Pilot studies in human populations have demonstrated a correlation between the level of antigen receptor trans-rearrangements and risk (at the population level) of lymphoid malignancy. Irradiation of newborn severe combined immune deficiency mice results in an increased risk of subsequent development of thymic lymphoma (100% of mice so irradiated are dead of thymic lymphoma by 20 weeks of age). We, therefore, assayed the occurrence of trans-rearrangements in this well-controlled mouse mutant system and found a 50-100-fold increase in the absolute number of TCRGV-TCRBJ trans-rearrangements compared to unirradiated littermates (and a comparable fold increase over age-matched BALB/c mice) at 2 weeks following irradiation. We also found a marked disproportion in generating trans-rearrangements versus intralocus rearrangements in the severe combined immune deficiency system compared to BALB/c, independent of irradiation. The trans-rearrangements noted were polyclonal in nature. These data, again, suggest that the absolute level of antigen receptor trans-rearrangements may serve as a biomarker of lymphoma risk.  相似文献

Spectrophotometric data followed by a suitable treatment of chemometric analysis were used for the simultaneous determination of monosodium glutamate (MSG), guanosine 5′-monophosphate (GMP) and inosine 5′-monophosphate (IMP) in stock cube samples, without any previous extraction step. By this way, the overlapping of the absorption spectra was resolved using a PLS-1 model. The concentration for experimental calibration matrix were varied between 5.03–34.2 μg mL−1 for IMP and GMP, and 448–1399 μg mL−1 for MSG. The relative errors of prediction (REPCV %) were 1.8, 2.8 and 3.1 for IMP, GMP and MSG, respectively.  相似文献

In this paper, we present a comprehensive discussion of the results obtained after in vitro exposure of human fetal fibroblasts and human adult fibroblasts to pulsed radiation in a wide band between 100 and 150 GHz and to continuous wave radiation at 25 GHz. The addition of antioxidants to frying oil reduces postprandial oxidative stress and the inflammatory response. ER stress may trigger both inflammation and oxidative stress processes. We aimed to determine the biological effects of the intake of four models of frying oils on postprandial ER stress in peripheral blood mononuclear cells. Twenty obese people received four breakfasts following a randomized crossover design, consisting of muffins made with different oils (virgin olive oil (VOO), sunflower oil (SFO), and a mixture of seed oils (SFO/canola oil) with either dimethylpolysiloxane (SOD) or natural antioxidants from olives (SOP) added), which were previously subjected to 20 heating cycles. ER stress was assessed by measuring the mRNA levels of sXBP1, BiP, CRT, and CNX in peripheral blood mononuclear cells. Our study showed that the intake of the muffins made with SFO induced the postprandial increase of the mRNA levels of the ER stress‐sensor sXBP1, and the ER stress related chaperones BiP and CRT (all p‐values <0.05). The harmful effects associated with the use of SFO as frying oil, in terms of inflammatory response and postprandial oxidative stress, may be partially mediated by the induction of postprandial ER stress.  相似文献
