Psoriasis is a chronic, immune-mediated inflammatory disease that affects around 125 million people worldwide. Several studies concerning the gut microbiota composition and its role in disease pathogenesis recently demonstrated significant alterations among psoriatic patients. Certain parameters such as Firmicutes/Bacteroidetes ratio or Psoriasis Microbiome Index were developed in order to distinguish between psoriatic and healthy individuals. The “leaky gut syndrome” and bacterial translocation is considered by some authors as a triggering factor for the onset of the disease, as it promotes chronic systemic inflammation. The alterations were also found to resemble those in inflammatory bowel diseases, obesity and certain cardiovascular diseases. Microbiota dysbiosis, depletion in SCFAs production, increased amount of produced TMAO, dysregulation of the pathways affecting the balance between lymphocytes populations seem to be the most significant findings concerning gut physiology in psoriatic patients. The gut microbiota may serve as a potential response-to-treatment biomarker in certain cases of biological treatment. Oral probiotics administration as well as fecal microbial transplantation were most reported in bringing health benefits to psoriatic patients. However, the issue of psoriatic bacterial gut composition, its role and healing potential needs further investigation. 1,25-Dihydroxycholecalciferol, the hormonally active vitamin D3 metabolite, is known to exhibit therapeutic effects against breast cancer, mainly by lowering the expression of estrogen receptors and aromatase activity. Previously, the safety of the vitamin D active metabolite (24R)-1,24-dihydroxycholecalciferol (PRI-2191) and 1,25(OH)2D3 analog PRI-2205 was tested, and the in vitro activity of these analogs against different cancer cell lines was studied. We determined the effect of the two vitamin D compounds on anastrozole (An) activity against breast cancer based on antiproliferative activity, ELISA, flow cytometry, enzyme inhibition potency, PCR, and xenograft study. Both the vitamin D active metabolite and synthetic analog regulated the growth of not only estrogen receptor-positive cells (T47D and MCF-7, in vitro and in vivo), but also hormone-independent cancer cells such as SKBR-3 (HER-2-positive) and MDA-MB-231 (triple-negative), despite their relatively low VDR expression. Combined with An, PRI-2191 and PRI-2205 significantly inhibited the tumor growth of MCF-7 cells. Potentiation of the antitumor activity in combined treatment of MCF-7 tumor-bearing mice is related to the reduced activity of aromatase by both An (enzyme inhibition) and vitamin D compounds (switched off/decreased aromatase gene expression, decreased expression of other genes related to estrogen signaling) and by regulation of the expression of the estrogen receptor ERα and VDR.  相似文献

Ce- or Sr-doped LaCoO3 bulk perovskites were prepared by citric acid method as well as 10 wt.% of LaCoO3 was deposited on alumina carrier stabilized with lanthanum. Properties of prepared materials were characterized by determination of surface area, acid-basic properties and XRD, XPS, TPDO2, H2-TPR measurements as well as catalytic activity and selectivity for ethanol combustion was tested. It was found that substitution of La in LaCoO3 with either Sr or Ce has only small effect on its activity in ethanol combustion. Strontium inserted into LaCoO3 structure increases basic character of the perovskite surface as well as selectivity to acetaldehyde (ACA). Substitution of La with cerium has no effect on the concentration of basic sites and does not affect the selectivity to ACA. Activity of LaCoO3-based catalysts in ethanol combustion and their selectivity to ACA formation can be explained on the basis of the presence of both -oxygen species and sites with basic character on the material surface.

Acid-basic properties of supported LaCoO3 are dominated by acidic character of the carrier. Results of XPS and H2-TPR measurements of LaCoO3 supported on La–Al2O3 suggest that perovskite remains in strong interaction with carrier and probably is partially decomposed. Deposition of perovskite on stabilized carrier significantly increases the rate of ethanol combustion.  相似文献

In the Fe–30Ni alloy investigated a martensitic transformation can occur both during quenching or plastic deformation. Martensite formed during plastic deformation, depending on the thermo-mechanical treatment applied, exhibits a different morphology from that achieved during quenching and forms the so-called composite-like structure. The morphology and volume fraction of martensite depends both on strain and temperature. In the present studies Fe–30Ni alloy was deformed by monotonic rolling in one path and perpendicular rolling in the temperature range MDMS. The aim of the investigations was a determination of martensite volume fraction depending on the strain and temperature. To examine the influence of strain, the alloy was deformed by rolling in one path or perpendicular rolling at a temperature of − 30 °C, in the strain range of 10–30%. The dependence of temperature was investigated by rolling with 30% strain in a temperature range from − 30 °C to − 80 °C. The variants of thermo-mechanical treatment performed enabled us to achieve different martensite morphologies and volume fractions. Microstructural analysis was performed by means of light microscopy and transmission electron microscopy. The results of quantitative microstructural analysis of martensite and retained austenite volume fractions formed in different thermo-mechanical treatments were compared with those obtained by magnetic measurements. The fraction of deformation-induced martensite determined varied from 2% to 86%. The partial volume fractions VVMF of martensite formed in different deformation directions were also determined. It was found that the influence of the temperature on the martensite volume fraction is more pronounced than the influence of strain.  相似文献

Extracellular vesicles are evaluated by nanoparticle tracking analysis (NTA), providing information on their hydrodynamic diameters, and by atomic force microscopy (AFM) to calculate their geometric diameters. The aim of this study is to explore the influence of Brownian movements in a sample drop and preparation time on imaging-based measurements and to determine the relationship between the geometric and hydrodynamic sizes of the extracellular vesicles measured by the AFM and the NTA, respectively. Exosomes derived from the human prostate cancer cell line PC3 were evaluated by NTA and AFM, and those results were compared with Monte Carlo simulations. The mean size, evaluated by AFM shortly after application on the mica substrate, is less than its real value. It obtains the correct value faster for a thinner sample drop. Fitting the log-normal distribution to the geometric and hydrodynamic diameters leads to the conclusion that the latter could arise from the former by linear scaling by a factor that could be used to characterize the analyzed extracellular vesicles. The size of the vesicles attached to the mica substrate depends on time. The effect of Brownian motion and stretch of the lipid bilayer should be considered in the context of exosome AFM studies.  相似文献

The aim of the article is to compare two types of fly ash (from the fluidized and pulverized coal combustion process) as a filler for rigid polyurethane foam. Pulverized fly ash (PFA) is widely used in building materials, while fluidized fly ash (FFA) is not currently recycled, but landfilled. The produced rigid polyurethane foams were reinforced with 5 and 10% by weight addition of fly ash from two different types of boilers. The foaming process, physical properties, morphologies and thermal degradation were subject to comparative analysis. The research indicated that fly ash intensifies the reactions of foam synthesis, most commonly, polyurethane (PU) foam with an addition of 10% PFA. What is interesting is that both ashes can be used in PU foam technology as they do not cause deterioration of the physical parameters. As shown, the addition of filler affects the morphology and impairs the brittleness. Additionally, the use of fly ash from coal combustion in the technology of polyurethane materials complies with the guidelines of the circular economy stated in the European Union legislation. Partial replacement of petrochemical components with waste filler also reduces the total energy consumption in the production of PU composites.  相似文献

The pathogenesis of the disorders of calcium metabolism is not fully understood. This review discusses the studies in which metabolomics was applied in this area. Indeed, metabolomics could play an essential role in discovering biomarkers and elucidating pathological mechanisms. Despite the limited bibliography, the present review highlights the potential of metabolomics in identifying the biomarkers of some of the most common endocrine disorders, such as primary hyperparathyroidism (PHPT), secondary hyperparathyroidism (SHPT), calcium deficiency, osteoporosis and vitamin D supplementation. Metabolites related to above-mentioned diseorders were grouped into specific classes and mapped into metabolic pathways. Furthermore, disturbed metabolic pathways can open up new directions for the in-depth exploration of the basic mechanisms of these diseases at the molecular level.  相似文献

The uptake and distribution of doxorubicin in the MCF7 line of breast-cancer cells were monitored by Raman measurements. It was demonstrated that bioavailability of doxorubicin can be significantly enhanced by applying Congo red. To understand the mechanism of doxorubicin delivery by Congo red supramolecular carriers, additional monolayer measurements and molecular dynamics simulations on model membranes were undertaken. Acting as molecular scissors, Congo red particles cut doxorubicin aggregates and incorporated them into small-sized Congo red clusters. The mixed doxorubicin/Congo red clusters were adsorbed to the hydrophilic part of the model membrane. Such behavior promoted transfer through the membrane.  相似文献
