The white-rot basidiomycete Pleurotus ostreatus produced sweet flavor compounds on a liquid medium. The major and minor compounds identified by GC-MS analysis were p-anisaldehyde (4-methoxybenzaldehyde) and 3-chloro-p-anisaldehyde (3-chloro-4-methoxybenzaldehyde), respectively. p-Anisaldehyde was only produced under static culture conditions. Differences in the type and quantity of flavor compounds produced among wild strains of P. ostreatus were observed. Aryl alcohol oxidase and manganese peroxidase activities increased parallel to the production of p-anisaldehyde. These results indicated that the biosynthesis of p-anisaldehyde is concerned in generating H2O2-activated peroxidase in the lignin-degradation system. Addition of L-tyrosine to the culture led to higher production of p-anisaldehyde. The flavor extract, which contains p-anisaldehyde, exhibited antimicrobial activity against Bacillus subtilis, Pseudomonas aeruginosa, Aspergillus niger and Fusarium oxysporum.  相似文献

The photosynthetic and anatomical responses of bryophytes to changes in gravity will provide crucial information for estimating how these plant traits evolved to adapt to changes in gravity in land plant history. We performed long-term hypergravity experiments at 10g for 4 and 8 weeks using the moss Physcomitrella patens with two centrifuges equipped with lighting systems that enable long-term plant growth under hypergravity with irradiance. The aims of this study are (1) to quantify changes in the anatomy and morphology of P. patens, and (2) to analyze the post-effects of hypergravity on photosynthesis by P. patens in relation to these changes. We measured photosynthesis by P. patens for a population of gametophores (e.g., canopy) in Petri dishes and plant culture boxes. Gametophore numbers increased by 9% for a canopy of P. patens, with 24–27% increases in chloroplast sizes (diameter and thickness) in leaf cells. In a canopy of P. patens, the area-based photosynthesis rate (A canopy) was increased by 57% at 10g. The increase observed in A canopy was associated with greater plant numbers and chloroplast sizes, both of which involved enhanced CO2 diffusion from the atmosphere to chloroplasts in the canopies of P. patens. These results suggest that changes in gravity are important environmental stimuli to induce changes in plant growth and photosynthesis by P. patens, in which an alteration in chloroplast size is one of the key traits. We are now planning an ISS experiment to investigate the responses of P. patens to microgravity.  相似文献

Certain strains of lactic acid bacteria present in commercial cheese starters, characterized by faint transparent colonies on an agar plate containing 1 mg kg −1 crystal violet (CVT), were identified as Lactococcus lactis subsp. (ssp) lactis biovar diacetylactis. The effect of citrate on the growth of these strains (CVT strains) in the presence of glucose was studied, in comparison with L. lactis strains. Molar growth yield from glucose (YG, g dry weight/mole of glucose consumed) for CVT strains grown on glucose plus citrate was significantly higher than the control (i.e. without citrate), but not for other L. lactis strains tested. Enhanced YG was also observed at a pH-controlled experiment, indicating that enhanced YG did not result from a buffering effect of citrate. CVT strains, in contrast to other strains of the same species, were shown to obtain enough energy to enhance YG on glucose–citrate mixtures.  相似文献

Surface contouring by phase-shifting digital holography is proposed and verified by experiments and numerical simulations. Digital holograms are recorded before and after mode hopping of a laser diode subject to current tuning, and the difference of the reconstructed phases at each wavelength is computed to deliver surface contours of a diffusely reflecting surface. Since normal incidence on the object is employed, the method does not need the removal of the tilt component and is free from the shadowing effect as advantages over the dual-incidence method proposed before by the first author.  相似文献

The water corrosion behavior of a typical fluoroaluminate glass was studied in both plate and powder form. The infrared spectra of the corroded plate sample exhibited an overall transmittance decrease but little absorption at 3450 cm–1. Transmittance in the visible region was substantially reduced with an increase of haziness. Microscopic observation revealed a uniform corrosion of the glass surface without any detectable cracks. Dissolution of the glass powder in water resulted in a slight pH increase of the solution. The leaching was congruent and the dynamic dissolution process monitored by solution conductivity followed a logarithmic correlation with time. The corrosion behavior was greatly different from that of fluoride glasses in other systems.  相似文献

We have optimized the conditions for visualizing microfilaments, microtubules, and coated pits in the cortical cytoplasm of high-pressure frozen and freeze-substituted plant cells, in both tobacco root tips and onion cotyledons, individual microfilaments and the supramolecular structure of coated pits can be seen clearly in freeze-substituted samples treated with OsO4 at 40 degrees C followed by 5% uranyl acetate. Treatment with uranyl acetate alone resulted in poorly stained cytoplasmic organelles, whereas microfilaments were difficult to discern in specimen treated with OsO4 alone. The combination of a 40 degrees C OsO4 staining step followed by staining with uranyl acetate at 4 degrees C should prove useful for more detailed plant cytoskeletal/membrane studies in the future.  相似文献

A wavelength-scanning heterodyne interference confocal microscope has proved to provide the tomographic image of the refractive indices of transparent and turbid media on the scale of geometrical depth when weakly reflected light with an optical power as low as of the order of 10(-14) W is used. The refractive indices of the transparent object and the turbid media were measured with accuracies of -0.5% and approximately 3%, respectively. This imaging method is advantageous for evaluating quantitative refractive indices and internal structures.  相似文献

A wavelength-scanning heterodyne interference confocal microscope quickly accomplishes the simultaneous measurement of the thickness and the refractive index of a sample by detection of the amplitude and the phase of the interference signal during a sample scan. However, the measurement range of the optical path difference (OPD) that is obtained from the phase changes is limited by the time response of the phase-locked loop circuit in the FM demodulator. To overcome this limitation and to improve the accuracy of the separation measurement, we propose an OPD detection using digital signal processing with a Hilbert transform. The measurement range is extended approximately five times, and the resolution of the OPD is improved to 5.5 from 9 microm without the electrical noise of the FM demodulator circuit. By applying this method for simultaneous measurement of thickness and the refractive index, we can measure samples 20-30-microm thick with refractive indices between 1 and 1.5.  相似文献

Analysis of nine kinds of sweeteners in foods by LC/MS   总被引:4,自引:0,他引:4  
A simple and rapid method for the simultaneous determination of nine kinds of sweeteners (acesulfame potassium, AK; sucralose, SUC; saccharin, SA; cyclamate, CYC; aspartame, APM; dulcin, DU; glycyrrhizic acid, GA; stevioside, STV; rebaudioside A, REB) in various foods by high-performance liquid chromatography-electrospray mass spectrometry (LC/ESI-MS) was developed. The LC separation was performed on a ZORBAX Eclipse XDB-C18 (2.1 mm x 150 mm) with a mobile phase of 5 mmol/L dibutylammonium acetate (DBAA) and acetonitrile-water (8: 2). Mass spectral acquisition was done in the negative ion mode by applying selected ion monitoring (SIM). The sweeteners were extracted from foods with 0.08 mol/L phosphate buffer (pH 7.0)- ethanol (1:1), and the extract was cleaned up on a Sep-pak Vac C18 cartridge after the addition of tetrabutylammonium bromide and phosphate buffer (pH 3.0). The recovery of the nine kinds of sweeteners from five kinds of foods fortified at the level 0.01 g/kg, 0.05 g/kg and 0.20 g/kg was 75.7-109.2%, and the between-day SD values were 0.5-10.9%. The quantification limits of AK, SA, CYC, APM and STV were 0.001 g/kg, and those of SUC, DU, GA and REB were 0.005 g/kg. A recovery test from each cleaned-up sample solution was necessary to detect ionization suppression.  相似文献

Wide-gap II-VI MgZnCdSe quaternary compounds were grown on InP substrates by molecular beam epitaxy, for the first time. Changing the Mg composition (x = 0 to 0.63), various Mgx(ZnyCd1_y)1_xSe lattice-matched to InP were grown. Mirror-like surface morphologies and streaky reflection high energy electron diffraction patterns of MgZnCdSe were obtained. With increased Mg compositions, the band-edge emissions wavelength in photoluminescence spectra was shifted from 572 nm (2.17 eV) to 398 nm (3.12 eV) at 15K. Furthermore, the absolute PL peak intensity increased drastically with increased band-edge emission, being accompanied by a relative decrement in the deep level emission intensities were also observed.  相似文献
