An optical nondestructive strain measurement technique was performed to analyze the mechanical deformation induced by an electrical field within the insulating materials. Poly(ethylene naphthalene 2,6‐dicarboxylate) (PEN) films were then subjected to constant electrical fields right up to their electrical breakdown. The experimental technique made it possible to follow the various stages of the mechanical behavior of PEN in real time. The final breakdown occurred in the observation zone and the related mechanical deformation was captured. A “margarita” structure was observed with a hole at the center. The experimental results indicated that the level of the induced‐mechanical deformations depended on the local environment. We defined two different zones representing the inside and the outside of the damaged area. The induced‐deformations were larger in the damaged zone. It was also observed that deformations increased when the sample had a lower degree of crystallinity. © 2008 Wiley Periodicals, Inc. J Appl Polym Sci, 2008  相似文献

The combined generation of heat and power (cogeneration) is praised by many as a technique for reducing the emissions of CO2 in industrialized nations. This is generally true but not always. In this article we discuss the impact of some major variables on the CO2 emission reduction capacity of cogeneration. Two sets of variables are predominant: the characteristics of the CHP process and the composition of the electricity generation sector. We consider a class of time-varying -valued control models, and with possibly unbounded costs. Melanoma is a complex and heterogenous disease, displays the deadliest form of skin cancer, and accounts for approx. 80% of all skin cancer deaths. In this study, we reported on the synthesis and pharmacological effects of a novel shikonin derivative (SK119), which is active in a nano-molar range and exhibits several promising in vitro effects in different human melanoma cells. SK119 was synthesized from shikonin as part of our search for novel, promising shikonin derivatives. It was screened against a panel of melanoma and non-tumorigenic cell lines using XTT viability assays. Moreover, we studied its pharmacological effects using apoptosis and Western blot experiments. Finally, it was combined with current clinically used melanoma therapeutics. SK119 exhibited IC50 values in a nano-molar range, induced apoptosis and led to a dose-dependent increase in the expression and protein phosphorylation of HSP27 and HSP90 in WM9 and MUG-Mel 2 cells. Combinatorial treatment, which is highly recommended in melanoma, revealed the synergistic effects of SK119 with vemurafenib and cobimetinib. SK119 treatment changed the expression levels of apoptosis genes and death receptor expression and exhibited synergistic effects with vemurafenib and cobimetinib in human melanoma cells. Further research indicates a promising potential in melanoma therapy.  相似文献

Reg-1α/lithostathine, a protein mainly associated with the digestive system, was previously shown to be overexpressed in the pre-clinical stages of Alzheimer’s disease. In vitro, the glycosylated protein was reported to form fibrils at physiological pH following the proteolytic action of trypsin. However, the nature of the protease able to act in the central nervous system is unknown. In the present study, we showed that Reg-1α can be cleaved in vitro by calpain-2, the calcium activated neutral protease, overexpressed in neurodegenerative diseases. Using chemical crosslinking experiments, we found that the two proteins can interact with each other. Identification of the cleavage site using mass spectrometry, between Gln4 and Thr5, was found in agreement with the in silico prediction of the calpain cleavage site, in a position different from the one reported for trypsin, i.e., Arg11-Ile12 peptide bond. We showed that the cleavage was impeded by the presence of the neighboring glycosylation of Thr5. Moreover, in vitro studies using electron microscopy showed that calpain-cleaved protein does not form fibrils as observed after trypsin cleavage. Collectively, our results show that calpain-2 cleaves Reg-1α in vitro, and that this action is not associated with fibril formation.  相似文献

The voltage-dependent L-type calcium channel isoform CaV1.2 is critically involved in many physiological processes, e.g., in cardiac action potential formation, electromechanical coupling and regulation of insulin secretion by beta cells. Gain-of-function mutations in the calcium voltage-gated channel subunit alpha 1 C (CACNA1C) gene, encoding the CaV1.2 α1-subunit, cause Timothy syndrome (TS), a multisystemic disorder that includes autism spectrum disorders and long QT (LQT) syndrome. Strikingly, TS patients frequently suffer from hypoglycemia of yet unproven origin. Using next-generation sequencing, we identified a novel heterozygous CACNA1C mutation in a patient with congenital hyperinsulinism (CHI) and associated hypoglycemic episodes. We characterized the electrophysiological phenotype of the mutated channel using voltage-clamp recordings and in silico action potential modeling experiments. The identified CaV1.2L566P mutation causes a mixed electrophysiological phenotype of gain- and loss-of-function effects. In silico action potential modeling supports that this mixed electrophysiological phenotype leads to a tissue-specific impact on beta cells compared to cardiomyocytes. Thus, CACNA1C variants may be associated with non-syndromic hyperinsulinemic hypoglycemia without long-QT syndrome, explained by very specific electrophysiological properties of the mutated channel. We discuss different biochemical characteristics and clinical impacts of hypoglycemia in the context of CACNA1C variants and show that these may be associated with significant morbidity for Timothy Syndrome patients. Our findings underline that the potential of hypoglycemia warrants careful attention in patients with CACNA1C variants, and such variants should be included in the differential diagnosis of non-syndromic congenital hyperinsulinism.  相似文献

UNICORE is a European Grid Technology with more than 10 years of history. Originating from the Supercomputing domain, the latest version UNICORE 6 has turned into a general-purpose Grid technology that follows established standards and offers a rich set of features to its users. The paper starts with an architectural insight into UNICORE 6, highlighting the workflow features, standards and the different clients. Organic pigments are important crystalline substances, and their properties and applications rely on size and shape control. Pigment Yellow 181 (PY181) is an industrial azo pigment that is light and weatherfast and suitable for high temperature processing. One disadvantage is its needle‐like shape in the default β‐phase, which makes the pigment difficult to process in industry, e.g., in polymer melts, where a spherical structure would be ideal. Here, we show for the first time, that polymer‐induced liquid precursor structures can be formed even in association to a chemical reaction. Furthermore, it is demonstrated that biomineralization principles can be exploited for the generation of advanced functional materials, such as pigments with novel complex morphology and different properties. Stable PY181 microspheres of nanoplates in the β‐phase were obtained in mixed solvents of water and isopropanol by direct azo coupling under the directing influence of a designed copolymer additive aminobenzoylaminobenzamide‐acetoacetyl‐poly(ethylene imine)‐block‐poly(ethylene glycol) (ABABA‐acetoacetyl‐PEI‐b‐PEG).  相似文献

Freestanding silicon nanocrystals (Si‐ncs) offer unique optical and electronic properties for new photovoltaic, thermoelectric, and other electronic devices. A method to fabricate Si‐ncs which is scalable to industrial usage has been developed in recent years. However, barriers to the widespread utilization of these nanocrystals are the presence of charge‐trapping defects and an oxide shell formed upon ambient atmosphere exposure hindering the charge transport. Here, we exploit low‐cost post‐growth treatment routes based on wet‐etching in hydrofluoric acid plus surface hydrosilylation or annealing enabling a complete native oxide removal and a reduction of the defect density by up to two orders of magnitude. Moreover, when compared with only H‐terminated Si‐ncs we report an enhancement of the conductivity by up to a factor of 400 for films of HF etched and annealed Si‐ncs, which retain a defect density below that of untreated Si‐ncs even after several months of air exposure. Further, we demonstrate that HF etched and hydrosilylated Si‐ncs are extremely stable against oxidation and maintain a very low defect density after a long‐term storage in air, opening the possibility of device processing in ambient atmosphere.  相似文献

Modularity and self-healing are two interesting properties that could help to design more flexible conveyors of micro-objects. In the Smart Blocks project, we propose to design a 2D modular and self-reconfigurable robot composed of centimeter-scale sliding blocks that embed their own actuators and control electronics. This article presents a proof-of-concept of the linkage and of the traveling system as well as an algorithm able to reconfigure a set of blocks from a spatial configuration to another one. Prototype blocks have been realized using electro-permanent magnets which show a good motion speed while saving power consumption during the linkage. Our reconfiguration algorithm is implemented in a simulator software showing in real-time the reconfiguration of the robot.  相似文献
