Most of the kinetic studies on nitrification have been performed in diluted salts medium. In this work, the ammonia oxidation rate (AOR) was determined by respirometry at different ammonia (0.01 and 33.5 mg N‐NH3 L?1), nitrite (0–450 mg N‐NO2? L?1) and nitrate (0 and 275 mg N‐NO3? L?1) concentrations in a saline medium at 30 °C and pH 7.5. Sodium azide was used to uncouple the ammonia and nitrite oxidation, so as to measure independently the AOR. It was determined that ammonia causes substrate inhibition and that nitrite and nitrate exhibit product inhibition upon the AOR. The effects of ammonia, nitrite and nitrate were represented by the Andrews equation (maximal ammonia oxidation rate, rAOMAX, = 43.2 [mg N‐NH3 (g VSSAO h)?1]; half saturation constant, KSAO, = 0.11 mg N‐NH3 L?1; inhibition constant KIAO, = 7.65 mg N‐NH3 L?1), by the non‐competitive inhibition model (inhibition constant, KINI, = 176 mg N‐NO2? L?1) and by the partially competitive inhibition model (inhibition constant, KINA, = 3.3 mg N‐NO3? L?1; α factor = 0.24), respectively. The rAOMAX value is smaller, and the KSAO value larger, than the values reported in diluted salts medium; the KIAO value is comparable to those reported. Process simulations with the kinetic model in batch nitrifying reactors showed that the inhibitory effects of nitrite and nitrate are significant for initial ammonia concentrations larger than 100 mg N‐NH4+ L?1. An experimental study on mixing, degree of suspension and power consumption in solid-liquid suspensions was done. A system similar to those found in anaerobic fermentation processes of animal manures was used, and an existing mixing equipment was adapted for the study. Power consumption and degree of suspension for both mechanical mixing and mixing by gas was determined. The influence of variables such as geometry, solids concentration, stirrer velocity, and gas velocity was studied, discussed, and compared to data from the literature. Best results were obtained for gas mixing, the power consumption being about one fourth of that required by mechanical agitation. Finally, extended correlations relating Power and Reynolds numbers for mechanical mixing and mixing by gas are proposed. 相似文献

In the present paper, we report results of surveys in 2003 in Japan and Denmark about patients' views about adverse events, focusing on the actions of healthcare staff involved in a medical accident. Results show that patients were more likely to indicate negative expectations to a doctor's reactions after a medical accident when asked in general terms than when asked in relation to concrete case stories. When asked in general terms, 66% (47%) of Japanese (Danish) respondents expected that doctors sometimes hold back on providing information to patients about a medical accident, while 37% (7%) did so when asked about a concrete, mild-outcome case. We examine some possible reasons for the relatively high level of distrust of Japanese patients, and we discuss whether the seemingly lower level of disclosure in Japan than in Denmark and the negative stories in the Japanese press may have an impact. We also suggest some implications for introducing a patient-centred or customer-centred approach to risk management in healthcare and other domains. 相似文献

The effects of different pH treatments with and without heating on the characteristics of wheat gluten suspension was investigated. At pH 1, maximum changes in colour were observed with a concurrent 65% decrease in protein free-thiol content compared to the control gluten. The SDS-Extractability of protein (SDS-EP) chromatogram eluted at lower retention time and the presence of bands at the top lane even during reducing conditions in SDS-PAGE gel suggested complex formation involving bonds other than disulphides. An increase in the free-amino group content and the presence of an additional peak at a higher retention time in the SDS-EP chromatogram was suggestive of hydrolysis. At pH 2 and 3, similar decreases in SDS-EPs and free-thiol content indicated formation of complexes. When heated, the free-thiol content of the dispersions increased compared to the non-heated dispersions indicating disruption of disulphide bonds with changes in gluten structure and size distribution. 相似文献

The influence of the type of backsheet on the electrical performance of test modules was evaluated before and after increasing time of accelerated ageing (damp heat [DH] exposure). Besides the measurement of the electrical power of the modules and the performance of the cells by electroluminescence, the ageing‐induced changes within the polymeric encapsulate and backsheets were investigated by means of vibrational spectroscopy and by thermo analytical methods. In addition, the permeability of the backsheets in the original and aged state was determined. This wide set of test parameters and methods allowed for the detection of correlations between (i) physical and chemical properties as well as their ageing‐induced changes of the materials and (ii) the module performance. A clear dependence of the relative loss in power output upon exposure under DH conditions for 2000 h could be observed for a set of identical test modules varied in composition only in the type of back cover used. While the modules containing gas‐tight backsheets and glass experienced only little loss in the relative power output, some modules with permeable backsheets showed a significant relative decrease in the power output and fill factor in dependence of the backsheet type used. Cell degradation could be visualised by recording electroluminescence images before and after the accelerated ageing test. The permeation properties of the backsheet used and their ageing‐induced changes seem to have an influence on the module performance. However, the absolute values neither of the water vapour transmission rate (WVTR) nor of the oxygen transmission rate (OTR) are directly linked to the loss in power output upon accelerated ageing under DH conditions. It could be shown that the ageing‐induced changes (relative transmission rates) between WVTR and OTR can be correlated with the module performance. These ageing‐induced changes in the permeation behaviour of the backsheets can be explained by (i) physical changes (e.g. post‐crystallisation, changes in the crystal structure or the crystalline microstructure) and (ii) chemical ageing effects such as a decrease in the molecular mass of the polyester (PET) polymer chains because of hydrolytic polymer degradation leading to a change in the crystallisation behaviour of PET. Hydrolytic degradation (= chemical ageing) of the PET core layer was observed (with varying extent) for all PET‐based backsheets and can, thus, not be directly correlated with the loss in performance of the corresponding test modules. The physical ageing effects, however, were detected only for those backsheets showing (i) strong deviating changes in the relative permeation rates for oxygen and water vapour upon accelerated ageing and (ii) a clear loss in electrical performance. Antifreeze proteins (AFPs) or thermal hysteresis (TH) proteins are biomolecular gifts of nature to sustain life in extremely cold environments. This family of peptides, glycopeptides and proteins produced by diverse organisms including bacteria, yeast, insects and fish act by non-colligatively depressing the freezing temperature of the water below its melting point in a process termed thermal hysteresis which is then responsible for ice crystal equilibrium and inhibition of ice recrystallisation; the major cause of cell dehydration, membrane rupture and subsequent cryodamage. Scientists on the other hand have been exploring various substances as cryoprotectants. Some of the cryoprotectants in use include trehalose, dimethyl sulfoxide (DMSO), ethylene glycol (EG), sucrose, propylene glycol (PG) and glycerol but their extensive application is limited mostly by toxicity, thus fueling the quest for better cryoprotectants. Hence, extracting or synthesizing antifreeze protein and testing their cryoprotective activity has become a popular topic among researchers. Research concerning AFPs encompasses lots of effort ranging from understanding their sources and mechanism of action, extraction and purification/synthesis to structural elucidation with the aim of achieving better outcomes in cryopreservation. This review explores the potential clinical application of AFPs in the cryopreservation of different cells, tissues and organs. Here, we discuss novel approaches, identify research gaps and propose future research directions in the application of AFPs based on recent studies with the aim of achieving successful clinical and commercial use of AFPs in the future. 相似文献

A rapid and selective liquid chromatography–electrospray ionisation mass spectrometry (LC–ESI-MS) method to screen citrus samples for limonoid glucosides and estimate their relative concentrations has been developed. This method utilises a phenyl stationary phase, whereas previous methods have relied on C-18. Samples may be analysed directly without treatment other than dilution. Peak areas from the extracted deprotonated molecular ion mass signals for individual limonoid glucosides were normalised against the sum of the areas to establish their relative concentrations. The method was successfully applied to the analysis of various juice, extracts, and liquid samples of partially purified limonoid glucosides. 相似文献

The multiple uses of the Danube River for navigation, hydropower production and as a drinking water supply influence its ecological quality. Flow restrictions due to regulation as well as impacts on the sediment regime such as retention in the catchment area and interruption of the sediment continuum result in changes of the hydromorphological structures and can produce major problems in the context of sediment management. In light of these facts, cross-border investigations of sediment transport at the Danube River are called for. As there are no uniform research approaches shared by Austria and Hungary, the EU-funded project SEDDON (Sediment Research and Management at the Danube River) aims to compare different monitoring and modeling techniques used to determine sediment transport, and to develop mutual solutions. Further, the laboratory equipment currently used for hydraulic model experiments is evaluated and a research channel with a free flowing discharge of up to 10 m3/s without pumping will be constructed, allowing large-scale joint laboratory tests concerning sediment to be performed. 相似文献
