. The post is cemented inside the canal lumen using polymer resins with Young's modulus lower than Dentine. In this conditions the stress concentration is located at the post-cement interface and in the cement bulk itself, preserving radicular Dentine from dangerous stress accumulation. The mechanical resistance of CFP posts cemented in human Dentine was evaluated by the means of mechanical pull-out tests assisted by the finite element analysis. The average bond strength and the critical stress values of the CHP-cement interface were 25 MPa and 50 MPa respectively. ©2000 Kluwer Academic Publishers  相似文献
