Calorie restriction (CR) is the most efficacious treatment to delay the onset of age-related changes such as mitochondrial dysfunction. However, the sensitivity of mitochondrial markers to CR and the age-related boundaries of CR efficacy are not fully elucidated. We used liver samples from ad libitum-fed (AL) rats divided in: 18-month-old (AL-18), 28-month-old (AL-28), and 32-month-old (AL-32) groups, and from CR-treated (CR) 28-month-old (CR-28) and 32-month-old (CR-32) counterparts to assay the effect of CR on several mitochondrial markers. The age-related decreases in citrate synthase activity, in TFAM, MFN2, and DRP1 protein amounts and in the mtDNA content in the AL-28 group were prevented in CR-28 counterparts. Accordingly, CR reduced oxidative mtDNA damage assessed through the incidence of oxidized purines at specific mtDNA regions in CR-28 animals. These findings support the anti-aging effect of CR up to 28 months. Conversely, the protein amounts of LonP1, Cyt c, OGG1, and APE1 and the 4.8 Kb mtDNA deletion content were not affected in CR-28 rats. The absence of significant differences between the AL-32 values and the CR-32 counterparts suggests an age-related boundary of CR efficacy at this age. However, this only partially curtails the CR benefits in counteracting the generalized aging decline and the related mitochondrial involvement.  相似文献

Molybdenum hemicarbide (Mo2C), which is widely applied in steel and metal ceramics as well as catalysts, was successfully synthesized by using a simple method of reducing MoO2 powders by CO. It was found that the final reduction product was all Mo2C under both isothermal and nonisothermal conditions. However, the reduction mechanisms were significantly different at lower and higher temperatures: at lower temperatures (1070 to 1342 K), reduction of MoO2 to Mo2C followed a one‐step reaction (simultaneous reduction and carburization), while at higher temperatures (1423 to 1485 K), MoO2 was first reduced to metallic Mo, and then Mo was carburized to Mo2C. Mo2C particles obtained at higher temperatures contained micrometer‐sized surface features which formed during the MoO2 to Mo reduction step.  相似文献

In many implementations of DNA computing, reliable detection of hybridization is of prime importance. We have applied several well-established DNA mutation scanning methods to this problem. Since they have been developed for speed and accuracy, these technologies are very promising for DNA computing. We have benchmarked a heteroduplex migration assay and enzymatic detection of mismatches on a 4 variable instance of 3SAT, using a previously described blocking algorithm. The first method is promising, but yielded ambiguous results. On the other hand, we were able to distinguish all perfect from imperfect duplexes by means of a CEL I mismatch endonuclease assay.  相似文献

Leaves of Brachystegia spiciformis represent a substantial fraction of the total aboveground litter in bush fallow fields with sandy soils in southern Mozambique, where annual rainfall exceeds 600 mm. This species is one of the most important in the miombo woodland that is the natural vegetation of the region. Proper knowledge of the decomposition of its litter is therefore crucial for understanding processes responsible for natural build-up of fertility in agricultural soils abandoned to bush fallow during shifting cultivation. This study investigated the effects of soil water content and soil temperature on loss of organic carbon (C) from decomposing leaves in litterbags with 1 mm mesh size. The litterbags were buried 50 mm deep in recently abandoned agricultural fields cleared of any vegetation (Bare) and in more than 15-year-old bush fallow fields (Fallow) of sites covering a climatic transect with annual rainfall from <400 mm to >1,000 mm. Two patterns of C loss were observed, one in coastal and wetter agroecosystems (rainfall >600 mm) and the other in inland and drier agroecosystems (rainfall <600 mm). In the wetter agroecosystems C loss was faster, whereas in drier agroecosystems it was more sensitive to rainfall pulses. Similarly, C loss was faster in fallow fields than in bare fields. During summer, bare fields reached soil temperatures higher than the estimated upper boundary favourable for C loss from decomposing leaf litter at all sites. A simple dynamic decomposition model describing the C fraction remaining in the litterbags was developed. Coefficients of determination (R 2) for the individual experimental units varied between 0.79 and 0.97. The general model for all sites and fields improved explanation of total variation from 81% to 86% when measured soil temperature and soil water content were used as modifiers of decomposition rate, compared with the standard negative exponential model. Root-mean-square error and systematic bias were 9.7% and 0.5% of initial C, respectively. Decomposition was more strongly affected by soil water content than by soil temperature and explained 75% of total variation. Thus, rainfall is the main driver of C loss from leaf litter in these agroecosystems.  相似文献

Two positive and negative selectable markers were created for use in mammalian cells. They are based on two genes for the resistance to Blasticidin S (BlaS) and on the thymidine kinase (Tk) gene of herpes simplex virus (HSV). The markers can be selected positively by their ability to induce BlaS resistance and negatively on the induced sensitivity towards gancyclovir (GANC). Both constructs are also expressed in Escherichia coli and transfer BlaS resistance to this organism as well, making these markers very suitable for the construction of shuttle vectors.  相似文献

BACKGROUND: Arabinoxylan (AX)‐derived preparations have raised particular interest by the suggestion that they exert prebiotic properties. Therefore an in vivo experiment was conducted to study the effects of xylooligosaccharides (XOS), arabinoxylooligosaccharides (AXOS) and soluble AX (S‐AX) on several groups of gut bacteria of chickens. Chickens were fed a control diet or the same diet supplemented with 2.5 g kg?1 of XOS (average degree of polymerisation (avDP) of 3, average degree of arabinose substitution (avDS) of 0.09), wheat bran‐derived AXOS (avDP 9, avDS 0.34) or wheat endosperm alkali‐solubilised AX (S‐AX, avDP > 6000, avDS 0.5). RESULTS: All treatment groups showed an increase in the number of caecal bifidobacteria after 2 weeks of feeding (P < 0.05), while the treatments did not impact numbers of Enterobacteriaceae and aerobic lactobacilli in the caeca relative to the control. XOS led to a tremendous increase in caecal bifidobacteria counts (108 g?1 for the XOS treatment versus 103 g?1 for the control treatment) after only 1 week of supplementation, while AXOS and S‐AX led to similar strong increases in bifidobacteria counts after 2 weeks of supplementation. Addition of S‐AX to the diet, but not of AXOS or XOS, led to a significantly decreased (P < 0.05) body weight gain after 2 weeks of feeding relative to animals on the control diet, consistent with its known viscosity‐related antinutritive effects in chickens. CONCLUSION: Similar to XOS, AXOS selectively stimulate the presence of bifidobacteria in the caeca of chickens, establishing their bifidogenic effect as a first indicator for prebiotic potential. When using a model to predict the behavior of a physical system of interest, engineers must be confident that, under the conditions of interest, the model is an adequate representation of the system. The process of building this confidence is called model validation. It requires that engineers have knowledge about the system and conditions of interest, properties of the model and their own tolerance for uncertainty in the predictions. To reduce time and costs, engineers often reuse preexisting models that other engineers have developed. However, if the user lacks critical parts of this knowledge, model validation can be as time consuming and costly as developing a similar model from scratch. In this article, we describe a general process for performing model validation for reused behavioral models that overcomes this problem by relying on the formalization and exchange of knowledge. We identify the critical elements of this knowledge, discuss how to represent it and demonstrate the overall process on a simple engineering example.
