Insulin resistance and type 2 diabetes are associated with elevated circulating levels of nonesterified FA (NEFA) and lipoprotein remnants. The dyslipidemia is an important contributor to the excess arterial disease associated with insulin resistance and type 2 diabetes, but the mechanisms involved are elusive. In the present study we examined the effect of NEFA on macrophages. For this purpose, we utilized human macrophages, prepared by treating THP-1 monocytes with phorbol ester. We found that albumin-bound NEFA at physiological levels increase the secretion of granulocyte macrophage-colony stimulating factor (GM-CSF) by the THP-1 macrophages in a dose-dependent manner. The effect was registered as an increase in mRNA, and the amount of GM-CSF secreted correlated with the accumulation of TAG and DAG in the cell. The NEFA-induced rise in GM-CSF appeared to be mediated by activation of protein kinase C, probably acting on extracellular signal-regulated kinases 1 and 2 and being calcium dependent. We speculate that increased secretion of GM-CSF by resident macrophages in the intima exposed chronically to high levels of NEFA, such as those present in insulin resistance, may contribute to a proatherogenic response of arterial cells.  相似文献

Acidianus brierleyi was demonstrated to remove pyritic sulphur from coal. A. brierleyi was also found capable of catalyzing the removal of what is normally reported as organic sulphur from coal 171US34. A kinetic analysis was performed by assuming a first order reaction. The first order kinetics allowed a comparison with literature data for Thiobacillus ferrooxidans. Also, formation of jarosite was taken into account in the analysis. The simple first order kinetics was observed to fit the data on removal of sulphur satisfactorily.  相似文献

Theminimum-degree greedy algorithm, or Greedy for short, is a simple and well-studied method for finding independent sets in graphs. We show that it achieves a performance ratio of (Δ+2)/3 for approximating independent sets in graphs with degree bounded by Δ. The analysis yields a precise characterization of the size of the independent sets found by the algorithm as a function of the independence number, as well as a generalization of Turán’s bound. We also analyze the algorithm when run in combination with a known preprocessing technique, and obtain an improved performance ratio on graphs with average degree , improving on the previous best of Hochbaum. Finally, we present an efficient parallel and distributed algorithm attaining the performance guarantees of Greedy. Gordon Gekko [29]. A preliminary version of this paper appeared at the 26th ACM Symposium on Theory of Computing, 1994. This paper presents a method for the detection of fouling in a cross-flow heat exchanger. A numerical model is used to generate data when the heat exchanger is clean and corresponding data when fouling occurs. In a first step, the model is used to generate a long time series by simulating a clean heat exchanger. This allows the determination of a neural network model of the heat exchanger. Then, hundred sets of data are generated by simulating a fouled heat exchanger and it is checked that the simple Cusum test can be used to detect fouling without any false alarm, whatever the reference time series is.  相似文献

We have created protein domains with extreme surface charge.These mutated domains allow for ion-exchange chromatographyunder conditions favourable for selective and efficient capture,using Escherichia coli as a host organism. The staphylococcalprotein A-derived domain Z (Zwt) was used as a scaffold whenconstructing two mutants, Zbasic1 and Zbasic2, with high positivesurface charge. Far-ultraviolet circular dichroism measurementsshowed that they have a secondary structure content comparableto the parental molecule Zwt. Although melting temperatures(Tm) of the engineered domains were lower than that of the wild-typeZ domain, both mutants could be produced successfully as intracellularfull-length products in E.coli and purified to homogeneity byion-exchange chromatography. Further studies performed on Zbasic1and Zbasic2 showed that they were able to bind to a cation exchangereven at pH values in the 9 to 11 range. A gene fusion betweenZbasic2 and the acidic human serum albumin binding domain (ABD),derived from streptococcal protein G, was also constructed.The gene product Zbasic2–ABD could be purified using cation-exchangechromatography from a whole cell lysate to more than 90% purity.  相似文献

The possibility of improving the keeping qualities of field-dried and hayloft-dried hay, with dry matter contents of 55 and 73% by adding sodium chloride, formic acid and propionic acid was investigated. Field-dried, sodium-chloride-treated hay was stored in containers, while controls and acid-treated hay were pressed into high-density bales. All hayloft-dried hay was stored in containers. Fungal attacks were very heavy, especially in the baled hay. The contaminating flora was examined at the temperature maximum and after 40 and 32 days storage for field-dried and hayloft-dried hay respectively. The genera Penicillium, Cladosporium and Trichothecium were predominant in hayloft-dried hay, and the differences between varying dry matter contents were insignificant. Mostly Aspergillus spp., but also Penicillium spp., occurred in the field-dried hay at 73% dry matter content but at 55% the flora consisted mostly of Aspergillus flavus, and the formic acid-treated hay contained almost a pure culture of this fungus. In addition, aflatoxin B1 and G1 (635–1000 μg/l) were demonstrated in this formic acid-treated hay.

Toxin formation in isolates of A. flavus from differently treated hay specimens was also investigated on various substrates.  相似文献

Aluminium bond wires are exposed to pulsed current with varying duty cycles, pulse widths and current amplitudes. How these parameters influence the temperature rise in the wire and what it takes to damage or break it are studied. There are two possible failure modes. One is burn-out due to high current amplitude, the other is metallurgical flexure fatigue at the bond heel. Both failure modes are described. The thermal time constant of the wire is determined for different wire lengths. SEM studies of wires during pulsing are made to see structural changes in the wire when it heats. The bond wire may suffer from damage, which affects the reliability, but it may still function electrically.  相似文献

Waste activated sludge was thickened by biological flotation without polymer flocculant dosage. The BIOFLOT® process utilizes the denitrifying ability of activated sludge bacteria. Gaseous products of anaerobic nitrate reduction cause spontaneous flotation of the sludge suspended solids. Laboratory tests confirmed the dependence of sludge thickening efficiency on available nitrate concentration, flotation time and temperature. Full-scale experiments were performed in a fully automatized unit for discontinuous sludge thickening from wastewater treatment plants with a capacity of up to 5000 I.E. Waste activated sludge from wastewater treatment plants at Pisek. Milevsko and Björnlunda was thickened from 6.2, 10.7 and 3.5 g/l MLSS to 59.4, 59.7 and 66.7 g/t MLSS, respectively. Concentrations of COD, ammonium and phosphate ions were decreased in sludge water. The average nitrate consumption for bioflotation was 21.2 mg NO1 per 1 g of MLSS of activated sludge. Flotation time ranged from 4 to 48 h.  相似文献

In laser projection systems the observer in the far field of the image points on the screen will recognize serious speckle noise. There are many methods to reduce or eliminate speckles in the near field by reducing or eliminating temporal or spatial coherence of the laser. But for the far field it is hardly possible to change the coherence properties of laser sources so that speckles will disappear. We propose a new method for eliminating speckles in the far field by using a diffractive optical element. The intensity modulation depth in the far-field speckle pattern can be reduced to a few percent while good beam quality is preserved.  相似文献
