This study examined the longitudinal effects of 2 first-grade universal preventive interventions on academic outcomes (e.g., achievement, special education service use, graduation, postsecondary education) through age 19 in a sample of 678 urban, primarily African American children. The classroom-centered intervention combined the Good Behavior Game (H. H. Barrish, Saunders, & Wolfe, 1969) with an enhanced academic curriculum, whereas a second intervention, the Family–School Partnership, focused on promoting parental involvement in educational activities and bolstering parents’ behavior management strategies. Both programs aimed to address the proximal targets of aggressive behavior and poor academic achievement. Although the effects varied by gender, the classroom-centered intervention was associated with higher scores on standardized achievement tests, greater odds of high school graduation and college attendance, and reduced odds of special education service use. The intervention effects of the Family–School Partnership were in the expected direction; however, only 1 effect reached statistical significance. The findings of this randomized controlled trial illustrate the long-term educational impact of preventive interventions in early elementary school. The transmission eigenvalue problem arises in scattering theory. The main difficulty in its analysis is the fact that, depending on the chosen formulation, it leads either to a quadratic eigenvalue problem or to a non-classical mixed problem. In this paper we prove the convergence of a mixed finite element approximation. This approach, which is close to the Ciarlet–Raviart discretization of biharmonic problems, is based on Lagrange finite elements and is one of the less expensive methods in terms of the amount of degrees of freedom. The convergence analysis is based on classical abstract spectral approximation result and the theory of mixed finite element methods for solving the stream function–vorticity formulation of the Stokes problem. Numerical experiments are reported in order to assess the efficiency of the method.  相似文献

Fungal β‐1,3‐glucan glucanosyltransferases are glucan‐remodeling enzymes that play important roles in cell wall integrity, and are essential for the viability of pathogenic fungi and yeasts. As such, they are considered possible drug targets, although inhibitors of this class of enzymes have not yet been reported. Herein we report a multidisciplinary approach based on a structure‐guided design using a highly conserved transglycosylase from Sacharomyces cerevisiae, that leads to carbohydrate derivatives with high affinity for Aspergillus fumigatus Gel4. We demonstrate by X‐ray crystallography that the compounds bind in the active site of Gas2/Gel4 and interact with the catalytic machinery. The topological analysis of noncovalent interactions demonstrates that the combination of a triazole with positively charged aromatic moieties are important for optimal interactions with Gas2/Gel4 through unusual pyridinium cation–π and face‐to‐face π–π interactions. The lead compound is capable of inhibiting AfGel4 with an IC50 value of 42 μm .  相似文献

We report on the synthesis of thin, transparent, and highly catalytic carbon nanotube films. Nanotubes catalyze the reduction of triiodide, a reaction that is important for the dye-sensitized solar cell, with a charge-transfer resistance as measured by electrochemical impedance spectroscopy that decreases with increasing film thickness. Moreover, the catalytic activity can be significantly enhanced by exposing the nanotubes to ozone in order to introduce defects. Ozone-treated, defective nanotube films could serve as catalytic, transparent, and conducting electrodes for the dye-sensitized solar cell. Other possible applications include batteries, fuel cells, and electroanalytical devices.  相似文献

Carotid plaques can be characterised by ultrasound by size and echogenicity. Both size and echogenicity are predictors of cardiovascular events. The aim of this study was to examine whether traditional risk factors and markers of inflammation and oxidation were associated with plaque size and echogenicity. Computerised analysis of carotid plaque size and echogenicity (grey scale median, GSM) were performed by ultrasound in a population-based health survey in 1,016 subjects aged 70 years (PIVUS study). Information on cardiovascular risk factors was collected, together with markers of inflammation and oxidation. Increased Framingham risk score, systolic blood pressure, higher BMI and decreased HDL, lower glutathione levels were related to echolucent plaques. Previous or present smoking was common with significantly more pack-years related to the echorich plaques. Plaque size was associated with increased Framingham risk score, systolic blood pressure, blood glucose levels, smoking, ApoB/A1 ratio, OxLDL, TNF alpha, HOMA insulin resistance, leucocyte count, decreased BCD-LDL and low levels of l-selectin. Low HDL, increased BMI and decreased glutathione levels were associated with the echolucency of carotid plaques, implying metabolic factors to play a role for plaque composition. Markers of inflammation were related to plaque size alone, implying inflammation to be predominantly associated with the amount of atherosclerosis. These results suggest that plaque size and echogenicity are influenced by different risk factors.  相似文献

Whole-cell biosensors based on reporter genes are finding a variety of applications in analytical chemistry. Despite their ability to selectively recognize the analyte in a complex mixture, few applications of such sensing devices to real sample analysis are reported. This is mainly due to nonspecific effects on the biosensor response caused by components of the sample matrix and by environmental changes. To overcome this problem, a bacterial biosensor with an internal correction mechanism of the analytical response was developed by introducing an additional reporter gene that provides a reference signal of the analytical performance of the biosensor. The first reporter (GFPuv), expressed in response to the concentration of L-arabinose, provides the analytical signal; the second reporter (EYFP), constitutively expressed if a constant amount of IPTG is added to each sample, was used as an internal reference. By inducing the biosensor with varying amounts of L-arabinose and a constant amount of IPTG, it was possible to obtain a dose-response curve for L-arabinose, together with a constant production of EYFP, which allowed for a dynamic evaluation of the metabolic activity of the cell. When tested in nonoptimal conditions (e.g., in the presence of either ethanol or deoxycholic acid at toxic concentrations), the presence of the internal reference system corrected the analytical response due to nonspecific interferences.  相似文献

Testing for arsenic pollution is commonly performed with chemical test kits of unsatisfying accuracy. Bacterial biosensors are an interesting alternative as they are easily produced, simple, and highly accurate devices. Here, we describe the development of a set of bacterial biosensors based on a nonpathogenic laboratory strain of Escherichia coli, the natural resistance mechanism of E. coli against arsenite and arsenate, and three reporter proteins: bacterial luciferase, beta-galactosidase and Green Fluorescent Protein (GFP). The biosensors were genetically optimized to reduce background expression in the absence of arsenic. In calibration experiments with the biosensors and arsenite-amended potable water, arsenite concentrations at 4 microg of As/L (0.05 microM) were routinely and accurately measured. The currently most quantitative system expressed the bacterial luciferase as reporter protein, responding proportional with a concentration range between 8 and 80 microg of As/L. Sensor cells could be stored as frozen batches, resuspended in plain media, and exposed to the aqueous test sample, and light emission was measured after 30-min incubation. Field testing for arsenite was achieved with a system that contained beta-galactosidase, producing a visible blue color at arsenite concentrations above 8 microg/L. For this sensor, a protocol was developed in which the sensor cells were dried on a paper strip and placed in the aqueous test solution for 30 min after which time color development was allowed to take place. The GFP sensor showed good potential for continuous rather than end point measurements. In all cases, growth of the biosensors and production of the strip test was achieved by very simple means with common growth media, and quality control of the sensors was performed by isolating the respective plasmids with the genetic constructs according to simple standard genetic technologies. Therefore, the biosensor cells and protocols may offer a realistic alternative for measuring arsenic contamination in potable water.  相似文献

ABSTRACT

The paper reports a study of the responses of fishing communities, Regência and Povoação, located at the mouth of the Doce River, to the collapse of the Fundão dam, owned by Samarco Mineração, in 2015. After the disaster, the fishermen’s social and cultural practices have undergone drastic changes. The arrival of tailings in the river has impaired their routines and income. The fishermen and their families indicate a sense of loss of identity and insecurity because of the uncertainty. They do not know if they will be able to resume fishing in these waters. This feeling contrasts with the results of a study conducted before the disaster.  相似文献

Flavour changes of six Belgian pale lager beers were studied in order to estimate the importance of different parameters and reactions in relation to the ageing process. An attempt was made to link analytical data with sensory evaluation using multivariate statistical analysis. Partial least squares regression techniques (PLSR) were employed on the analytical and sensory data. As apparent from the PLSR model, significant indicators of lager beer ageing are aldehyde markers (especially total aldehydes, furfural, hexanal, 2‐methylpropanal, 2‐methylbutanal, and 3‐methylbutanal), cold and permanent haze, and beer colour. Conversely, compounds or parameters that load negatively in the PLSR model for beer ageing are trans‐isohumulones, cis‐isohumulones, total bitterness, the T/C‐ratio, polyphenolic markers (especially proanthocyanidins), the flavanoid content, and, to a lesser extent, the TB‐index and reducing power (TRAP). The integrated analytical‐sensorial methodology is proposed as a useful tool for evaluation of the flavour instability of pale lager beers.  相似文献

Despite modern surgical trauma care, bleeding contributes to one-third of trauma-related death. A significant improvement was obtained through the introduction of tranexamic acid (TXA), which today is widely used in emergency and elective orthopedic surgery to control bleeding. However, concerns remain regarding potential adverse effects on bone turnover and regeneration. Therefore, we employed standardized cell culture systems including primary osteoblasts, osteoclasts, and macrophages to evaluate potential effects of TXA on murine bone cells. While osteoblasts derived from calvarial digestion were not affected, TXA increased cell proliferation and matrix mineralization in bone marrow-derived osteoblasts. Short-term TXA treatment (6 h) failed to alter the expression of osteoblast markers; however, long-term TXA stimulation (10 days) was associated with the increased expression of genes involved in osteoblast differentiation and extracellular matrix synthesis. Similarly, whereas short-term TXA treatment did not affect gene expression in terminally differentiated osteoclasts, long-term TXA stimulation resulted in the potent inhibition of osteoclastogenesis. Finally, in bone marrow-derived macrophages activated with LPS, simultaneous TXA treatment led to a reduced expression of inflammatory cytokines and chemokines. Collectively, our study demonstrates a differential action of TXA on bone cells including osteoanabolic, anti-resorptive, and anti-inflammatory effects in vitro which suggests novel treatment applications.  相似文献
