Lipases are hydrolytic enzymes that break the ester bonds of triglycerides, generating free fatty acids and glycerol. Extracellular lipase activity has been reported for the nonconventional yeast Kluyveromyces marxianus, grown in olive oil as a substrate, and the presence of at least eight putative lipases has been detected in its genome. However, to date, there is no experimental evidence on the physiological role of the putative lipases nor their structural and catalytic properties. In this study, a bioinformatic analysis of the genes of the putative lipases from K. marxianus L-2029 was performed, particularly identifying and characterizing the extracellular expected enzymes, due to their biotechnological relevance. The amino acid sequence of 10 putative lipases, obtained by in silico translation, ranged between 389 and 773 amino acids. Two of the analysed putative proteins showed a signal peptide, 25 and 33 amino acids long for KmYJR107Wp and KmLIP3p, and a molecular weight of 44.53 and 58.23 kDa, respectively. The amino acid alignment of KmLIP3p and KmYJR107Wp with the crystallized lipases from a patatin and the YlLip2 lipase from Yarrowia lipolytica, respectively, revealed the presence of the hydrolase characteristic motifs. From the 3D models of putative extracellular K. marxianus L-2029 lipases, the conserved pentapeptide of each was determined, being GTSMG for KmLIP3p and GHSLG for KmYJR107Wp; besides, the genes of these two enzymes (LIP3 and YJR107W) are apparently regulated by oleate response elements. The phylogenetic analysis of all K. marxianus lipases revealed evolutionary affinities with lipases from abH15.03, abH23.01, and abH23.02 families.  相似文献

Summary The crystallization and melting behaviour of poly(butylene terephthalate) has been studied in the pure state and in its blends with a polyarylate of bisphenol A and isophthalic/terephthalic acids. Differential scanning calorimetry has been used as experimental technique and the effects of different thermal treatments have been analyzed. Results show the hindrance for the crystallization of poly(butylene terephthalate) imposed by the presence of polyarylate, as well as the existence of multiple melting after isothermal crystallization. Discusses the theoretical and clinical applications of the postmodern paradigm of social constructionism as a basis for counseling culturally different clients. Such applications avoid the trap of perpetuating the status-quo when working with minority and marginalized clientele. A B16 melanoma-specific CD8+ T cell line (AB1) was established from the spleen cells of C57BL/6 mice cured of B16 melanoma with interleukin (IL)-12 treatment. The AB1 line exclusively used T cell receptor Vbeta11. The AB1 cells exhibited a cytolytic activity against both syngeneic B16 melanoma and allogeneic P815 mastocytoma, whereas a cold inhibition assay revealed specificity of the AB1 cells against B16 melanoma. Their lostability to kill a class I loss variant of B16 melanoma was restored by the transfection of H-2Kb gene. In addition, their interferon (IFN)-gamma production was significantly suppressed by the addition of anti-H-2Kb monoclonal antibody, and RT-PCR analysis showed that the AB1 line expressed the mRNA encoding IFN-gamma, but not IL-4 or IL-10. The experiment using synthetic peptides of tyrosinase-related protein-2 (TRP-2) revealed that the AB1 cells could recognize TRP-2(181-188) peptide. Moreover, the AB1 cells showed an in vivo antitumor effect against established pulmonary metastases of B16 melanoma. Overall, these results indicate that the Tc1-type Vbeta11+ AB1 cells exert an antitumor activity against syngeneic B16 melanoma through recognition of TRP-2(181-188) peptide in an H-2Kb-restricted manner.  相似文献

Student academic underachievement is a concern of paramount importance in Europe, where around 15% of the students in the last high school courses do not achieve the minimum knowledge academic requirement. In this paper, we propose a model based on a system of differential equations to study the dynamics of the students’ academic performance in the German region of the North Rhine-Westphalia. This approach is supported by the idea that both good and bad study habits, are a mixture of personal decisions and influence of classmates. This model allows us to forecast the student academic performance by means of confidence intervals over the next few years.  相似文献

We report the effects of the Sr2+ and Ni2+ co-doping of BiFeO3 on the crystal structure and multiferroic properties of Bi1?xSrxFe1-yNiyO3 (x?=?0.05, 0.0?≤?y?≤?0.10, and Δy?=?0.05) that is synthesized using assisted high-energy ball milling. The mixtures of Bi2O3, Fe2O3, SrO and NiO were milled for 5?h, pressed at 900?MPa, and sintered at 800?°C in order to obtain cylindrical test pieces. X-ray diffraction and Rietveld refinement elucidated the effects of Sr2+ and Ni2+ on the crystal structure. Co-doping with SrNi in suitable proportions stabilizes rhombohedral BiFeO3. High contents of Ni2+ promote the precipitation of secondary phases in the forms of NiFe2O4 and Bi25FeO40. The magnetic behavior was examined by means of vibrating sample magnetometry. The results showed a change in the magnetic order from antiferromagnetic for the undoped sample to the ferromagnetic order for the co-doped samples. This change is attributed to the modulations in the magnetic moment due to crystal structure distortions. All samples show high relative permittivity values, which were enhanced by doping with Sr2+. Ni2+ cations increase the dielectric dissipation factor; this enhancement is related to their interactions with cations of a different oxidation state, such as Fe3+, Fe2+, Ni2+, Bi3+ and Sr2+ in the crystal structure of BiFeO3. The magnetoelectric coupling that was evaluated using magnetodielectric measurements was above 4% at 1?kHz for the higher applied magnetic field of 18?kOe.  相似文献

ABSTRACT

In this study, the efficiency of gaseous ozone (O3) injected in the soil as an oxidizing agent for the inactivation of F. oxysporum was evaluated under laboratory conditions. The results show the treatment reached an inactivation efficiency of 76% after an applied dose of 0.40 g O3 kg ?1 soil. This shows that the injection of O3 can be a viable alternative to control pathogenic organisms in the soils. Nevertheless, it is clear that more studies on determining the effects of this treatment on soil quality are needed.  相似文献
