1,25-Dihydroxycholecalciferol, the hormonally active vitamin D3 metabolite, is known to exhibit therapeutic effects against breast cancer, mainly by lowering the expression of estrogen receptors and aromatase activity. Previously, the safety of the vitamin D active metabolite (24R)-1,24-dihydroxycholecalciferol (PRI-2191) and 1,25(OH)2D3 analog PRI-2205 was tested, and the in vitro activity of these analogs against different cancer cell lines was studied. We determined the effect of the two vitamin D compounds on anastrozole (An) activity against breast cancer based on antiproliferative activity, ELISA, flow cytometry, enzyme inhibition potency, PCR, and xenograft study. Both the vitamin D active metabolite and synthetic analog regulated the growth of not only estrogen receptor-positive cells (T47D and MCF-7, in vitro and in vivo), but also hormone-independent cancer cells such as SKBR-3 (HER-2-positive) and MDA-MB-231 (triple-negative), despite their relatively low VDR expression. Combined with An, PRI-2191 and PRI-2205 significantly inhibited the tumor growth of MCF-7 cells. Potentiation of the antitumor activity in combined treatment of MCF-7 tumor-bearing mice is related to the reduced activity of aromatase by both An (enzyme inhibition) and vitamin D compounds (switched off/decreased aromatase gene expression, decreased expression of other genes related to estrogen signaling) and by regulation of the expression of the estrogen receptor ERα and VDR.  相似文献

Neoplasms derived from follicular tissue are extremely rare. Clinically, they are reported as non-symptomatic, slow-growing nodules. These lesions are mainly benign, but the malignant type can occur. Mainly middle-aged people (50–60 years of age) are affected. These carcinomas are mainly localized on the head and neck or torso. They can be locally aggressive and infiltrate surrounding tissue and metastasize to regional lymph nodes. In the minority of cases, distant metastases are diagnosed. Quick and relevant diagnosis is the basis of a treatment for all types of tumors. The patient’s life expectancy depends on multiple prognostic factors, including the primary tumor size and its mitotic count. Patients should be referred to a specialized skin cancer center to receive optimal multidisciplinary treatment. This article tries to summarize all the information that is currently available about pathogenesis, diagnosis, and treatment methods of follicular tumors.  相似文献

Protein solubility is based on the compatibility of the specific protein surface with the polar aquatic environment. The exposure of polar residues to the protein surface promotes the protein’s solubility in the polar environment. The aquatic environment also influences the folding process by favoring the centralization of hydrophobic residues with the simultaneous exposure to polar residues. The degree of compatibility of the residue distribution, with the model of the concentration of hydrophobic residues in the center of the molecule, with the simultaneous exposure of polar residues is determined by the sequence of amino acids in the chain. The fuzzy oil drop model enables the quantification of the degree of compatibility of the hydrophobicity distribution observed in the protein to a form fully consistent with the Gaussian 3D function, which expresses an idealized distribution that meets the preferences of the polar water environment. The varied degrees of compatibility of the distribution observed with the idealized one allow the prediction of preferences to interactions with molecules of different polarity, including water molecules in particular. This paper analyzes a set of proteins with different levels of hydrophobicity distribution in the context of the solubility of a given protein and the possibility of complex formation.  相似文献

Operation of mobile robots in off-road environment requires the attention to the torque saturation problem that occurs in the wheels DC motors while climbing hills. In the present work, off-road conditions are utilized to benefit while avoiding torque saturation. Energy optimization algorithm using predictive control is implemented on a two-DC motor-driven wheels mobile robot while crossing a ditch. The predictive control algorithm is simulated and compared with the PID control and the open-loop control. Predictive control showed more capability to avoid torque saturation and noticeable reduction in the energy consumption. Furthermore, using the wheels motors armature current instead of the supply voltage as control variable in the predictive control showed more efficient speed control. Simulation results showed that in case of known ditch dimensions ahead of time, the developed algorithm is feasible. Experimental examination of the developed energy optimization algorithm is presented. The experimental results showed a good agreement with the simulation results. The effects of the road slope and the prediction horizon length on the consumed energy are evaluated. The analytical study showed that the energy consumption is reduced by increasing the prediction horizon until it reaches a limit at which no more energy reduction is obtained. This limit is proportional to the width of the ditch in front of the mobile robot. Curve fitting is applied to the obtained results to address further the effect of the parameters on the energy consumption.  相似文献

Linseed cake (LC), a byproduct of linseed oil extraction, is used as a functional filler for production of biodegradable composites. To determine the influence of residual linseed crude oil contained in lignocellulosic filler on the properties of the poly(lactic acid) (PLA)-based composites with 5–30% filler content, two types of LC were analyzed: a defatted and an unmodified one. Complex analysis of the composites' properties change was conducted in relation to their structure modification caused by the addition of a waste filler. It was found that the addition of LC resulted in simultaneous plasticization and improved crystallization of PLA. Lignocellulosic particles and crude linseed oil contained in the LC powder provided a modifying effect, influencing the level of crystallinity and mechanical and thermomechanical properties. Manipulator mounted on an unmanned satellite could be used for performing orbital capture maneuver in order to repair satellites or remove space debris from orbit. Use of manipulators for such purposes presents unique challenges, as high level of autonomy is required and the motion of the manipulator influences the position and orientation of the manipulator-equipped satellite. This paper presents a new control system that consists of two modules: trajectory planning module (based on trajectory optimization algorithm) and Model Predictive Controller. Both modules take into account the free-floating nature of the satellite-manipulator system. Proposed control system was tested in numerical simulations performed for a simplified planar case. In the first set of simulations Nonlinear Model Predictive Control (NMPC) was used to ensure realization of a square reference end-effector trajectory, while in the second set control system was used for optimizing and then ensuring realization of the trajectory that leads to grasping of the rotating target satellite. Simulations were performed with disturbances and with the assumed non-perfect knowledge of parameters of the satellite-manipulator system. Results obtained with NMPC are better than results obtained with the controller based on the Dynamic Jacobian inverse and with the Modified Simple Adaptive Control (MSAC).  相似文献

This research was primarily conducted to verify the presence of synergistic effects in ternary cementitious systems containing portland cement (OPC), class C fly ash (FA) and silica fume (SF). A subsequent objective of the study was to quantify the magnitude of the synergy and to determine its source. For a ternary mixture containing 20% FA and 5% SF by mass, the synergistic effect was observed mostly at later ages (7 days onward) and it resulted in an increased compressive strength and resistance to chloride ion penetration as well as a reduced rate of water absorption (sorptivity) compared to predictions based on individual effects of FA and SF in respective binary systems. The observed synergy was attributed to both chemical and physical effects. The chemical effect manifested itself in the form of an increased amount of hydration products. The physical effect associated with packing density was, somewhat contrary to general belief, not due to an optimized particle size distribution of the binder components of the ternary cementitious system. Instead, it was the result of smaller initial inter-particle spacing caused by lower specific gravities of both FA and SF which, in turn, led to a lower volumetric w/cm. If the mixture design was adjusted to account for these differences, the physical effect would be diminished.  相似文献

Linseed cake (LC) is a by-product of agricultural industry which does not have any large-scale industrial applications. The possibilities of its utilization as filler with plasticizing ability for high density polyethylene (HDPE)-based composites have been investigated. Composites containing 5, 10, 20 and 30 wt% of the waste filler have been prepared using a melt mixing method. The influence of the LC on the mechanical and thermomechanical properties of the composites, as well as their water absorption and morphology, have been evaluated by the following methods: static tensile test, impact strength assessment using Dynstat method, hardness measurements, differential scanning calorimetry, dynamic mechanical thermal analysis, scanning electron microscopy observations, measurements of Vicat’s softening temperature and water uptake test. Application of different measuring techniques allows for describing complex modification effects of the composites’ properties changes induced by the presence of lignocellulosic filler with high oil content. The results of the study proved a pronounced influence of LC on high density polyethylene-based composites, especially a plasticizing effect of crude linseed oil contained by the waste filler particles. LC also has been assumed to affect the polymeric matrix crystallization process. It was found that complex modification of polyethylene results from simultaneously occurring different phenomena including: plasticization of the HDPE by linseed oil, improved crystallinity of the semicrystalline matrix, presence of the rigid lignocellulosic particles dispersed in polymer and accumulation of the oil in the interfacial regions.  相似文献
