Kaempferol is a flavonoid that occurs in tea and in many vegetables and fruits, including broccoli, cabbage, beans, grapes, apples, and strawberries. The efficacy of Kaempferol has been demonstrated in the treatment of breast, esophageal, cervical, ovarian, and liver cancers and leukemia, which very likely arises from its prooxidant properties and the activation of pro-apoptotic pathways. Indeed, this matter has already been the focus of a number of published studies and reviews. The aim of the present study was to elucidate the antioxidant vs. prooxidant properties of flavonoids in the presence of the redox-active metal, copper (II) ion, by means of the Fenton reaction. The specific motivation of this work is that, since an increased level of Cu(II) ions is known to be associated with many disease states such as neurological conditions (Alzheimer’s disease) and cancer, any interaction between these ions and flavonoids might affect the outcome of therapeutic uses of the latter. The structure of the Cu-kaempferol complex in DMSO was investigated by means of low temperature EPR spectroscopy, which confirmed the existence of at least two distinct coordination environments around the copper (II) ion. UV vis-spectra of kaempferol and its Cu(II) complex in DMSO revealed an interaction between the 5-OH (A ring) group and the 4-CO (C ring) group of kaempferol with Cu(II) ions. An ABTS assay confirmed that kaempferol acted as an effective radical scavenger, and that this effect was further enhanced in the form of the Cu(II)-kaempferol complex. Quantitative EPR spin trapping experiments, using DMPO as the spin trap, confirmed suppression of the formation of a mixture of hydroxyl, superoxide, and methyl radicals, in a Fenton reaction system, upon coordination of kaempferol to the redox-active Cu(II) ions, by 80% with respect to the free Cu(II) ions. A viscometric study revealed a better DNA-intercalating ability of the Cu-kaempferol complex than for free kaempferol, essential for conferring anticancer activity of these substances. The results of the viscometric measurements were compared with those from a DNA damage study of Cu-kaempferol complexes in a Fenton reaction system, using gel electrophoresis. At low concentrations of kaempferol (Cu–kaempferol ratios of 1:1 and 1:2), a very weak protective effect on DNA was noted, whereas when kaempferol was present in excess, a significant DNA-protective effect was found. This can be explained if the weakly intercalated kaempferol molecules present at the surface of DNA provide protection against attack by ROS that originate from the Fenton reaction involving intercalated Cu(II)-kaempferol complexes. Following the application of ROS scavengers, L-histidine, DMSO, and SOD, gel electrophoresis confirmed the formation of singlet oxygen, hydroxyl radicals, and superoxide radical anions, respectively. We propose that the prooxidant properties of Cu-kaempferol complexes may provide anticancer activity of these substances. When present in excess, kaempferol displays antioxidant properties under Cu-Fenton conditions. This suggests that kaempferol might prove a suitable candidate for the prevention or treatment of oxidative stress related medical conditions that involve a disturbed metabolism of redox metals such as copper, for example, Menkes disease, and neurological disorders, including Alzheimer’s disease. For the potential use of kaempferol in clinical practice, it will be necessary to optimize the dose size and critical age of the patient so that this flavonoid may be beneficial as a preventive drug against cancer and neurological disorders.  相似文献

By using a bio-mechanochemical approach combining mechanochemistry (ball milling) and green synthesis for the first time, silver nanoparticles (Ag NPs) with antibacterial activity were successfully synthesized. Concretely, eggshell membrane (ESM) or Origanum vulgare L. plant (ORE) and silver nitrate were used as environmentally friendly reducing agent and Ag precursor, respectively. The whole synthesis took 30?min in the former and 45?min in the latter case. The photon cross-correlation measurements have shown finer character of the product in the case of milling with Origanum. UV–Vis measurements have shown the formation of spherical NPs in both samples. TEM study has revealed that both samples are composites of nanosized silver nanoparticles homogenously dispersed within the organic matrices. It has shown that the size and size distribution of the silver nanoparticles is smaller and more uniform in the case of eggshell membrane matrix implying lower silver mobility within this matrix. The antibacterial activity was higher for the silver nanoparticles synthesized with co-milling with Origanum plant than in the case of milling with eggshell membrane.  相似文献

Adhesives are widely used in electronic packaging and are of vital importance in the reliability of electronic systems. A deep understanding of the degradation mechanism of adhesives under corrosion load plays a key role in lifetime prediction. Unfortunately, most of the common reliability tests are destructive. The present approach combines the nondestructive methods, electrochemical impedance spectroscopy and the Fourier-transform infrared spectroscopy, as powerful tools in a complementary manner to describe the degradation mechanism and kinetics of two epoxy-based adhesives, which are commonly used in electronic packaging. It is demonstrated that the application quality is the dominating impact on the optimization of the lifetime.  相似文献

Many ecological interactions in communities take place between consumers and the organisms they feed on. Continuous surplus of specific nutritional compounds in the diet may lead to evolutionary changes in the metabolic capacity of the consumer, leaving the biosynthesis of such compounds prone to genetic decay and render organisms auxotrophic. A nutrient that is essential to many organisms is the unsaturated fatty acid, linoleic acid (LA; 18:2n-6), which is important in the maintenance of cell membrane fluidity and as a precursor for signaling molecules. LA is readily synthesized in bacteria, protozoa and plants, but it was long thought that all animals lack this ability. Although the majority of animals lack the ability for LA biosynthesis, an increasing number of studies have shown that LA is commonly synthesized in arthropods. Here, we investigated a basal hexapod group, Collembola, to shed light on early evolution of LA synthetic ability in arthropods and its relation to dietary composition. We use stable isotope labeling to detect biosynthesis of LA in Collembola fed with 13C–OA oleic acid (OA; 18:1n-9), a precursor of LA. Our data demonstrate that LA biosynthesis is common among Collembola with 10 out of 16 tested species being able to synthesize LA and 4 species lacking this ability. However, we did not find clear evidence for a relationship between LA synthetic ability and the natural diet of species. Thus, the selective pressures underlying LA biosynthesis might be species-specific and further research will shed new light on understanding this evolutionary process.  相似文献

Nano- and submicro-scaled ZrO2 particles were codeposited into Ni matrix by an electrochemical plating process using a Watts bath. The incorporation has been studied in relation to dispersion stability. The ZrO2 particles were characterized through zeta potential electro-acoustic measurements and particle size distribution. SEM and EDX analysis of the codeposited layers were performed to characterise the coating morphology and to determine the particle concentration in the layer. A content up to 10 vol.% of ZrO2 was found in Ni matrix. As electrostatic repulsion within the dispersion increases, particle agglomeration slows down and the concentration of non-agglomerated particles rises resulting in an enhanced amount of particles incorporated in the electroplated Ni layers. The effect of electrophoresis and diffusion on particle transport to the cathode is discussed.  相似文献

Background: Cardiovascular surgery is confronted by a lack of suitable materials for patch repair. Acellular animal tissues serve as an abundant source of promising biomaterials. The aim of our study was to explore the bio-integration of decellularized or recellularized pericardial matrices in vivo. Methods: Porcine (allograft) and ovine (heterograft, xenograft) pericardia were decellularized using 1% sodium dodecyl sulfate ((1) Allo-decel and (2) Xeno-decel). We used two cell types for pressure-stimulated recellularization in a bioreactor: autologous adipose tissue-derived stromal cells (ASCs) isolated from subcutaneous fat of pigs ((3) Allo-ASC and (4) Xeno-ASC) and allogeneic Wharton’s jelly mesenchymal stem cells (WJCs) ((5) Allo-WJC and (6) Xeno-WJC). These six experimental patches were implanted in porcine carotid arteries for one month. For comparison, we also implanted six types of control patches, namely, arterial or venous autografts, expanded polytetrafluoroethylene (ePTFE Propaten® Gore®), polyethylene terephthalate (PET Vascutek®), chemically stabilized bovine pericardium (XenoSure®), and detoxified porcine pericardium (BioIntegral® NoReact®). The grafts were evaluated through the use of flowmetry, angiography, and histological examination. Results: All grafts were well-integrated and patent with no signs of thrombosis, stenosis, or aneurysm. A histological analysis revealed that the arterial autograft resembled a native artery. All other control and experimental patches developed neo-adventitial inflammation (NAI) and neo-intimal hyperplasia (NIH), and the endothelial lining was present. NAI and NIH were most prominent on XenoSure® and Xeno-decel and least prominent on NoReact®. In xenografts, the degree of NIH developed in the following order: Xeno-decel > Xeno-ASC > Xeno-WJC. NAI and patch resorption increased in Allo-ASC and Xeno-ASC and decreased in Allo-WJC and Xeno-WJC. Conclusions: In our setting, pre-implant seeding with ASC or WJC had a modest impact on vascular patch remodeling. However, ASC increased the neo-adventitial inflammatory reaction and patch resorption, suggesting accelerated remodeling. WJC mitigated this response, as well as neo-intimal hyperplasia on xenografts, suggesting immunomodulatory properties.  相似文献
