A processing technique to measure cement impedance from ultrasonic pulse-echo waveforms, called normalized single mode (NSM) processing, is described. The technique is based on a physical model that represents the transducer in terms of its continuous plane wave kernel. Narrowband filtering to process a single resonant mode and all processing parameters are normalized to the modal frequency. The performance of NSM and a digital simulation of the present analog commercial processing, W2/W1, is compared for several environmental effects: casing thickness, eccentering, and added Gaussian noise. The computation is based on waveforms from a mathematical model and laboratory data. Results on model data show that NSM processing significantly reduces the impedance errors due to thickness variations, eccentering, and noise. Results on laboratory data indicate that the techniques yield comparable results for smooth machined casings, but that NSM processing significantly reduces measurement error and variance in a sample of the actual oil field casing. 相似文献

Our goal is an automated 2D-image-pair registration algorithm capable of aligning images taken of a wide variety of natural and man-made scenes as well as many medical images. The algorithm should handle low overlap, substantial orientation and scale differences, large illumination variations, and physical changes in the scene. An important component of this is the ability to automatically reject pairs that have no overlap or have too many differences to be aligned well.We propose a complete algorithm, including techniques for initialization, for estimating transformation parameters, and for automatically deciding if an estimate is correct. Keypoints extracted and matched between images are used to generate initial similarity transform estimates, each accurate over a small region. These initial estimates are rank-ordered and tested individually in succession. Each estimate is refined using the Dual-Bootstrap ICP algorithm, driven by matching of multiscale features. A three-part decision criteria, combining measurements of alignment accuracy, stability in the estimate, and consistency in the constraints, determines whether the refined transformation estimate is accepted as correct. Experimental results on a data set of 22 challenging image pairs show that the algorithm effectively aligns 19 of the 22 pairs and rejects 99.8% of the misalignments that occur when all possible pairs are tried. A tandem mass spectrometer for improved transmission and analysis of large macromolecular assemblies
We report the design and first applications of a tandem mass spectrometer (a quadrupole time-of-flight mass spectrometer) optimized for the transmission and analysis of large macromolecular assemblies. Careful control of the pressure gradient in the different pumping stages of the instrument has been found to be essential for the detection of macromolecular particles. Such assemblies are, however, difficult to analyze by tandem-MS approaches, because they give rise to signals above m/z 3,000-4,000, the limit for commercial quadrupoles. By reducing the frequency of the quadrupole to 300 kHz and using it as a narrow-band mass filter, we show that it is possible to isolate ions from a single peak at m/z 22,000 in a window as narrow as 22 m/z units. Using cesium iodide cluster signals, we show that the mass range in the time-of-flight (TOF) analyzer extends beyond m/z 90,000, in theory to more than m/z 150,000. We also demonstrate that the resolution of the instrument is greater than 3,000 at m/z 44,500. Tandem-MS capabilities are illustrated by separating components from heterooligomeric assemblies formed between tetrameric transthyretin, thyroxine, retinol-binding protein, and retinol. Isolation of a single charge state at m/z 5,340 in the quadrupole and subsequent collision-induced dissociation (CID) in the gas-filled collision cell leads to the formation of ions from individual subunits and subcomplexes, identified by their mass and charge in the TOF analyzer. 相似文献

A wavelength-independent method for optical gating, based on the optical Kerr effect, has been demonstrated. Using this method, we produced 100-ps, 10-kW, two-wavelength pulses (10.3 and 10.6 microm) with a signal-to-background ratio contrast of 10(5) by slicing a long CO2 pulse. The occurrence of material adhesion and formation of oversize particles in the product yield during one-pot spheroid production by rotary processing leads to a less predictable process and a decrease in the usable portion of the total product yield obtained from each production run. The use of variable speeds of the rotating frictional base plate during the spheronization run was investigated for achieving optimal spheroid production. When the base plate speed was increased during liquid addition, the greater centrifugal forces generated improved liquid distribution and the mixing of the moist powder mass, resulting in a decrease in the amount of oversize particles formed. When the base plate was maintained at a high speed throughout the run, the amount of oversize particles and mean spheroid size increased, and a greater “between batch” mean spheroid size variability was also observed. The findings showed that, when higher speeds were used, the residence time must be adjusted accordingly to avoid excessive coalescence and growth while maintaining even liquid distribution. A “low-high-low” speed variation during rotary processing may be used to produce spheroids with a narrow size distribution and with a minimal amount of oversize particles in the total product yield. 相似文献

The original formulation of spatially variant apodization for complex synthetic aperture radar imagery concentrated on integer-oversampled data. Noninteger-oversampled data presented previously [IEEE Trans. Aerosp. Electron. Syst. 31, 267 (1995)] suggested use of different weightings in the algorithm. An alternative noninteger-oversampled approach that employs the same apodization concept but uses local spatial interpolation is presented. With this approach the combined image formation, apodization, and detection of 1.3x-versus-2.0x oversampled data can be performed in half the time without loss of image quality. 相似文献

By in silicio analysis, we have discovered that there are seven open reading frames (ORFs) in Saccharomyces cerevisiae whose protein products show a high degree of amino acid sequence similarity to the aryl alcohol dehydrogenase (AAD) of the lignin-degrading fungus Phanerochaete chrysosporium. Yeast cultures grown to stationary phase display a significant aryl alcohol dehydrogenase activity by degrading aromatic aldehydes to the corresponding alcohols. To study the biochemical and the biological role of each of the AAD genes, a series of mutant strains carrying deletion of one or more of the AAD-coding sequences was constructed by PCR-mediated gene replacement, using the readily selectable marker kanMX. The correct targeting of the PCR-generated disruption cassette into the genomic locus was verified by analytical PCR and by pulse-field gel electrophoresis (PFGE) followed by Southern blot analysis. Double, triple and quadruple mutant strains were obtained by classical genetic methods, while the construction of the quintuple, sextuple and septuple mutants was achieved by using the marker URA3 from Kluyveromyces lactis, HIS3 from Schizosaccharomyces pombe and TRP1 from S. cerevisiae. None of the knock-out strains revealed any mutant phenotype when tested for the degradation of aromatic aldehydes using both spectrophotometry and high performance liquid chromatography (HPLC). Specific tests for changes in the ergosterol and phospholipids profiles did not reveal any mutant phenotype and mating and sporulation efficiencies were not affected in the septuple deletant. Compared to the wild-type strain, the septuple deletant showed an increased resistance to the anisaldehyde, but there is a possibility that the nutritional markers used for gene replacement are causing this effect. 相似文献

The behavior of macromolecular systems at different temperatures is often crucial to their biological activity and function. While heat-induced changes of individual proteins are readily monitored by a number of spectroscopic methods, changes in noncovalent complexes of biomolecules are more challenging to interpret. Nanoelectrospray mass spectrometry is becoming increasingly powerful in the study of large noncovalent complexes, and here we describe the design, characterization, and application of a novel probe that allows the thermocontrol of the solution in the electrospray capillary. The transition temperature for the unfolding of the protein lysozyme is readily obtained and correlates closely with that measured by fluorescence spectroscopy, thereby demonstrating the validity of this approach. We apply this technique to the study of the 200-kDa complex of the small heat shock protein TaHSP16.9, revealing both its dissociation into suboligomeric species and an increase in its size and polydispersity at elevated temperatures. In contrast, gas-phase activation of this complex is also carried out and yields a dissociation pathway fundamentally different from that observed for thermal activation in solution. As such, this probe allows the study of the reversible heat-induced changes of noncovalent complexes in a biologically relevant manner. 相似文献
