(1) Background: Several properties of silver nanoparticles (AgNPs), such as cytotoxic, anticancer, and antimicrobial activities, have been subjects of intense research; however, important aspects such as nanoparticle aggregation are generally neglected, although a decline in colloidal stability leads to a loss of the desired biological activities. Colloidal stability is affected by pH, ionic strength, or a plethora of biomolecules that interact with AgNPs under biorelevant conditions. (2) Methods: As only a few studies have focused on the relationship between aggregation behavior and the biological properties of AgNPs, here, we have systematically evaluated this issue by completing a thorough analysis of sterically (via polyvinyl-pyrrolidone (PVP)) stabilized AgNPs that were subjected to different circumstances. We assessed ultraviolet–visible light absorption, dynamic light scattering, zeta potential measurements, in vitro cell viability, and microdilution assays to screen both colloidal stability as well as bioactivity. (3) Results: The results revealed that although PVP provided outstanding biorelevant colloidal stability, the chemical stability of AgNPs could not be maintained completely with this capping material. (4) Conclusion: These unexpected findings led to the realization that stabilizing materials have more profound importance in association with biorelevant applications of nanomaterials than just being simple colloidal stabilizers.  相似文献

Long-term exercise induces physiological cardiac adaptation, a condition referred to as athlete’s heart. Exercise tolerance is known to be associated with decreased cardiac passive stiffness. Passive stiffness of the heart muscle is determined by the giant elastic protein titin. The adult cardiac muscle contains two titin isoforms: the more compliant N2BA and the stiffer N2B. Titin-based passive stiffness may be controlled by altering the expression of the different isoforms or via post-translational modifications such as phosphorylation. Currently, there is very limited knowledge about titin’s role in cardiac adaptation during long-term exercise. Our aim was to determine the N2BA/N2B ratio and post-translational phosphorylation of titin in the left ventricle and to correlate the changes with the structure and transverse stiffness of cardiac sarcomeres in a rat model of an athlete’s heart. The athlete’s heart was induced by a 12-week-long swim-based training. In the exercised myocardium the N2BA/N2B ratio was significantly increased, Ser11878 of the PEVK domain was hypophosphorlyated, and the sarcomeric transverse elastic modulus was reduced. Thus, the reduced passive stiffness in the athlete’s heart is likely caused by a shift towards the expression of the longer cardiac titin isoform and a phosphorylation-induced softening of the PEVK domain which is manifested in a mechanical rearrangement locally, within the cardiac sarcomere.  相似文献

Daily variation in the estrogenic activity of effluent released by a modern sewage treatment plant (STP) was measured and its effects on the physiology, behavior, and reproductive success of male fish were evaluated. As measured by an estrogen receptor binding assay, the daily estrogenic activity of this effluent was both high and extremely variable (42 +/- 25.4 [mean +/- SD] ng 17beta-estradiol (E2) equivalents/L; n = 18). Liver VTG mRNA expression in male fathead minnows (FHM) covaried with the binding assay estimates, suggesting that these fluctuations are biologically relevant. Tests which exposed male FHMs to either fluctuating levels of E2, a constant concentration of E2 (time-weighted to reflect average concentrations), or control (no E2) demonstrated that while the estrogenic activity of this effluent was detrimental to male spawning success, the fact that its concentration varied in a daily manner was without additional influence. The variability of the effluent's estrogenicity suggests that studies concerned with the effects of STP effluents should collect multiple daily samples and then test them on an appropriate time-weighted basis.  相似文献

The combination of the sonochemical activation of Ni(NO3)2 and Co(NO3)2 in the presence of Fe(NO3)3 and polyethylene glycol and consecutive heat treatment of the formed metal hydroxides offers a cheap and efficient method for the preparation of nickel ferrite and cobalt ferrite magnetic nanoparticles, which can be successfully applied in the selective capture of fluorescently derivatized N-glycans from human serum. XRD measurement revealed that, besides the ferrite phase, nickel and cobalt oxides also form during heat treatment. The amount of simple metal oxides can be well controlled by the temperature of the heat treatment, since increasing temperature yielded higher spinel content. For both nickel and cobalt, the best heat treatment temperature was found to be 673 K, where the samples contained 84.1% nickel ferrite, and in the case of cobalt, almost pure (99.6%) cobalt ferrite could be prepared. FT-IR and zeta potential measurements indicated the presence of surface OH groups, which aided in the dispersion of the particles in water and, in addition, can promote the adsorption of polar compounds. The practical applicability of the magnetic nanopowders was demonstrated in the purification of fluorescently derivatized N-glycans (from human serum). Cobalt ferrite was found to be the most effective. Owing to the easy preparation and the simplicity of the magnetic separation the pure cobalt ferrite, magnetic nanoparticles could be efficient tools for the selective enrichment of serum N-glycans in HPLC measurements.  相似文献

To date, there has been relatively little research comparing different types of online gamers. The main aim of this study was to provide robust benchmark data on different types of Massively Multiplayer Online Game (MMOG) players using a large sample of online gamers. An online survey was used to recruit 4,374 Hungarian online gamers from websites offering different types of MMOGs. In addition to sociodemographic characteristics, the study also collected data on gaming preference, amount of time spent gaming, amount of money spent on the game, and whether they played at an amateur or professional level. A latent profile analysis of gaming preferences differentiated between eight specific gamer types, of which four types emerged as clear categories, indicating clear preference for a specific type of game (role-playing games, first-person shooter games, real-time strategy games, and other games). Overall, 79% of gamers belonged to these four categories. First-person shooter gamers were almost exclusively male, younger aged, lower educated, and of lower socioeconomic status. Real-time strategy gamers were older. Female gamers were most likely to play “Other” games and/or role-playing games. In relation to time spent gaming, role-playing gamers appeared to be the most vulnerable. The results indicated that a significant number of gamers have clear gaming preferences, and these specific gaming types are associated with significant differences regarding sociodemographic and gaming characteristics of gamers.  相似文献

Transmission-line laser models (TLLMs) are time-domain models suitable for the simulation of complex phenomena in semiconductor lasers. TLLMs include time-domain filters based on transmission-line stubs to model the spectral dependence of the material gain. In this paper, numerical simulations are presented which show that the accuracy of these gain models is dependent on the model's iteration timestep. Analytical formulae are derived that relate the accuracy of the filters to the timestep, filter centre frequency, and filter bandwindth. A new wideband stub filter which allows the material gain to be modelled using a larger timestep is presented. This is equivalent to a digital infinite-impulse-response filter, which is more computationally efficient than finite-impulse-response filters, and is unconditionally stable. The new gain model can improve the computational speed for simulating for multimode Fabry-Perot lasers by a factor of 10–100.  相似文献
