In this report the effects of single doses of ionizing radiation on the mRNA expression of several proteins involved in multiple drug resistance were analyzed. Murine NIH 3T3 cells treated with single doses of 5, 10 and 20 Gy during the time interval from 1.5 to 72 h after irradiation were compared with their corresponding controls at the same points of time. The glutathione S-transferase-pi (GST pi) level was elevated in cells treated with 10 or 20 Gy from 24 to 72 h after irradiation compared with the control. Topoisomerase II alpha and thymidylate synthase were decreased in irradiated cells 24-72 h after exposure. These down-regulations were associated with cellular proliferation, determined by mRNA expression of the proliferation marker histone 3. Irradiated cells exhibited no alteration in the P-glycoprotein or glutathione peroxidase mRNA content. The finding that GST pi mRNA was overexpressed after irradiation was validated by investigations on a human lung carcinoma cell line (LXF 289) on the mRNA and protein level. Thus, our results indicate that irradiation alters the expression of proteins involved in multidrug resistance and may, therefore, play a role in clinical drug response.  相似文献

Physical forces play a fundamental role in the regulation of cell function in many tissues, but little is known about how cells are able to sense mechanical loads and realize signal transduction. Adhesion receptors like integrins are candidates for mechanotransducers. We used a magnetic drag force device to apply forces on integrin receptors in an osteoblastic cell line and studied the effect on tyrosine phosphorylation as a biochemical event in signal transduction. Mechanical stressing of both the beta1 and the alpha2 integrin subunit induced an enhanced tyrosine phosphorylation of proteins compared with integrin clustering. Application of cyclic forces with a frequency of 1 Hz was more effective than a continuous stress. Using Triton X-100 for cell extraction, we found that tyrosine-phosphorylated proteins became physically anchored to the cytoskeleton due to mechanical integrin loading. This cytoskeletal linkage was dependent on intracellular calcium. To see if mechanical integrin stressing induced further downstream signaling, we analyzed the activation of mitogen-activated protein (MAP) kinases and found an increased phosphorylation of MAP kinases due to mechanical stress. We conclude that integrins sense physical forces that control gene expression by activation of the MAP kinase pathway. The cytoskeleton may play a key role in the physical anchorage of activated signaling molecules, which enables the switch of physical forces to biochemical signaling events.  相似文献

The complex nature of cable joints leads to errors when PD analysis is carried out using conventional equivalent circuits. A method for analyzing electrostatic discharge (ESD) generators and coupling to equipment under test in the frequency domain is proposed. In ESD generators, the pulses are excited by the voltage collapse across relay contacts. The voltage collapse is replaced by one port of a vector network analyer (VNA). All the discrete and structural elements that form the ESD current pulse and the transient fields are excited by the VNA as if they were excited by the voltage collapse. In such a way, the method allows analyzing the current and field-driven linear coupling without having to discharge an ESD generator, eliminating the risk to the circuit and allowing the use of the wider dynamic range of a network analyzer relative to a real-time oscilloscope. The method is applicable to other voltage-collapse-driven tests, such as electrical fast transient, ultrawideband susceptibility testing but requires a linear coupling path.  相似文献

The discharge current and the transient fields of an electrostatic discharge (ESD) generator in the contact mode are numerically simulated using the finite-difference time-domain method. At first the static field is established. Then the conductivity of the relay contact is changed, which initiates the discharge process. The simulated data are used to study the effect of design choices on the current and fields. They are compared to measured field and current data using a multidecade broadband field and current sensors. The model allows accurate prediction of the fields and currents of ESD generators, thus it can be used to evaluate different design choices.  相似文献

Investigation of a dc power delivery network, consisting of a multilayer PCB using area fills for power and return, involves the distributed behavior of the power/ground planes and the parasitics associated with the lumped components mounted on it. Full-wave methods are often employed to study the power integrity problem. While full-wave methods can be accurate, they are time and memory consuming. The cavity model of a rectangular structure has previously been employed to efficiently analyze the simultaneous switching noise (SSN) in the power distribution network. However, a large number of modes in the cavity model are needed to accurately simulate the impedance associated with the vias, leading to computational inefficiency. A fast approach is detailed herein to accelerate calculation of the summation associated with the higher-order modes. Closed-form expressions for the parasitics associated with the interconnects of the decoupling capacitors are also introduced. Combining the fast calculation of the cavity models of regularly shaped planar circuits, a segmentation method, and closed-form expressions for the parasitics, an efficient approach is proposed herein to analyze an arbitrary shaped power distribution network. While it may take many hours for a full-wave method to do a single simulation, the proposed method can generally perform the simulation with good accuracy in several minutes. Another advantage of the proposed method is that a SPICE equivalent circuit of the power distribution network can be derived. This allows both frequency and transient responses to be done with SPICE simulation.  相似文献

The high frequency properties of coaxial power cables are modeled using time- and frequency-domain numerical simulations. This is required due to the complex helical structure of the outer metallic screen. The finite element (FEM) and finite difference time domain methods (FDTD) have been employed to study the effect of screen spiralization. It is established that this screen design causes a dependence of the cable high frequency characteristics on the surrounding medium. The development and deployment of radio frequency identification (RFID) systems render a novel distributed sensor network which enhances visibility into manufacturing processes. In RFID systems, the detection range and read rates will suffer from interference among high-power reading devices. This problem grows severely and degrades system performance in dense RFID networks. Consequently, medium access protocols (MAC) protocols are needed for such networks to assess and provide access to the channel so that tags can be read accurately. In this paper, we investigate a suite of feasible power control schemes to ensure overall coverage area of the system while maintaining a desired read rate. The power control scheme and MAC protocol dynamically adjust the RFID reader power output in response to the interference level seen during tag reading and acceptable signal-to-noise ratio (SNR). We present novel distributed adaptive power control (DAPC) as a possible solution. A suitable back off scheme is also added with DAPC to improve coverage. A generic UHF wireless testbed is built using UMR/SLU GEN4-SSN for implementing the protocol. Both the methodology and hardware implementation of the schemes are presented, compared, and discussed. The results of hardware implementation illustrate that the protocol performs satisfactorily as expected.  相似文献

Electrostatic discharge (ESD) generators are used for testing the robustness of electronics toward ESD. Most generators are built in accordance with the IEC 61000-4-2 specifications. Using only a few parameters, this standard specifies the peak current, the rise time and the falling edge. Lacking a transient field specification, test results vary depending on which generator is used, even if the currents are quite similar. Such a specification is needed to improve the test repeatability. As for the current, the specification should be based on a reference human metal ESD event. While keeping the presently set peak current and rise time values, such a reference ESD (5 kV, 850-/spl mu/m arc length) is identified and specifications for current derivative, fields, and induced voltages are derived. The reference event parameters are compared to typical ESD generators.  相似文献
