BACKGROUND: The polymerase chain reaction (PCR) is a powerful tool that is being increasingly used for detection of transgenic DNA. PCR requires only a minute quantity of template, but sensitive and accurate testing requires DNA of sufficient purity and free from inhibitors such as plant polysaccharides. Several standard protocols are available for this purpose, but they usually involve several steps, imply destruction of the maize kernel, or are time‐consuming. Our aim was to develop a fast and simple extraction method to isolate a raw DNA‐containing solution from maize tissues suitable for use as a template in a PCR‐based detection assay with specific oligonucleotides directed to the identification of event MON810. RESULTS: The NaOH‐based DNA extraction method we report here is time‐saving (5 min) and can be used to isolate DNA‐containing solutions from a small maize leaf portion (down to 1 mg) or from a single overnight‐germinated kernel. PCR performed with selected primers yielded reproducible detection of transgenic DNA. CONCLUSION: The main advantages of the procedure are the quick extraction step, the possibility of non‐destructive testing of maize kernels, and the robustness of the PCR‐based detection, a consequence of the selection of MON810‐matching oligonucleotides yielding intense and highly specific amplicons. Various aspects of global environmental change affect plant photosynthesis, the primary carbon input in ecosystems. Thus, accurate methods of measuring plant photosynthesis are important. Remotely sensed spectral indices can monitor in detail the green biomass of ecosystems, which provides a measure of potential photosynthetic capacity. In evergreen vegetation types, however, such as Mediterranean forests, the amount of green biomass changes little during the growing season and, therefore, changes in green biomass are not responsible for changes in photosynthetic rates in those forests. This study examined the net photosynthetic rates and the diametric increment of stems in a Mediterranean forest dominated by Quercus ilex using three spectral indices (normalized difference vegetation index (NDVI), enhanced vegetation index (EVI), and photochemical reflectance index (PRI)) derived from Moderate Resolution Imaging Spectroradiometer (MODIS) sensors. Average annual EVI accounted for 83% of the variability of the diametric increment of Q. ilex stems over a 10 year period. NDVI was marginally correlated with the diametric increment of stems. This study was the first to identify a significant correlation between net photosynthetic rates and radiation use efficiency at the leaf level using PRI derived from satellite data analysed at the ecosystem level. These results suggest that each spectral index provided different and complementary information about ecosystem carbon uptake in a Mediterranean Q. ilex forest.  相似文献

New flexible aliphatic oxazolidone‐isocyanurate networks (AISOX) are obtained by reacting a low molecular weight diisocyanate (4,4′‐methylene dicyclohexyl diisocyanate, H12MDI) and a macro‐diepoxyde (poly(ethylene glycol) diglycidyl ether, Mn = 526, PEGDGE) in different molar ratio. The curing reaction, carried out from 25 °C to 200 °C, is studied by using DSC and FTIR. The effect of the molar ratio of the two monomers on thermal and mechanical properties of AISOX resins is investigated by DSC, thermogravimetric analysis, stress?strain measurements and optical microscopy. Independently from the feed composition, it is observed that the reaction steps are: (i) partial hydrolysis of isocyanate caused by water traces, (ii) incomplete trimerization of isocyanate to give isocyanurate, and (iii) formation of oxazolidone and complete conversion of isocyanate. At the highest concentration of the soft macrodiepoxyde (PEGDGE), the AISOX resin is in the rubbery state at room temperature and shows an elastomeric behavior. © 2016 Wiley Periodicals, Inc. J. This study combines the use of corn starch and Tetradesmus obliquus microalgae for the production of antioxidant starch films as flexible packaging material. Starch was plasticized with glycerol and blended with 1 w% polyallylamine chosen as an agent to modify the film physical properties. The addition of polyallylamine improved film water stability and water vapor transmission rate as well as mechanical stiffness and tenacity. The dried Tetradesmus obliquus microalgae, which showed an EC50 value of 2.8 mg/mg DPPH (2.2-Diphenyl-1-picrylhydrazyl radical), was then used as antioxidant filler. The addition of microalgae provided the films with good antioxidant activity, which increased with microalgae content increasing. To our knowledge, this is the first study reporting the development of sustainable bioactive packaging films composed of almost 100% starch, and follows the European union’s goals on plastics strategy concerning the promotion of bio-based, compostable plastics and the setting up of approaches to prevent food waste with a simple plastic packaging.  相似文献

Oxygen high pressure (up to 16 MPa) has been introduced in the oxygen-annealing step necessary to make the YBa2Cu3Ox phase superconducting (change x from 6 to about 7). It enables a displacement in the equilibrium phase diagram towards higher temperatures, which means the possibility to achieve the same final oxygen content as the one at low temperature, but with the benefit of higher diffusion rates. Initial development made on thin bars (1.5–3 mm thick) has confirmed the interest of using a high pressure of oxygen. TEM observations have shown an increase of twin density associated with higher Jc. This is in agreement with other works claiming the possibility of higher Jc by twin engineering, and more precisely by twin refinement while annealing at high temperature. We report the successful application of this process without any adjustment to so-called thin-wall single-domain samples. These samples are obtained by growing a crystal on a pellet already shaped with an array of holes. The advantage is that, as far as diffusion processes are concerned, the typical length is not anymore the diameter of the sample, but the thickness of the walls between holes. The trapped field of 16 mm diameter Y123 thin-wall single-domain samples was doubled (0.6 T vs. 0.3 T at 77 K) in a rather short annealing time (about 3 days). Microstructures as well as magneto-optical observations of plain and thin-wall samples evidenced a reduction of cracks in the thin-wall samples. Improved performances were confirmed by further characterizations performed from 77 down to 20 K using the pulse-field facilities of the LNCMP at Toulouse.  相似文献

A gold electrode surface was modified using a dinuclear copper complex [CuII2 (Ldtb)(μ-OCH3)](BPh4) and then coated with a chitosan film. This biomimetic polymer film-coated electrode was employed to eliminate the interference from ascorbic acid and uric acid in the sensitive and selective determination of dopamine. The optimized conditions obtained for the biomimetic electrode were 0.1 M phosphate buffer solution (pH 8.0), complex concentration of 2.0 × 10−4 M, 0.1% of chitosan and 0.25% of glyoxal. Under the optimum conditions, the calibration curve was linear in the concentration range of 4.99 × 10−7 to 1.92 × 10−5 M, and detection and quantification limits were 3.57 × 10−7 M and 1.07 × 10−6 M, respectively. The recovery study gave values of 95.2-102.6%. The lifetime of this biomimetic sensor showed apparent loss of activity after 70 determinations. The results obtained with the modified electrode for dopamine quantification in the injection solution matrix were in good agreement with those of the pharmacopoeia method.  相似文献

The heart of palm is a food extracted from several species of palm trees. Only 10–20% of the raw material is transformed into canned hearts of palm, therefore this agro‐industry generates a large quantity of solid residue. The aim of this work was to extract and characterise a juice produced from this solid residue. This extract is compared with the traditional fruit juices (orange and pineapple juices), obtaining higher contents of minerals, such as magnesium (22.80 mg/100 g) and potassium (276.90 mg/100 g), phenolic compounds (gallic, 3,4‐dihydroxybenzoic, chlorogenic, syringic and caffeic acids) and high free radical scavenging capacity (expressed as 2,2‐diphenyl‐1‐picrylhydrazyl), which suggest high antioxidant activity. Additionally, a new soft drink was formulated using this king palm juice as its main ingredient. The soft drink samples formulated with this juice had high sensory acceptability and high intention of consumption, which suggests an important and low‐cost application for this industrial residue.  相似文献

A trial was carried out on seven Massese ewes, from which individual milk samples were taken during lactation. All the enzyme activities investigated (oxidase activity of xanthine oxidoreductase, gamma-glutamyltranspeptidase, alkaline phosphatase and 5′-nucleotidase) were affected by the lactation phase, with the exception of the dehydrogenase activity of xanthine oxidoreductase (XOR). A higher oxidase activity (XO) of XOR was found when the diameter of milk fat globules was smaller. In addition, the absence of XO in colostrum led to the hypothesis that its antibacterial role is independent of its activity. The increase in alkaline phosphatase activity during lactation requires further research to better define the criteria for pasteurised sheep milk.  相似文献
