We have screened the p53 status of 156 human cell lines, including 142 tumor cell lines from 27 different tumor types and 14 cell lines from normal tissues by using functional analysis of separated alleles in yeast. This assay enables us to score wild-type p53 expression on the basis of the ability of expressed p53 to transactivate the reporter gene HIS3 via the p53-responsive GAL1 promotor in Saccharomyces cerevisiae. Of 142 tumor cell lines, at least 104 lines (73.2%) were found to express the mutated p53 gene: 94 lines (66.2%) were mutated in both alleles, three lines (2.1%) were heterozygous, and no p53 cDNA was amplified from seven lines (4.9%). Of the 14 cell lines originating from normal tissues, all the transformed or immortalized cell lines expressed mutant p53 only. Yeast cells expressing mutant p53 derived from 94 cell lines were analyzed for temperature-sensitive growth. p53 cDNA from eight cell lines showed p53-dependent temperature-sensitive growth, growing at 30 degrees C but not at 37 degrees C. Four temperature-sensitive p53 mutations were isolated: CAT-->CGT at codon 214 (H214R), TAC-->TGC at codon 234 (Y234C), GTG-->ATG at codon 272 (V272M), and GAG-->AAG (E285K). Functionally wild-type p53 was detected in 38 tumor cell lines (26.8%) and all of the diploid fibroblasts at early and late population doubling levels. These results strongly support the previous findings that p53 inactivation is one of the most frequent genetic events that occurs during carcinogenesis and immortalization. 相似文献

We showed that human adult red blood cells (RBCs) produce prostaglandin E1 (PGE1) and E2 (PGE2). RBCs that were mechanically stressed in the presence of extracellular Ca2+ by being injected rapidly through a fine needle produced PGE1 and PGE2 within 30 min after this mechanical stress. The amounts of PGE1 and PGE2 produced by 1 x 10(9) mechanically stressed RBCs were approximately 50 pg and 100 pg, respectively, which were determined in the cytosolic fraction from sonicated RBCs using a competitive enzyme immunoassay method. A Western blot analysis using anti-cyclooxygenase-2 antibody revealed a band at the 70-kDa position in the samples from RBCs producing PGE1 and PGE2. Treatment with 10 micrograms/mL indomethacin completely inhibited the productions of PGE1 and PGE2. The present results may indicate a new role of RBCs in microcirculation. 相似文献

ABSTRACTFukushima Daiichi nuclear power plants (1F) were damaged by unprecedented severe accident in the great east Japan earthquake, and seawater and freshwater had been injected as an emergency countermeasure for the core cooling. Although, the primary containment vessel (PCV) was not supposed to be exposed to diluted seawater, the PCV will be exposed to diluted seawater environment until fuel debris removal is completed. Therefore, it is necessary to establish a countermeasure of corrosion for the PCVs made of carbon steel. In this study, the effect of the addition of corrosion inhibitor, which is one of the corrosion countermeasures, was investigated by two types of corrosion tests. As a result of the immersion corrosion test, it was found that any of the three kinds of corrosion inhibitor could suppress corrosion of carbon steel. In addition, as a result of the inhibitor interim addition test, it was found that corrosion of carbon steel covered with corrosion products could be suppressed by optimizing the additive amount of corrosion inhibitor in the cooling water. 相似文献

By time-resolved reflectivity measurements with sub-10 fs laser pulses at 395 nm, the coherent phonons of aligned bundles of single-walled carbon nanotubes are observed for various polarization directions of the pump and probe pulses. In the isotropic reflectivity measurement, we observe the radial breathing modes, G, and even D modes, while in the anisotropic reflectivity mode, only the G mode appears. A complex polarization dependence of the G band phonon amplitude in the isotropic reflectivity is explained by the superposition of G band phonons with different symmetries. 相似文献

Compensation for repeatable tracking errors in hard drives usingdiscrete-time repetitive controllers
Presents the repeatable runout (RRO) compensation in hard disk drives (HDDs). To cancel the RRO, we applied repetitive controllers to the tracking system. Through experiments, we obtained a high reduction ratio of from 10 to 45 dB for up to 10th harmonics component of the RRO. Concentrations of population and business activities result in high energy demand in urban areas. This requires the construction of underground substations. Oil‐free, nonflammable, nonexplosive equipment is recommended for underground substations. For this reason, gas‐insulated transformers have been developed. A diagnostic method for gas‐insulated transformers is thus required. This paper provides an experimental survey of the main components of decomposition gas generated by various faults in gas‐insulated transformers carried out through simplified model tests. The phenomena of overheating and partial discharges are modeled, taking the actual materials related to each fault into account. For example, CO, CO2, and aldehydes are produced by overheating of pressboards and PET films. The amount of gas produced increases with rising temperature. While various gases are produced from a partial discharge, the principal components are SO2 and SOF2.
