Anti-inflammatory activity of myricetin-3-O-beta-D-glucuronide and related compounds

OBJECTIVE AND DESIGN: The anti-inflammatory effect of myricetinglucuronide (MGL) was investigated and structurally-related compounds were compared to examine the structure/activity-relationship in carrageenan-induced rat paw edema. MATERIALS AND SUBJECTS: In vitro studies were performed using rat basophilic leukemia (RBL-1) cells, human polymorphonuclear leukocytes (PMNL), COX-1 from ram seminal vesicle, COX-2 from sheep placenta and human venous blood. For the in vivo tests male Wistar rats were used, for the ex vivo test perfused rabbit ears. TREATMENT: 1-300 microg/kg MGL or myricetinmethylglucuronate and 0.1-5 mg/kg other related compounds administered p.o. (carrageenan edema). 5, 50 and 150 microg/kg MGL p.o. for 14 days (Freund's adjuvant arthritis), 5 and 50 microg/kg p.o. for 6 days (ulceration). METHODS: Anti-inflammatory effects were measured in carrageenan edema and in adjuvant arthritis. Incidence of gastric lesions was tested in an ulcerogenicity model in vivo. Influence on COX was determined in the perfused rabbit ear, in PMNL and in a test assay using COX-1 and COX-2. 5-LOX activity was studied using PMNL and RBL-1. The influence on platelet aggregation was evaluated measuring light transmission. RESULTS: MGL exerted a marked and dose-dependent anti-inflammatory effect in acute (carrageenan edema, ED50 15 microg/kg, indomethacin ED50 10 mg/kg) and chronic (adjuvant arthritis, inhibition at 150 microg/kg 18.1 % left paw, 20.6% right paw, indomethacin 3 mg/kg 18.0% and 19.4%)) models of inflammation. In the perfused rabbit ear 1 microg MGL inhibited the release of PGI2, PGD2 and PGE2 to the same extent as 1 microg indomethacin. The inhibition of COX-1 in the intact cell system was IC50 = 0.5 microM, that of indomethacin 0.0038 microM. In the isolated enzyme preparations of COX-1 and COX-2 the IC50 was 10 microM and 8 microM, that of indomethacin 9.2 mM and 2.4 microM. In the RBL-1 and PMNL test assay the inhibition of 5-LOX was 0.1 microM and 2.2 microM. An orally administered dose of 50 microg/kg/day induced no gastric ulcers in rats treated for 6 days. The investigations on carrageenan edema showed a close relationship between the structure of MGL and the anti-inflammatory effect. CONCLUSIONS: MGL is a COX-1, COX-2 and 5-LOX inhibitor. In view of the moderate in vitro activity and the very potent in vivo activity an additive mechanism must be involved. Small changes in the molecular structure lead to the loss or reduction of the anti-inflammatory activity.     

The effect of large strains on ageing of a filled elastomer

Strain endurance experiments on a filled elastomer were performed at several temperatures. It was found that superimposed prestraining accelerated the ageing process. This process was caused by microstructural changes in the elastomer. In order to evaluate the mechanical properties of a filled elastomer it is necessary to take into consideration not only the ageing effects but also the loading history.     

Management of rhesus isoimmunization by preimplantation genetic diagnosis

Intracranial osteolipomas are rare lesions which occur in the region of the tuber cinereum. All cases reported to date have been incidental autopsy findings. We describe a patient who presented with a variety of neurological symptoms and had a typical osteolipoma surgically removed.     

Parotid gland dysfunction in a murine model of acute graft versus host disease [aGVHD]

We report a rare case of meningeal melanocytoma in the left frontal region. A 45-year-old man complained of a headache. Magnetic resonance (MR) scanning showed characteristic patterns: a slightly high signal intensity mass in the left frontal region on the T1-weighted image and a low signal intensity on the T2-weighted image. The patient underwent gross total removal of the tumor. The postoperative course was uneventful. After two years, there was a small local recurrence. The histological finding of the tumor showed meningeal melanocytoma. To our knowledge, this is the second published report of a meningeal melanocytoma in the supratentorial region.     

Single-pulse, two-line temperature-measurement technique using KrF laser-induced O(2) fluorescence

A new single-pulse, two-line laser-induced O(2) fluorescence (LIF) temperature-measurement technique was demonstrated. The fluorescence spectrum obtained with multichannel detection following simultaneous excitation of two coincident transitions in the 0-6 and the 2-7 bands of the B(3)Σ(-)(u)-X(3)Σ(-)(g) Schumann-Runge system was used to determine the gas temperature. The rms error of 100-pulse average LIF temperature measurements, referenced to their corresponding thermocouple measurements, was 1.3% over a temperature range of 1300-1800 K in atmospheric air. Photon shot noise was found to be the primary source of uncertainty for these measurements in a quiescent environment. Single-pulse temperature-measurement uncertainties (1 σ) ranged from approximately 13% at 1300 K to 7% at 1800 K.     

One-dimensional antireflection gratings in (100) silicon: a numerical study

The antireflection properties of V-grooved gratings in (100)crystalline silicon are studied numerically by use of rigorouselectromagnetic theory. This study shows that these gratings canexhibit antireflective behavior only for TM-polarizedradiation. The V-grooved structures are analyzed as a function ofgrating period, duty cycle, and depth of a SiO(2) mask layerthat is added to the tops of the V-grooved mesas. Specificantireflection grating designs (the duty cycle and depth versus theperiod) are presented that illustrate TM-polarized reflectivity muchless than 10(-3) with periods as high as 80% the wavelengthof incident radiation. These designs exhibit good tolerance tofabrication errors and grating's plane deviations in aplanar-diffraction mounting.     

Usage of bioactivated PCL nanofiber as a fluoxetine capturing matrix in milk

ABSTRACT

In this study, for the first time, polycaprolactone (PCL) nanofiber matrix was bioactivated for the removal of fluoxetine from milk. Bioactivated nanofiber was prepared by immobilizing fluoxetine antibody on PCL nanofiber matrix. The fluoxetine removal efficiency of bioactivated nanofiber in milk was found to be approximately 93.6%. This removal did not significantly change the biochemical composition of milk. In conclusion, as a novel product, bioactivated nanofibrous PCL matrix can be used for the removal of drugs or unwanted chemicals from breast milk or from other fluids.     

Aptitude development of civil engineers for the management of human resources in construction projects

The paper analyses the singular role human resource management (HRM) plays within the context of construction management, where it differs significantly from its application in other disciplines. A synthesis of HRM research work, in general and in the construction industry in particular, combined with the writer's experience in developing and teaching specialized HRM courses in the civil engineering faculties of two universities, leads to the conclusion that: prospective construction project managers must receive HRM training in appreciably greater scope and depth than is given in most civil engineering schools today; and the peculiar environmental conditions and technology prevalent in construction warrant a specialized adaptation of HRM skills. A syllabus is outlined and discussed.
