Microbiological contamination of chicken meat depends on the conditions under which the animals are reared, slaughtered and processed. The aim of this study was to determine the influence of farm origin and processing stages at slaughterhouse on the microbial safety and quality of chicken. Samples of chicken carcasses from three different farms were taken from a slaughterhouse. Mesophiles, Escherichia coli, coagulase positive Staphylococcci counts, presence of Listeria monocytogenes,Campylobacter and Salmonella were determined at five sampling points: after defeathering, after evisceration, after washing, after chilling and after cutting. Chilling reduced log numbers of mesophiles, coagulase positive Staphylococci and E. coli by 0.85, 1.52 and 2.2 log units, respectively. Salmonella was not detected after chilling. High prevalence of Campylobacter spp was observed at all the stages ranging between 84% and 100%. L. monocytogenes was not detected in chicken carcasses after defeathering. However, it was detected after evisceration and after washing and chilling. The most critical stage for Lmonocytogenes contamination was the portioning operation, the prevalence in breast and legs being 88% and 84%, respectively.  相似文献

The decellularized extracellular matrix (ECM) obtained from human and porcine adipose tissue (AT) is currently used to prepare regenerative medicine bio-scaffolds. However, the influence of these natural biomaterials on host immune response is not yet deeply understood. Since macrophages play a key role in the inflammation/healing processes due to their high functional plasticity between M1 and M2 phenotypes, the evaluation of their response to decellularized ECM is mandatory. It is also necessary to analyze the immunocompetence of macrophages after contact with decellularized ECM materials to assess their functional role in a possible infection scenario. In this work, we studied the effect of four decellularized adipose matrices (DAMs) obtained from human and porcine AT by enzymatic or chemical methods on macrophage phenotypes and fungal phagocytosis. First, a thorough biochemical characterization of these biomaterials by quantification of remnant DNA, lipids, and proteins was performed, thus indicating the efficiency and reliability of both methods. The proteomic analysis evidenced that some proteins are differentially preserved depending on both the AT origin and the decellularization method employed. After exposure to the four DAMs, specific markers of M1 proinflammatory and M2 anti-inflammatory macrophages were analyzed. Porcine DAMs favor the M2 phenotype, independently of the decellularization method employed. Finally, a sensitive fungal phagocytosis assay allowed us to relate the macrophage phagocytosis capability with specific proteins differentially preserved in certain DAMs. The results obtained in this study highlight the close relationship between the ECM biochemical composition and the macrophage’s functional role.  相似文献

Osteogenesis imperfecta is a rare genetic disorder characterized by bone fragility, due to alterations in the type I collagen molecule. It is a very heterogeneous disease, both genetically and phenotypically, with a high variability of clinical phenotypes, ranging from mild to severe forms, the most extreme cases being perinatal lethal. There is no curative treatment for OI, and so great efforts are being made in order to develop effective therapies. In these attempts, the in vivo preclinical studies are of paramount importance; therefore, serious analysis is required to choose the right murine OI model able to emulate as closely as possible the disease of the target OI population. In this review, we summarize the features of OI murine models that have been used for preclinical studies until today, together with recently developed new murine models. The bone parameters that are usually evaluated in order to determine the relevance of new developing therapies are exposed, and finally, current and innovative therapeutic strategies attempts considered in murine OI models, along with their mechanism of action, are reviewed. This review aims to summarize the in vivo studies developed in murine models available in the field of OI to date, in order to help the scientific community choose the most accurate OI murine model when developing new therapeutic strategies capable of improving the quality of life.  相似文献

Fumarylacetoacetate hydrolase (FAH) is the fifth enzyme in the tyrosine catabolism pathway. A deficiency in human FAH leads to hereditary tyrosinemia type I (HT1), an autosomal recessive disorder that results in the accumulation of toxic metabolites such as succinylacetone, maleylacetoacetate, and fumarylacetoacetate in the liver and kidney, among other tissues. The disease is severe and, when untreated, it can lead to death. A low tyrosine diet combined with the herbicidal nitisinone constitutes the only available therapy, but this treatment is not devoid of secondary effects and long-term complications. In this study, we targeted FAH for the first-time to discover new chemical modulators that act as pharmacological chaperones, directly associating with this enzyme. After screening several thousand compounds and subsequent chemical redesign, we found a set of reversible inhibitors that associate with FAH close to the active site and stabilize the (active) dimeric species, as demonstrated by NMR spectroscopy. Importantly, the inhibitors are also able to partially restore the normal phenotype in a newly developed cellular model of HT1.  相似文献

The freeze-drying method is proposed as an effective synthesis process for the obtaining of LiFePO4/C composites. The citric acid is used as a complexing agent and carbon source. After the low temperature annealing, the freeze-dried solution leads to a homogeneous carbon covered LiFePO4 sample. The chemical characterization of the material included ICP and elemental analysis, infrared spectroscopy, X-ray diffraction, magnetic measurements and thermal analysis. SEM and TEM microscopies indicate an aggregate morphology with tiny particles of lithium iron phosphate inside a carbon matrix. Impedance spectroscopy showed a 8.0 × 10−7 S cm−1 conductivity value. Cyclic voltammetry graphics displayed the two peaks corresponding to the Fe(II)/Fe(III) reaction and demonstrated the good reversibility of the material. The specific capacity value obtained at C/40 rate was 164 mAh g−1, with a slight decrease on greater C-rates reaching 146 mAh g−1 at C/1. The capacity retention study has evidenced good properties, with retention over 97% of the maximum values in the first 50 cycles, which allows an effective performance of the freeze-dried sample as cathodic material in lithium-ion batteries.  相似文献

The incidence of bone-related disorders is continuously growing as the aging of the population in developing countries continues to increase. Although therapeutic interventions for bone regeneration exist, their effectiveness is questioned, especially under certain circumstances, such as critical size defects. This gap of curative options has led to the search for new and more effective therapeutic approaches for bone regeneration; among them, the possibility of using extracellular vesicles (EVs) is gaining ground. EVs are secreted, biocompatible, nano-sized vesicles that play a pivotal role as messengers between donor and target cells, mediated by their specific cargo. Evidence shows that bone-relevant cells secrete osteoanabolic EVs, whose functionality can be further improved by several strategies. This, together with the low immunogenicity of EVs and their storage advantages, make them attractive candidates for clinical prospects in bone regeneration. However, before EVs reach clinical translation, a number of concerns should be addressed. Unraveling the EVs’ mode of action in bone regeneration is one of them; the molecular mediators driving their osteoanabolic effects in acceptor cells are now beginning to be uncovered. Increasing the functional and bone targeting abilities of EVs are also matters of intense research. Here, we summarize the cell sources offering osteoanabolic EVs, and the current knowledge about the molecular cargos that mediate bone regeneration. Moreover, we discuss strategies under development to improve the osteoanabolic and bone-targeting potential of EVs.  相似文献

New applications and emerging markets in electromobility and large‐scale stationary energy storage require the development of new electrochemical systems with higher energy density than current batteries. Rechargeable metal–air batteries, mainly lithium–air and zinc–air systems, are considered one of the most promising candidates. In contrast to lithium, zinc is abundant, inexpensive and its electrodeposition in aqueous electrolytes is relatively easy. Unfortunately, achieving a rechargeable zinc–air battery is still hindered by various technical problems related to the reversibility and lifetime of the electrodes. The most widely used electrolyte in zinc–air batteries has been the classical aqueous alkaline. In this context and with the main objective of providing a complete overview, we studied a wide number of articles starting from the beginning of the development of secondary zinc–air batteries (1970–1980s) to more recent works, with the aim of compiling all available information. It is essential to revise older papers to find relevant information that may get otherwise forgotten and not taken into account to develop new solutions. This information could also be applied in other storage systems based on zinc as nickel–zinc, zinc hybrid or zinc‐ion. Copyright © 2016 John Wiley & Sons, Ltd.  相似文献

This paper presents an increase to x = 0.67 of the zirconium content in the conductive Bi2−xZrxO3+δ solid solution. Complete incorporation of Zr in the βIII-Bi2O3 structure, confirmed by X-ray powder diffraction, has produced a phase with a lower volume and superior conductivity than those predicted by an earlier study. The observed βIII-δ Bi2−xZrxO3+δ phase transition around 730 °C has been characterised for the first time and shows a segregation of a mixture of predominantly γ-Bi2O3 and approximately 30% of the ZrO2, before total reincorporation of the Zr in the high temperature δ-phase.  相似文献
