Conducting, mechanically durable, elastic nanocomposite films were prepared with chitosan (CS) as the polymer matrix, graphene obtained from highly exfoliated graphite as the nanofiller, and poly(vinyl pyrrolidone) (PVP) as the stabilizer of the graphene sheets. The maximum graphene content in the composites without a loss of uniformity and other useful properties increased up to 4.0 wt %. The resulting composites were characterized by scanning electron microscopy, Raman spectroscopy, X‐ray diffraction analysis, mechanical testing, and electrical conductivity testing to determine the effects of the addition of graphene on the morphology and mechanical and electrical properties of the CS–PVP–graphene nanocomposite films. In this study, we took an approach to making nanocomposites from the perspectives of green chemistry, environmental protection, regenerative medicine, and low cost. © 2017 Wiley Periodicals, Inc. J. Synthetic targeted optimization of plant promoters is becoming a part of progress in mainstream postgenomic agriculture along with hybridization of cultivated plants with wild congeners, as well as marker-assisted breeding. Therefore, here, for the first time, we compiled all the experimental data—on mutational effects in plant proximal promoters on gene expression—that we could find in PubMed. Some of these datasets cast doubt on both the existence and the uniqueness of the sought solution, which could unequivocally estimate effects of proximal promoter mutation on gene expression when plants are grown under various environmental conditions during their development. This means that the inverse problem under study is ill-posed. Furthermore, we found experimental data on in vitro interchangeability of plant and human TATA-binding proteins allowing the application of Tikhonov’s regularization, making this problem well-posed. Within these frameworks, we created our Web service Plant_SNP_TATA_Z-tester and then determined the limits of its applicability using those data that cast doubt on both the existence and the uniqueness of the sought solution. We confirmed that the effects (of proximal promoter mutations on gene expression) predicted by Plant_SNP_TATA_Z-tester correlate statistically significantly with all the experimental data under study. Lastly, we exemplified an application of Plant_SNP_TATA_Z-tester to agriculturally valuable mutations in plant promoters. 相似文献

The effects of insulin on the bioenergetic and thermogenic capacity of brown adipocyte mitochondria were investigated by focusing on key mitochondrial proteins. Two-month-old male Wistar rats were treated acutely or chronically with a low or high dose of insulin. Acute low insulin dose increased expression of all electron transport chain complexes and complex IV activity, whereas high dose increased complex II expression. Chronic low insulin dose decreased complex I and cyt c expression while increasing complex II and IV expression and complex IV activity. Chronic high insulin dose decreased complex II, III, cyt c, and increased complex IV expression. Uncoupling protein (UCP) 1 expression was decreased after acute high insulin but increased following chronic insulin treatment. ATP synthase expression was increased after acute and decreased after chronic insulin treatment. Only a high dose of insulin increased ATP synthase activity in acute and decreased it in chronic treatment. ATPase inhibitory factor protein expression was increased in all treated groups. Confocal microscopy showed that key mitochondrial proteins colocalize differently in different mitochondria within a single brown adipocyte, indicating mitochondrial mosaicism. These results suggest that insulin modulates the bioenergetic and thermogenic capacity of rat brown adipocytes in vivo by modulating mitochondrial mosaicism. 相似文献

A new approach to making a photopolymeric composition was first elaborated in this work based on the use of poly‐N,N′‐(m‐phenylene)isophtalamide as the heterochain polymer matrix together with 4,4′‐diphenyl oxide diacrylamide as the crosslinking agent. Reproducible three‐dimensional (3D) objects have been formed using a laser stereolithograph completed with a laser with wavelength of 405 nm. According to the thermogravimetric analysis, the thermal stability of the formed objects could be changed through a control from 260 to 405 °C. Moreover, it was proven that the tensile strength of samples of the crosslinked films reached 90.1 ± 3.2 MPa at the elongation at break of 12.4 ± 2.3%, depending on the forming parameters of such samples. © 2018 Wiley Periodicals, Inc.
