Pycnodysostosis, a rare autosomal recessive skeletal dysplasia, is caused by a deficiency of cathepsin K. Patients have impaired bone resorption in the presence of normal or increased numbers of multinucleated, but dysfunctional, osteoclasts. Cathepsin K degrades collagen type I and generates N-telopeptide (NTX) and the C-telopeptide (CTX) that can be quantified. Levels of these telopeptides are increased in lactating women and are associated with increased bone resorption. Nothing is known about the consequences of cathepsin K deficiency in lactating women. Here we present for the first time normalized blood and CTX measurements in a patient with pycnodysostosis, exclusively related to the lactation period. In vitro studies using osteoclasts derived from blood monocytes during lactation and after weaning further show consistent bone resorption before and after lactation. Increased expression of cathepsins L and S in osteoclasts derived from the lactating patient suggests that other proteinases could compensate for the lack of cathepsin K during the lactation period of pycnodysostosis patients.  相似文献

METIS-II was an EU-FET MT project running from October 2004 to September 2007, which aimed at translating free text input without resorting to parallel corpora. The idea was to use “basic” linguistic tools and representations and to link them with patterns and statistics from the monolingual target-language corpus. The METIS-II project has four partners, translating from their “home” languages Greek, Dutch, German, and Spanish into English. The paper outlines the basic ideas of the project, their implementation, the resources used, and the results obtained. It also gives examples of how METIS-II has continued beyond its lifetime and the original scope of the project. On the basis of the results and experiences obtained, we believe that the approach is promising and offers the potential for development in various directions.  相似文献

In the present study, the authors tested the effects of working-memory load on math problem solving in 3 different cultures: Flemish-speaking Belgians, English-speaking Canadians, and Chinese-speaking Chinese currently living in Canada. Participants solved complex addition problems (e.g., 58 + 76) in no-load and working-memory load conditions, in which either the central executive or the phonological loop was loaded. The authors used the choice/no-choice method to obtain unbiased measures of strategy selection and strategy efficiency. The Chinese participants were faster than the Belgians, who were faster and more accurate than the Canadians. The Chinese also required fewer working-memory resources than did the Belgians and Canadians. However, the Chinese chose less adaptively from the available strategies than did the Belgians and Canadians. These cultural differences in math problem solving are likely the result of different instructional approaches during elementary school (practice and training in Asian countries vs. exploration and flexibility in non-Asian countries), differences in the number language, and informal cultural norms and standards. The relevance of being adaptive is discussed as well as the implications of the results in regards to the strategy choice and discovery simulation model of strategy selection (J. Shrager & R. S. In this study, we investigated a new instrument: the Southampton Test of Empathy for Preschoolers (STEP). The test incorporated 8 video vignettes of children in emotional scenarios, assessing a child's ability to understand (STEP-UND) and share (STEP-SHA) in the emotional experience of a story protagonist. Each vignette included 4 emotions (angry, happy, fearful, sad) that reflected emotion judgments based on the protagonist's facial expression, situation, verbal cues, and desire. The STEP was administered to 39 preschool children, and internal reliability, concurrent validity, and construct validity were addressed. The results showed good internal consistency. They also highlighted moderate concurrent validity with parent-rated empathy, a measure of facial indices, and construct validity with teacher-rated prosocial behavior. The energy potential for energy crops and biomass residues in the Netherlands is assessed. The analysis explores the possible use of land for biomass production in the future. Various government memorandums and analyses of the expected future land use in various sectors have served as the basis for the assessment of the supply of and the demand for land in the future. In this study the potential supply of agricultural land is based on expected productivity increments in agriculture and assumptions with respect to the future demand for agricultural products. Various future claims for infrastructure, forestry, urban areas and nature are subtracted from the expected supply. The net projected supply of land ranges from zero to 52 000 ha in 2000 to 110 000-250 000 ha in 2015. The supply of agricultural land depends however on a number of supra-national factors, such as the European agricultural policy, world market developments and the agricultural production in the countries in Eastern Europe. Uncertainties remain, therefore, and the projected supply of agricultural land should be considered as a possible scenario based on current trends. If the calculated land potential is used for energy crops like miscanthus and short rotation coppice, this land could contribute 0-10 PJ in 2000 and 27-59 PJ in 2015. Secondary biomass yields, such as those from forestry, agricultural residues, wood from prunings, etc., could contribute a further 34 PJ in 2000, decreasing to approximately 28 PJ in 2015. Taken together these potentials could satisfy 1-1.5% of the energy requirements of the Netherlands in 2000 and 1.5-2.5% in 2015, provided that energy farming is an economically feasible activity for farmers.  相似文献

High hydrostatic pressure (HHP) inactivation of three Listeria monocytogenes strains (EGDe, LO28, and Scott A) subjected to 350 MPa at 20 degrees C in ACES buffer resulted in survival curves with significant tailing for all three strains. A biphasic linear model could be fitted to the inactivation data, indicating the presence of an HHP-sensitive and an HHP-resistant fraction, which both showed inactivation according to first-order kinetics. Inactivation parameters of these subpopulations of the three strains were quantified in detail. EGDe showed the highest D-values for the sensitive and resistant fraction, whereas LO28 and Scott A showed lower HHP resistance for both fractions. Survivors isolated from the tail of LO28 and EGDe were analyzed, and it was revealed that the higher resistance of LO28 was a stable feature for 24% (24 of 102) of the resistant fraction. These HHP-resistant variants were 10 to 600,000 times more resistant than wild type when exposed to 350 MPa at 20 degrees C for 20 min. Contrary to these results, no stable HHP-resistant isolates were found for EGDe (0 of 102). The possible effect of HHP survival capacity of stress-resistant genotypic and phenotypic variants of L. monocytogenes on the safety of HHP-processed foods is discussed.  相似文献

Current microlithography used in high-volume integrated circuit manufacturing employs some form of optical projection technology. The most advanced tools use deep-ultraviolet (DUV) radiation having a wavelength of 248 nm and are used to print 250-nm features. These tools will likely be extended for use at the 180-nm generation and perhaps below. New DUV tools using 193-nm radiation are actively under development and are expected to be used for 130-nm generation and perhaps even 100-nm generation. Extending these DUV optical projection tools for manufacturing in the 100-200-nm region will be paced by the development of new high numerical aperture imaging systems and highly complex phase shift masks. For future generations of integrated circuits with minimum feature sizes below 100 nm, 193-nm tools will have great difficulty meeting all manufacturing requirements. This paper describes an alternate optical approach, for sub-100-nm generations, based on extreme ultraviolet radiation at around 13 nm, called extreme ultraviolet lithography (EUVL). This approach uses a laser-produced plasma source of radiation, a reflective mask, and a 4× reduction all-reflective imaging system. The technology is currently in the engineering development phase for an alpha machine. Determining the heating mechanism (or mechanisms) that causes the outer atmosphere of the Sun, and many other stars, to reach temperatures orders of magnitude higher than their surface temperatures has long been a key problem. For decades, the problem has been known as the coronal heating problem, but it is now clear that 'coronal heating' cannot be treated or explained in isolation and that the heating of the whole solar atmosphere must be studied as a highly coupled system. The magnetic field of the star is known to play a key role, but, despite significant advancements in solar telescopes, computing power and much greater understanding of theoretical mechanisms, the question of which mechanism or mechanisms are the dominant supplier of energy to the chromosphere and corona is still open. Following substantial recent progress, we consider the most likely contenders and discuss the key factors that have made, and still make, determining the actual (coronal) heating mechanism (or mechanisms) so difficult.  相似文献

Herein we describe the synthesis and conformational analysis of a series of bicyclic thymidine derivatives and their evaluation as inhibitors of thymidine monophosphate kinase from Mycobacterium tuberculosis (TMPKmt), based on previously discovered bicyclic sugar nucleosides. With a K(i) value of 2.3 microm, 1-[3-aminomethyl-3,5-dideoxy-2-O,6-N-(thiocarbonyl)-beta-D-ribofuranosyl]thymine emerged as the most potent TMPK inhibitor of this series. Moreover, this promising compound displays inhibitory potency against Mycobacteria cultures with an IC(99) value of 100 microg mL(-1), thus promoting TMPKmt for the first time as a validated target for further inhibitory design. Attempts to rationalise the observed structure-activity relationship (SAR) involving molecular modelling and conformational analysis are described.  相似文献

YoungCinchona ledgeriana plants contain two types of alkaloid: indole alkaloids in the leaves and quinoline alkaloids in the root. FromCinchona leaves, a crude alkaloid extract was made, containing the cinchophylline type of indole alkaloids and a small amount of 5-methoxytryptamine. The leaf alkaloid extract exerted a strong detrimental effect on the growth of larvae of the polyphagous beet armyworm,Spodoptera exigua (Lepidoptera). Feeding of larvae on an artificial diet containing the leaf alkaloids at the same concentrations as those found in the plant resulted in significant growth reduction, retardation in development, and mortality of the larvae. Cinchophyllines are composed of 5-methoxytryptamine coupled to a corynantheal unit. When incorporated into the artificial diet, 5-methoxytryptamine alone had no effect on the 5.exigua larvae. Corynantheal, however, had a strong detrimental effect on growth of the larvae, its effect being comparable to that of the leaf alkaloid extract. In contrast to the indole-type leaf alkaloids, the quinolinetypeCinchona root alkaloids did not affect growth and development of the larvae. These results suggest that the indole-type alkaloids, which inCinchona plants are present at the highest concentrations in the young, vulnerable leaflets, are involved in the chemical defense of the plant against herbivorous insects.  相似文献
