Dps (DNA-binding protein from starved cells) is well known for the structural protection of bacterial DNA by the formation of highly ordered intracellular assemblies under stress conditions. Moreover, this ferritin-like protein can perform fast oxidation of ferrous ions and subsequently accumulate clusters of ferric ions in its nanocages, thus providing the bacterium with physical and chemical protection. Here, cryo-electron microscopy was used to study the accumulation of iron ions in the nanocage of a Dps protein from Escherichia coli. We demonstrate that Fe2+ concentration in the solution and incubation time have an insignificant effect on the volume and the morphology of iron minerals formed in Dps nanocages. However, an increase in the Fe2+ level leads to an increase in the proportion of larger clusters and the clusters themselves are composed of discrete ~1–1.5 nm subunits.  相似文献

In the last decades, mesh-free methods for simulating various cutting processes have been used very widely as they can eliminate numerical problems in the simulation of material failure and large plastic deformations. This paper deals with the results from modelling the orthogonal cutting of AISI 1045 steel using smoothed particle hydrodynamics (SPH) method. Moreover, it is determined how the parameters of the SPH solver such as initial smoothing length, initial particle density and coefficient for the timestep increase affect the prediction error for the values of cutting force and chip compression ratio as well as computing time. The optimum values of the SPH solver parameters are determined by minimising an objective function. The best balance between the prediction error of machining variables and computing time is achieved for an initial particle density of 40 μm and a coefficient for the timestep increase of 0.4.  相似文献

Proton‐conducting gel electrolytes based on poly(methyl methacrylate) (PMMA), poly(vinylidene fluoride) (PVdF), and mixtures of PMMA with PVdF or poly(vinyl chloride) doped by acid solutions in aprotic solvents were synthesized and are discussed in this article. The gel conductivity as a function of the concentrations of acid and polymer and the polymeric matrix composition has been analyzed. Extreme dependence of the conductivity on acid and polymer concentrations was found. It was revealed that within the acid concentration range studied, the gel conductivity was higher than the conductivity of the corresponding liquid electrolytes used for the synthesis. The increase in the electrical conductivity with the growth of the systems viscosity is discussed as an indication of a certain involvement of the polymer matrix in the increase of the charge carrier mobility within the frame of a Grotthuss mechanism. © 2014 Wiley Periodicals, Inc. Recently, we reported on the photovoltaic current observed in poled capacitors with polycrystalline Pb(ZrTi)O(3) (PZT) films, where (111)-oriented PZT grains are separated by an ultrathin semiconductor PbO phase. This photocurrent is driven by the depolarization field, which is generated by residual uncompensated polarization charge located on grain boundaries near electrodes. We showed that the photocurrent can serve as a criterion of existence of the depolarization field and demonstrated that this field is retained in the film for at least one year. Here, we present new experimental and numerical results which confirm the proposed conception of the photovoltaic effect. We study the photocurrent depending on the kind of electrodes, preliminary illumination in an open-circuit regime, and light intensity of LED, and give evidence of retention of the depolarization field in the films for at least for one and one-half years. The numerical study of the photovoltaic effect at extremely high photogeneration rate shows that total compensation of the polarization charge by photoexcited carriers in these structures is impossible. This photovoltaic effect can be used for nondestructive readout in ferroelectric memory.  相似文献

Controlling the release rate of biocides (antifouling agents) from a paint coating is a key issue for the development of multi-season antifouling marine coatings. One promising approach is the use of nanoparticles onto which biocides are adsorbed to prevent premature depletion of the biocide. Adsorption of one novel (Medetomidine) and six commercially available and widely used antifouling biocides (Chlorothalonile, Dichlofluanid, Diuron, Irgarol, Seanine, Tolylfluanid) onto oxide nanoparticles (Al2O3, CuO, MgO, SiO2, TiO2, ZnO) was investigated by HPLC and NMR in different organic solvents. Large differences in adsorption strength depending on the type of nanoparticle and solvent employed were observed. It was shown that nanoparticles coordinate preferentially with the imidazole moiety of Medetomidine. Independent of the type of particle this interaction was considerably stronger in comparison to the other biocides. However, the interaction strength was strongly dependant on the type of solvent, where the largest strongest interaction was achieved in o-xylene. In addition field tests were performed where a considerable decrease in release rate was displayed from coatings containing Medetomidine adsorbed to nanoparticles compared to coatings containing Medetomidine as single additive.  相似文献

High-quality genome sequences help to elucidate the genetic basis of numerous biological processes and track species evolution. For flax (Linum usitatissimum L.)—a multifunctional crop, high-quality assemblies from Oxford Nanopore Technologies (ONT) data were unavailable, largely due to the difficulty of isolating pure high-molecular-weight DNA. This article proposes a scheme for gaining a contiguous L. usitatissimum assembly using Nanopore data. We developed a protocol for flax nuclei isolation with subsequent DNA extraction, which allows obtaining about 5 μg of pure high-molecular-weight DNA from 0.5 g of leaves. Such an amount of material can be collected even from a single plant and yields more than 30 Gb of ONT data in two MinION runs. We performed a comparative analysis of different genome assemblers and polishers on the gained data and obtained the final 447.1-Mb assembly of L. usitatissimum line 3896 genome using the Canu—Racon (two iterations)—Medaka combination. The genome comprised 1695 contigs and had an N50 of 6.2 Mb and a completeness of 93.8% of BUSCOs from eudicots_odb10. Our study highlights the impact of the chosen genome construction strategy on the resulting assembly parameters and its eligibility for future genomic studies.  相似文献

Stemness and epithelial–mesenchymal plasticity are widely studied in the circulating tumor cells of breast cancer patients because the roles of both processes in tumor progression are well established. An important property that should be taken into account is the ability of CTCs to disseminate, particularly the viability and apoptotic states of circulating tumor cells (CTCs). Recent data demonstrate that apoptosis reversal promotes the formation of stem-like tumor cells with pronounced potential for dissemination. Our study focused on the association between different apoptotic states of CTCs with short- and long-term treatment outcomes. We evaluated the association of viable CTCs, CTCs with early features of apoptosis, and end-stage apoptosis/necrosis CTCs with clinicopathological parameters of breast cancer patients. We found that the proportion of circulating tumor cells with features of early apoptosis is a perspective prognosticator of metastasis-free survival, which also correlates with the neoadjuvant chemotherapy response in breast cancer patients. Moreover, we establish that apoptotic CTCs are associated with the poor response to neoadjuvant chemotherapy, and metastasis-free survival expressed at least two stemness markers, CD44 and CD133.  相似文献

Sputtered metal gratings have been realized using lift-off process based on bilayer resist electron beam lithography (EBL). The lithography mask is composed of PMMA (poly(methylméthacrylate)) layer deposited under HSQ (hydrogen silsesquioxane) inorganic resist. EBL is performed in HSQ, whereas PMMA is used to ease final lift-off. We demonstrate the possibility of patterning by lift-off metals with different sputtering yields and deposition conditions. Gratings with period of 200 nm and filling factor of 50% are obtained.  相似文献

Latex-based coatings for protection of tree seedlings against pest insect feeding are evaluated with respect to surface-, mechanical-, and release properties and antifeedant activity. The latex dispersion Eudragit copolymer (EC) was used to form the coatings, 2,6-di-tert-butyl-4-methylphenol (BHT) and cis-dihydropinidine (Alk) as antifeedants, and a thickener and a alkylglucoside based nonionic surfactant were used as additives to optimize the release- and mechanical properties of coatings. Coating characterization was performed with respect to surface morphology (atomic force microscopy, AFM) and surface wetting (contact angle), as well as to mechanical (tensile stress- and tensile strain at break) properties. Surface smoothness and wettability as well as elasticity increased with addition of the surfactant. The optimized coatings were found to be elastic and water resistant at 3–6 wt.% of BHT and 3 wt.% of surfactant. BHT was released into SDS/water at very low rates. Several formulations of BHT and Alk were efficient in preventing the feeding on conifer bark by a pine insect, Hylobius abietis both in laboratory (no-choice) and in field (3 months) tests.  相似文献
