The contents of Alu- and L1-containing TaqI restriction fragments were assessed by Southern blot analyses across YAC contigs already assembled by other means and localized within Xq24-q28. Fingerprinting patterns of YACs in contigs were concordant, and using software based on that of M. V. Olson et al. (1986, Proc. Natl. Acad. Sci. USA 83: 7826) to analyze digitized data on fragment sizes, fingerprinting itself could establish matches among about 40% of a test group of 435 YACs. At 100-kb resolution, both repetitive elements were found throughout the region, with no apparent enrichment of Alu or L1 in DNA of G compared to that found in R bands. However, consistent with a random overall distribution, delimited regions of up to 100 kb contained clusters of repetitive elements. The local concentrations may help to account for the reported differential hybridization of Alu and L1 probes to segments of metaphase chromosomes.  相似文献

Slow strain rate tests were performed on quenched and tempered AISI 4340 steel to measure the extent of hydrogen embrittlement caused by electroplating with zinc-cobalt alloys. The effects of bath composition and pH were studied and compared with results for electrodeposited cadmium and zinc-10%nickel. It was found that zinc-1%cobalt alloy coatings caused serious hydrogen embrittlement (EI 0.63); almost as severe as that of cadmium (EI 0.78). Baking cadmium plated steel for 24 h at 200 °C gave full recovery of mechanical properties but specimens plated with zinc-1%cobalt and then baked still failed in 89% of the time of unplated controls. It was shown that hydrogen uptake and embrittlement could be controlled by depositing thin layers of cobalt or nickel at the steel/coating interface. For example, the least embrittlement was caused by zinc-10%nickel (EI 0.037) due to a nickel rich layer with very low hydrogen diffusion coefficient that formed during the initial stages of electroplating. Similarly, a 0.5 μm nickel layer was effective in lowering the embrittlement caused by zinc-1%cobalt to that of zinc-10%nickel. Furthermore, a 0.5 μm cobalt layer deposited before a zinc-1%cobalt coating gave virtually 100% recovery of mechanical properties after baking.  相似文献

Investigating the dynamical and physical properties of cosmic dust can reveal a great deal of information about both the dust and its many sources. Over recent years, several spacecraft (e.g., Cassini, Stardust, Galileo, and Ulysses) have successfully characterised interstellar, interplanetary, and circumplanetary dust using a variety of techniques, including in situ analyses and sample return. Charge, mass, and velocity measurements of the dust are performed either directly (induced charge signals) or indirectly (mass and velocity from impact ionisation signals or crater morphology) and constrain the dynamical parameters of the dust grains. Dust compositional information may be obtained via either time-of-flight mass spectrometry of the impact plasma or direct sample return. The accurate and reliable interpretation of collected spacecraft data requires a comprehensive programme of terrestrial instrument calibration. This process involves accelerating suitable solar system analogue dust particles to hypervelocity speeds in the laboratory, an activity performed at the Max Planck Institut fu?r Kernphysik in Heidelberg, Germany. Here, a 2 MV Van de Graaff accelerator electrostatically accelerates charged micron and submicron-sized dust particles to speeds up to 80 km s(-1). Recent advances in dust production and processing have allowed solar system analogue dust particles (silicates and other minerals) to be coated with a thin conductive shell, enabling them to be charged and accelerated. Refinements and upgrades to the beam line instrumentation and electronics now allow for the reliable selection of particles at velocities of 1-80 km s(-1) and with diameters of between 0.05 μm and 5 μm. This ability to select particles for subsequent impact studies based on their charges, masses, or velocities is provided by a particle selection unit (PSU). The PSU contains a field programmable gate array, capable of monitoring in real time the particles' speeds and charges, and is controlled remotely by a custom, platform independent, software package. The new control instrumentation and electronics, together with the wide range of accelerable particle types, allow the controlled investigation of hypervelocity impact phenomena across a hitherto unobtainable range of impact parameters.  相似文献

For understanding both the environmental behavior and developing remediation treatments for chromium ore processing residue (COPR) it is important to identify all the potentially soluble sources of Cr(VI). Hydrogarnet has been identified as a major phase in COPR and it has been previously speculated that it has a capacity to host Cr(VI). Here we provide direct evidence of this capacity by demonstrating the incorporation of Cr(VI) into laboratory synthesized hydrogarnet. Electron microscopy and energy dispersive X-ray microanalysis show incorporation of approximately 17000-22000 mg Cr(VI) kg(-1) hydrogarnet. X-ray powder diffraction data show that peak intensities are altered by chromium substitution and that chromium substituted hydrogarnets have a smaller unit cell than the pure Ca-Al end member. This is consistent with substitution of hydroxyl tetrahedra by smaller chromate tetrahedra. Electron energy loss spectroscopy confirms the tetrahedral coordination and hexavalent oxidation state of chromium in the hydrogarnets. The maximum amount of hexavalent chromium that can be introduced synthetically corresponds to a replacement of about one out of every eight hydroxyl tetrahedral per unit cell by a CrO4(2-) tetrahedra and tallies closely with the amount of chromium measured in hydrogarnets from COPR. Chromium-bearing hydrogarnet is the most abundant crystalline phase in millions of tons of COPR contaminating land around Glasgow, Scotland, and was recently identified in COPR from sites in North America. Calculations based on its abundance and its Cr(VI) content indicate that hydrogarnet can host as much as 50% of the Cr(VI) found in some COPR samples.  相似文献

Idiopathic thrombocytopenic purpura (ITP, also known as immune thrombocytopenic purpura) in adults is principally a disease of young women. Although in some patients the onset is acute and complete resolution occurs, in most patients, the onset is insidious and the course is chronic. In spite of the relative frequency of ITP, there are important unresolved issues in its diagnosis and management. For this reason, the American Society of Hematology (ASH) chose ITP as the disease topic for its initial sponsored practice guideline in 1993. A major conclusion of the published guideline was the lack of firm evidence on which to base diagnostic procedures and management strategies. This review describes the clinical features of ITP in adults, emphasizes the principal unresolved issues in diagnosis and management, and outlines the critical areas for future research.  相似文献

Six wells were drilled in the North Falkland Basin in 1998. Five of these wells recorded oil shows, and up to 32% gas was also recorded in mud returns to the rig floor. However, none of the wells encountered commercially viable petroleum accumulations.
The syn-rift and early post-rift intervals contain thick, lacustrine claystones with oil source potential as indicated by TOC values up to 7.5% and Rock-Eva1 S2 values of up to 102 kg HC per tonne of rock. These source rocks were immature or only marginally mature in five of the wells but had attained maturity in one of them. Modelling suggests that the main source interval may well be within the peak oil generation window in deeper, undrilled parts of the basin. Calculations of the amount of oil expelled range up to 60 billion barrels.
Most of the wells tested a closely-related set of plays in large structures asSociated with a sandstone interval near the top of the late syn-rifi to early post-rij? source-rock succession. Post-drilling geological modelling of the basin suggests that oil is unlikely to have migrated into this sandstone play at the localities tested, and that the wells consequently failed largely due to a lack of charge. Howevel; the play maintains exploration potential elsewhere. Other plays, particularly those stratigraphically asSociated with the base rather than the top of the source rock, may have a higher chance of exploration success.  相似文献

This paper focuses on the performance of the time-division multiple-access technique with fixed allocation scheme when the memory buffer size at each ground station is finite. Specifically, a queueing model is formulated in order to derive the expected waiting time of the messages in the buffer and the probability of message blocking. Two cases are discussed. single-packet messages and multipacket messages, both with multiple time slots available to the ground station. Numerical results also are provided to facilitate determining the best tradeoff between memory buffer size and the performance parameters. These results suggest that assigning a single slot per ground station is more efficient than multiple slots.  相似文献

In a previous communication we showed from rapid isotopic exchange measurements that the exchangeability of the substrate water at the water oxidation catalytic site in the S3 state undergoes biphasic kinetics although the fast phase could not be fully resolved at that time [Messinger, J., Badger, M., and Wydrzynski, T. (1995) Proc. Natl. Acad. Sci. U.S.A. 92, 3209-3213]. We have since improved the time resolution for these measurements by a further factor of 3 and report here the first detailed kinetics for the fast phase of exchange. First-order exchange kinetics were determined from mass spectrometric measurements of photogenerated O2 as a function of time after injection of H218O into spinach thylakoid samples preset in the S3 state at 10 degreesC. For measurements made at m/e = 34 (i. e., for the mixed labeled 16,18O2 product), the two kinetic components are observed: a slow component with k1 = 2.2 +/- 0.1 s-1 (t1/2 approximately 315 ms) and a fast component with k2 = 38 +/- 4 s-1 (t1/2 approximately 18 ms). When the isotopic exchange is measured at m/e = 36 (i.e., for the double labeled 18,18O2 product), only the slow component (k1) is observed, clearly indicating that the substrate water undergoing slow isotopic exchange provides the rate-limiting step in the formation of the double labeled 18,18O2 product. When the isotopic exchange is measured as a function of temperature, the two kinetic components reveal different temperature dependencies in which k1 increases by a factor of 10 over the range 0-20 degreesC while k2 increases by only a factor of 3. Assuming simple Arrhenius behavior, the activation energies are estimated to be 78 +/- 10 kJ mol-1 for the slow component and 39 +/- 5 kJ mol-1 for the fast component. The different kinetic components in the 18O isotopic exchange provide firm evidence that the two substrate water molecules undergo separate exchange processes at two different chemical sites in the S3 state, prior to the O2 release step (t1/2 approximately 1 ms at 20 degreesC). The results are discussed in terms of how the substrate water may be bound at two separate metal sites.  相似文献
