The optical and electronic design features of a reliable microspectrofluorometer that allows successful microfluorometric studies in areas of brain ischemia over a period of 2.0 hours are described in detail. The apparatus allows moment-to-moment recording of the oxidation-reduction state of brain mitochondria in areas of ischemia and therefore affords the potential of measuring the immediate effect of a variety of drugs designed either to increase blood flow in areas of ischemia or to support marginal metabolism. Problems of choice of anesthetic agent, hemoglobin artifact, photodecomposition, and preparation reliability are discussed.  相似文献

Objective

This study evaluates the inter-site and intra-site reproducibility of 7 Tesla brain imaging and compares it to literature values for other field strengths.

Materials and methods

The same two subjects were imaged at eight different 7 T sites. MP2RAGE, TSE, TOF, SWI, EPI as well as B1 and B0 field maps were analyzed quantitatively to assess inter-site reproducibility. Intra-site reproducibility was measured with rescans at three sites.

Results

Quantitative measures of MP2RAGE scans showed high agreement. Inter-site and intra-site reproducibility errors were comparable to 1.5 and 3 T. Other sequences also showed high reproducibility between the sites, but differences were also revealed. The different RF coils used were the main source for systematic differences between the sites.

Conclusion

Our results show for the first time that multi-center brain imaging studies of the supratentorial brain can be performed at 7 T with high reproducibility and similar reliability as at 3T. This study develops the basis for future large-scale 7 T multi-site studies.

The corrosion resistance of aluminium surfaces is closely linked to the surface state after a grinding process. For years, iron-containing abrasive materials were suspected to lead to increased corrosion susceptibility after processing of aluminium surfaces. To prove a possible correlation between the iron content of an abrasive and the corrosion behaviour of aluminium components, scientific investigations and experimentally practical corrosion tests are necessary. For the current investigation, specimens of a technical Al-Si alloy from the same batch were used. The test specimens were mechanically ground with various resin-bonded model abrasives containing different iron contents. The performed corrosion tests did not reveal a negative influence of the different iron-containing abrasives on the corrosion behaviour of the Al–Si alloy. However, the most sensitive measuring method (electrochemical noise) showed differences in the surface activity depending on the type of abrasive.  相似文献

Multiple sites within Germany operate human MRI systems with magnetic fields either at 7 Tesla or 9.4 Tesla. In 2013, these sites formed a network to facilitate and harmonize the research being conducted at the different sites and make this technology available to a larger community of researchers and clinicians not only within Germany, but also worldwide. The German Ultrahigh Field Imaging (GUFI) network has defined a strategic goal to establish a 14 Tesla whole-body human MRI system as a national research resource in Germany as the next progression in magnetic field strength. This paper summarizes the history of this initiative, the current status, the motivation for pursuing MR imaging and spectroscopy at such a high magnetic field strength, and the technical and funding challenges involved. It focuses on the scientific and science policy process from the perspective in Germany, and is not intended to be a comprehensive systematic review of the benefits and technical challenges of higher field strengths.

Objective

To quantify the periventricular venous density in neuromyelitis optica spectrum disease (NMOSD) in comparison to that in patients with multiple sclerosis (MS) and healthy control subjects.

Materials and methods

Sixteen patients with NMOSD, 16 patients with MS and 16 healthy control subjects underwent 7.0-Tesla (7T) MRI. The imaging protocol included T2*-weighted (T2*w) fast low angle-shot (FLASH) and fluid-attenuated inversion recovery (FLAIR) sequences. The periventricular venous area (PVA) was manually determined by a blinded investigator in order to estimate the periventricular venous density in a region of interest-based approach.

Results

No significant differences in periventricular venous density indicated by PVA were detectable in NMOSD versus healthy controls (p = 0.226). In contrast, PVA was significantly reduced in MS patients compared to healthy controls (p = 0.013).

Conclusion

Unlike patients with MS, those suffering from NMOSD did not show reduced venous visibility. This finding may underscore primary and secondary pathophysiological differences between these two distinct diseases of the central nervous system.

The retina is a complex neurological tissue and is extremely sensitive to an insufficient supply of oxygen. Hypoxia plays a major role in several retinal diseases, and often results in the loss of cells that are essential for vision. Cyclosporine A (CsA) is a widely used immunosuppressive drug. Furthermore, treatment with CsA has neuroprotective effects in several neurologic disorders. No data are currently available on the tolerated concentration of CsA when applied to the retina. To reveal the most effective dose, retinal explants from rat eyes were exposed to different CsA concentrations (1–9 µg/mL). Immunohistochemistry with brain-specific homeobox/POU domain protein 3a (Brn3a) and TUNEL staining was performed to determine the percentage of total and apoptotic retinal ganglion cells (RGCs), as well as the responses of micro- and macroglial cells. Furthermore, optical coherence tomography (OCT) scans were performed to measure the changes in retinal thickness, and recordings with multielectrode array (MEA) were performed to evaluate spontaneous RGC spiking. To examine the neuroprotective effects, retinas were subjected to a hypoxic insult by placing them in a nitrogen-streamed hypoxic chamber prior to CsA treatment. In the biocompatibility tests, the different CsA concentrations had no negative effect on RGCs and microglia. Neuroprotective effects after a hypoxic insult on RGCs was demonstrated at a concentration of 9 µg/mL CsA. CsA counteracted the hypoxia-induced loss of RGCs, reduced the percentage of TUNEL+ RGCs, and prevented a decrease in retinal thickness. Taken together, the results of this study suggest that CsA can effectively protect RGCs from hypoxia, and the administered concentrations were well tolerated. Further in vivo studies are needed to determine whether local CsA treatment may be a suitable option for hypoxic retinal diseases.  相似文献

A series of 50 experiments was performed to investigate particle size and yield from three laboratory spray dryers with two-fluid nozzles, using an aqueous solution of egg albumin as the feed. First principles theory suggests that the main parameters affecting final particle size should be atomization gas flowrate and velocity, feed solution flowrate, solution concentration, and inlet and outlet gas temperature. The experiments demonstrated that atomization gas velocity at the nozzle correlated better than volumetric gas flowrate to the final particle size obtained. Increasing atomization gas flow gave a decrease in particle size, levelling off at high nozzle gas velocities approaching sonic values. Liquid flowrate had little effect, and hybrid ratios such as the ALM (atomization gas to liquid flow ratio) did not correlate well to measured final particle size for this system. This seems likely to apply to aqueous systems with temperature limitations, where liquid flowrates will be relatively low compared to gas flowrates (both atomization and main gas flow). Particle size generally increased with solution concentration, and in some cases scaled exactly, assuming a constant initial droplet size and final particle density. All experiments were performed below the boiling point and therefore temperature had little effect. Particles of 2–3 microns were obtained at high atomization gas flows, indicating initial droplet sizes of 4–7 microns at most, while lower flowrates gave substantially larger particles and a wider size distribution.  相似文献

With increasing flexibility of technology and a shift towards competence being the core of competitive edge in worklife, the need for new organizational concepts or models which givejoint optimization across human and technological dimensions has been acknowledged in leading, innovative enterprises. National crossdisciplinary research based productivity programmes are appearing in several countries. Due to internationalization and the general shortcomings of bureaucratic organizational forms, regional networks of enterprises in cooperation with public R&D institutions seem to provide answers to needs of regions to remain competitive. The paper discusses the possible role of social science in a multilevel, participative strategy for developing new, democratic and productive organizational forms in worklife. Aspects of national productivity programmes in Scandinavia, Germany and Australia are discussed in connection with the situation in Italy as analyzed through some recent research projects.  相似文献
