A previous analysis of tRNA sequences suggested a correlation between the absence of a nucleotide at position 47 (nt 47) in the extra loop and the presence of a U13:G22 base pair in the D-stem. We have evaluated the significance of this correlation by determining the in vivo activity of tRNAs containing either a C13:G22 or a U13:G22 pair in tRNA molecules with or without nt 47. Although this correlation might reflect some malfunction of tRNAs lacking nt 47, but containing the C13:G22, assays of the in vivo suppressor activity showed that this tRNA is actually more active than the tRNA with the features found in the database, i.e., a U13:G22 base pair and no nt 47. Moreover, analogous constructs with a GGC anticodon permitted the growth of an Escherichia coli strain deleted for tRNA(Ala)GGC genes equally well. On the other hand, long-term growth experiments with competing E. coli strains harboring the tRNA lacking nt 47, either with the C13:G22 or the U13:G22 base pair demonstrated that the U13:G22 tRNA overtook the C13:G22 strain even when the starting proportion of strains favored the C13:G22 strain. Thus, the preference for the U13:G22 tRNA lacking nt 47 in the sequence database is most likely due to factors that come into play during extended growth or latency rather than to the ability of the tRNA to engage in protein synthesis. 相似文献

To give operators of high-speed, highly-manoeuvrable vehicles the ability to keep pace with their increasingly dynamic environment, stereoscopic 3-D displays may soon replace conventional 2-D displays. Important to the development of stereoscopic 3-D displays is the interaction of perceived depth created by hues (chromostereopsis) and perceived depth created by presenting different images of a single object to the left and right eye of the observer (stereopsis). The purpose of this research is to evaluate the interaction of chromostereopsis and stereopsis on a stereoscopic CRT by determining the level of accuracy with which subjects can properly interpret the relative depth differences of adjacent symbols containing six levels of hue and seven levels of stereoscopic disparity. This research demonstrated that hue, disparity, and the interaction of hue and disparity significantly influenced one's perception of depth on a stereoscopic monitor. The results suggest that caution should be exercised by the stereoscopic 3-D display format designer when choosing hues to represent images located in close proximity on a stereoscopic display. Due to the chromostereoscopic effect on the perception of depth, hues on extreme ends of the colour spectrum should not be used in situations where less than 3.39 arc minutes of disparity difference is being portrayed on a stereoscopic display, unless the hues are consistently being used to alter the depth presented by stereoscopic disparity, or the chromostereoscopic depth resulting from certain hues is consistently nullified by altering disparity levels accordingly. 相似文献

Background: High-level occupational vinyl chloride (VC) exposures have been associated with hepatic hemangiosarcoma, which typically develops following a long latency period. Although VC is genotoxic, a more comprehensive mode of action has not been determined and diagnostic biomarkers have not been established. The purpose of this study is to address these knowledge gaps through plasma metabolomics. Methods: Plasma samples from polyvinyl chloride polymerization workers who developed hemangiosarcoma (cases, n = 15) and VC exposure-matched controls (n = 17) underwent metabolomic analysis. Random forest and bioinformatic analyses were performed. Results: Cases and controls had similar demographics and routine liver biochemistries. Mass spectroscopy identified 606 known metabolites. Random forest analysis had an 82% predictive accuracy for group classification. 60 metabolites were significantly increased and 44 were decreased vs. controls. Taurocholate, bradykinin and fibrin degradation product 2 were up-regulated by greater than 80-fold. The naturally occurring anti-angiogenic phenol, 4-hydroxybenzyl alcohol, was down-regulated 5-fold. Top affected ontologies involved: (i) metabolism of bile acids, taurine, cholesterol, fatty acids and amino acids; (ii) inflammation and oxidative stress; and (iii) nicotinic cholinergic signaling. Conclusions: The plasma metabolome was differentially regulated in polyvinyl chloride workers who developed hepatic hemangiosarcoma. Ontologies potentially involved in hemangiosarcoma pathogenesis and candidate biomarkers were identified. 相似文献

ABSTRACTBecause there are fewer tools available to probe the interactions therein, the effect of the fundamental chemistry of the organic diluent on solvent extraction equilibria has been under-characterized relative to the aqueous. As a result, diluents for solvent extraction are often selected for an application not for their utility as a medium for reaction, but for other (often equally) important reasons (like low flammability). To begin to improve this imbalance in the science, twenty different diluents have been used in a study of the extraction of radiotracer 152/154Eu3+ from dilute nitric acid solutions using the extractant 2-ethylhexylphosphonic acid mono-2-ethylhexyl ester (HEH[EHP]). To increase the utility of the study and to honor the memory of Professor Jan Rydberg, this investigation was conducted by a cadre of comparatively inexperienced separation scientists (who are as a result no longer inexperienced separation scientists) as a radioanalytical chemistry and solvent extraction educational exercise. Slope analysis was used to determine the apparent stoichiometry of the extracted metal complex. The results discussed in the following indicate that, while the pH dependence exhibits the expected three H+ exchanged per metal ion extracted, the extractant dependence suggests that the number of protonated extractant molecules in the extracted complex changes with the organic diluent. The experimentally observed “extractant dependency” ranges from 2.5 to 3.0 dimer equivalent molecules per extracted metal ion. Ironically, in the diluents exhibiting the highest apparent M:(HA)3 stoichiometry, HEH[EHP] extracts Eu3+ less efficiently. Europium luminescence spectroscopy was used to probe for changes in the first coordination sphere of the complex in different diluents. The coupled-dipole approximation has been used to model polarized light-scattering data obtained from the sperm of the octopus Eledone cirrhosa. Mueller scattering-matrix elements (which describe how a sample alters the intensity and degree of polarization of scattered light) were measured as a function of angle. The sample was modeled as a helical fiber believed to correspond to a DNA protein complex. It was necessary to propose an inherent anisotropy in the polarizability of the fiber in order to fit the data. The direction of the principle axes of the polarizability were determined by comparing the model with experimental data. The results suggest that the 2-nm DNA fibers are perpendicular to the thick fiber that defines the helical geometry of the octopus sperm head. 相似文献

The thermal properties were investigated for hot‐pressed zirconium diboride—transition‐metal boride solid solutions. The transition‐metal additives included hafnium, niobium, tungsten, titanium, and yttrium. The nominal additions were equivalent to 3 at.% of each metal with respect to zirconium. Powders were hot‐pressed to nearly full density at 2150°C using 0.5 wt% carbon as a sintering aid. Thermal diffusivity was measured using the laser flash method. Thermal conductivity was calculated from the thermal diffusivity results using temperature‐dependent values for density and heat capacity. At 25°C, the thermal conductivity ranged from 88 to 34 W·(m·K)?1 for specimens with various additives. Electrical resistivity measurements and the Wiedemann–Franz law were used to calculate the electron contribution of the thermal conductivity and revealed that thermal conductivity was dominated by the electron contribution. The decrease in thermal conductivity correlated with a decrease in unit cell volume, indicating that lattice strain may affect both phonon and electron transport in ZrB2. 相似文献

Environmental flows are now a standard part of sustainable water management globally but are only rarely implemented. One reason may be insufficient engagement of stakeholders and their priority outcomes in the environmental flow‐setting process. A recent environmental flow assessment (EFA) in the Kilombero basin of the Rufiji River in Tanzania concentrated on a broad‐based investigation of stakeholders' use and perceptions of the ecosystem services provided by the river as a framework for the assessment of flow regimes that would maintain them. The EFA process generally followed the Building Block Methodology but within an enhanced stakeholder engagement framework. Engagement began with the involvement of institutional stakeholders to explain the purpose of the EFA and to elicit their priority outcomes. Extensive interactions with direct‐use stakeholders followed to investigate their uses of and priorities for the rivers. Results were used by the EFA specialist team in choosing flow indicators and defining measurable environmental objectives. The specialists then met to reach a consensus of the flow requirements. The EFA results were lastly reported back to stakeholders. During the Kilombero EFA, we learned that stakeholders at all levels have a good awareness of the natural services provided by a healthy river and can contribute to the setting of environmental objectives for the rivers and floodplain. These can be factored into the biophysical assessments of river flows required to maintain habitats, processes, water quality, and biodiversity. It is therefore important to allocate significant resources to stakeholder engagement. It now remains to be seen if enhanced stakeholder engagement, including the increased understanding and capacity built among all stakeholders, will increase support for the implementation of the recommended flows. 相似文献
