A detailed long range restriction map of the region defined by markers D6S149 and D6S193 on chromosome 6q27 has been constructed. This was achieved by YAC cloning and contig assembling of the same region. Seven YAC clones were found to span the almost 1000 Kb region flanked by the two markers which on the genetic map resulted to be 1.9 cM apart. With some of the characterized YAC clones we undertook a molecular cytogenetic analysis of 20 benign ovarian tumors. The rationale for this was the recent mapping to a region of chromosome 6q27, flanked by markers D6281 and D6S133, of a locus for the SV40-mediated immortalization of human cells (SEN6 gene). Noteworthy we found that the the D6S149-D6S193 region (comprised in the larger D6S281-D6S133 physical interval) was altered in all samples analysed adding support to the occurrence of a immortalization step in this type of tumors. 相似文献

TP53 gene mutations occur in 70% of oesophageal adenocarcinomas (OACs). Given the central role of p53 in controlling cellular response to therapy we investigated the role of mutant (mut-) p53 and SLC7A11 in a CRISPR-mediated JH-EsoAd1 TP53 knockout model. Response to 2 Gy irradiation, cisplatin, 5-FU, 4-hydroxytamoxifen, and endoxifen was assessed, followed by a TaqMan OpenArray qPCR screening for differences in miRNA expression. Knockout of mut-p53 resulted in increased chemo- and radioresistance (2 Gy survival fraction: 38% vs. 56%, p < 0.0001) and in altered miRNA expression levels. Target mRNA pathways analyses indicated several potential mechanisms of treatment resistance. SLC7A11 knockdown restored radiosensitivity (2 Gy SF: 46% vs. 73%; p = 0.0239), possibly via enhanced sensitivity to oxidative stress. Pathway analysis of the mRNA targets of differentially expressed miRNAs indicated potential involvement in several pathways associated with apoptosis, ribosomes, and p53 signaling pathways. The data suggest that mut-p53 in JH-EsoAd1, despite being classified as non-functional, has some function related to radio- and chemoresistance. The results also highlight the important role of SLC7A11 in cancer metabolism and redox balance and the influence of p53 on these processes. Inhibition of the SLC7A11-glutathione axis may represent a promising approach to overcome resistance associated with mut-p53. 相似文献

Bulk and quantum well GaAs1?xBix/GaAs layers with Bi mole fractions from 0.02 to 0.10 are grown by molecular-beam epitaxy at temperatures ranging from 280 to 320?°C. The samples are characterized using temperature and pump-power dependent photoluminescence measurements covering 8–300?K and 1–250?mW (7–1,800?W/cm2), respectively. The results indicate that there is strong reduction in bandgap energy with the incorporation of small amounts of Bi and that GaAsBi most likely forms a weak type-I band alignment with GaAs. 相似文献

Repetitive transcranial magnetic stimulation (rTMS) is a non-invasive brain stimulation technique that has the potential to treat a variety of neurologic and psychiatric disorders. The extent of rTMS-induced neuroplasticity may be dependent on a subject’s brain state at the time of stimulation. Chronic low intensity rTMS (LI-rTMS) has previously been shown to induce beneficial structural and functional reorganisation within the abnormal visual circuits of ephrin-A2A5-/- mice in ambient lighting. Here, we administered chronic LI-rTMS in adult ephrin-A2A5-/- mice either in a dark environment or concurrently with voluntary locomotion. One day after the last stimulation session, optokinetic responses were assessed and fluorescent tracers were injected to map corticotectal and geniculocortical projections. We found that LI-rTMS in either treatment condition refined the geniculocortical map. Corticotectal projections were improved in locomotion+LI-rTMS subjects, but not in dark + LI-rTMS and sham groups. Visuomotor behaviour was not improved in any condition. Our results suggest that the beneficial reorganisation of abnormal visual circuits by rTMS can be significantly influenced by simultaneous, ambient visual input and is enhanced by concomitant physical exercise. Furthermore, the observed pathway-specific effects suggest that regional molecular changes and/or the relative proximity of terminals to the induced electric fields influence the outcomes of LI-rTMS on abnormal circuitry. 相似文献

A surface of epoxy-impregnated hardened cement paste was investigated using a novel atomic force microscopy (AFM) imaging mode that allows for the quantitative mapping of the local elastic modulus. The analyzed surface was previously prepared using focussed ion beam milling. The same surface was also characterized by electron microscopy and energy-dispersive X-ray spectroscopy.We demonstrate the capability of this quantitative nanomechanical mapping to provide information on the local distribution of the elastic modulus (from about 1 to about 100 GPa) with a spatial resolution in the range of decananometers, that corresponds to that of low-keV back-scattered electron imaging. Despite some surface roughness which affects the measured nanomechanical properties it is shown that topography, adhesion and Young's modulus can be clearly distinguished.The quantitative mapping of the local elastic modulus is able to discriminate between phases in the cement paste microstructure that cannot be distinguished from the corresponding back-scattered electron images. 相似文献

Non-isothermal TG/DTG measurements are widely used to determine kinetic parameters of oxidation and gasification of carbons, e.g. by multi-heating rate methods. Thereby it is important to avoid spurious effects due to diffusion limitations. In this work, TG/DTG experiments with activated carbon were conducted under isothermal and non-isothermal conditions. The agreement of the experimental results with results of simulations based on the intrinsic kinetics, pore diffusion and external diffusion in/to the sample (crucible) is very good. The simulations show that mass transfer has an unwanted influence on TG measurements, if the concentration of the gaseous reactant (O2 and CO2) is too low and/or the heating rate is too high. Based on literature data, the simulation was extended to blast furnace coke with a very low reactivity and to highly reactive coke from pyrolysis of miscanthussinensis. The simulations and theoretical considerations lead to a criterion to exclude the influence of mass transfer on TG measurements a priori. This criterion is valid for combustion as well as for gasification with steam or carbon dioxide. 相似文献

This article examines the usefulness of a thick thermally sprayed interlayer (plasma-sprayed Ni-50%Cr, plasma-sprayed Al2O3-13%TiO2, or high-velocity oxygen-fuel-sprayed WC-17%Co) for enhancing the wear resistance and the corrosion protectiveness of a diamond-like
carbon (DLC)-based thin film deposited onto a carbon steel substrate. Scratch tests indicate that the Al2O3-13%TiO2 and WC-17%Co interlayers definitely increase the critical spallation load of the thin film, but the Al2O3-13%TiO2 interlayer itself undergoes brittle fracture under high-contact loads. Accordingly, during ball-on-disk tests at room temperature,
no cracking and spallation occur in the DLC-based film deposited onto the WC-17%Co interlayer, whereas the one onto the Al2O3-13%TiO2 interlayer is rapidly removed because the interlayer itself is fractured. At 300 °C, by contrast, the DLC-based film on the
Al2O3-13%TiO2 interlayer offers the best tribological performance, possibly thanks to the increased toughness of the ceramic interlayer
at this temperature. Electrochemical polarization tests indicate that the thin film/WC-Co systems possess the lowest corrosion
