BACKGROUND: Both fibroblast-mediated cytokine gene therapy and bone marrow transplantation (BMT) have proven to be efficient protocols for the recovery of bone marrow depression. In this report, the effects of fibroblast-mediated interleukin (IL)-6 gene therapy, in combination with BMT, on the recovery of irradiation-induced bone marrow depression were investigated. METHODS: NIH3T3 fibroblast cells engineered to secrete IL-6 (NIH3T3-IL-6) or NIH3T3 cells transduced with the neomycin gene (NIH3T3-Neo), in combination with 10(7), 10(6), or 10(5) syngeneic bone marrow cells, were implanted into irradiated mice. RESULTS: The platelets and white blood cells in the peripheral blood of the irradiated mice increased greatly 12 days after implantation of NIH3T3-IL-6 cells and BMT, the white blood cell counts were restored to a normal level 32 days after the combined therapy, and the platelet number was obviously higher than that in mice implanted with NIH3T3-Neo and BMT. Twenty and 25 days after the combined therapy, the mice showed accelerated recovery of colony-forming unit (CFU)-granulocyte/macrophages and CFU-megakaryocytes when compared with the mice implanted with NIH3T3-Neo cells and BMT. Ten days after lethal irradiation with gamma rays, the spleens formed more CFU-spleen in mice implanted with NIH3T3-IL-6 cells and BMT than in mice injected with phosphate-buffered saline or NIH3T3-Neo cells. Combined therapy with NIH3T3-IL-6 cell implantation and BMT delayed the survival period of the hematopoietic-depressed mice significantly when compared with therapy with NIH3T3-Neo cell implantation and BMT. CONCLUSIONS: These data demonstrated that the combined therapy of fibroblast-mediated IL-6 gene therapy and BMT could significantly promote the recovery of irradiation-induced hematopoietic depression. 相似文献

Beneficial use impairments (BUIs) under the Great Lakes Water Quality Agreement identify environmental issues requiring remedial action within the Great Lakes Areas of Concern (AOCs). We conducted this study to support the assessment of the wildlife component of BUI 3: degradation of fish and wildlife populations. We compared bird and amphibian (frogs and toads) data from the Toronto and Region Conservation Authority’s Terrestrial Long-term Monitoring Program in the Toronto and Region AOC to an adjacent, but otherwise similar, reference watershed, Duffins Creek. Twelve of 13 targets were met within the AOC for forest bird, wetland bird, meadow bird and amphibian populations based on averages of mean annual values at sites within the AOC that were within two standard deviations of averages at sites in the Duffins Creek reference watershed between 2008 and 2017. Even though wildlife populations within the AOC were within the normal range of variability expected from a reference watershed, they were often at lower levels than within the Duffins Creek reference watershed. In addition, forest bird and amphibian populations were negatively affected by urbanization within the AOC and meadow bird indices declined. We conclude that while wildlife populations within the AOC currently meet targets for BUI 3, they continue to be negatively impacted by numerous stressors that are primarily related to past and ongoing urbanization. Thus, continued restoration of wildlife habitat and protection of existing habitat within the AOC is highly recommended. 相似文献

A method of controlling anchoring energy of surface interaction of liquid crystal photoaligning substances is considered to enhance the liquid crystal display performances. An important parameter of the dye's molecular structure that determines the ratio of polar and azimuthal anchoring energy is the ability to form dimers. The values of dimerization thermodynamic potentials have been found. The probability of the formation of dye molecules dimers is evaluated. The bonds conjugation transfer via intermolecular hydrogen bond is revealed, and anisotropy of polarizability of the hydrogen bond is evaluated. Protein–protein interactions (PPIs) play a fundamental role in various biological functions; thus, detecting PPI sites is essential for understanding diseases and developing new drugs. PPI prediction is of particular relevance for the development of drugs employing targeted protein degradation, as their efficacy relies on the formation of a stable ternary complex involving two proteins. However, experimental methods to detect PPI sites are both costly and time-intensive. In recent years, machine learning-based methods have been developed as screening tools. While they are computationally more efficient than traditional docking methods and thus allow rapid execution, these tools have so far primarily been based on sequence information, and they are therefore limited in their ability to address spatial requirements. In addition, they have to date not been applied to targeted protein degradation. Here, we present a new deep learning architecture based on the concept of graph representation learning that can predict interaction sites and interactions of proteins based on their surface representations. We demonstrate that our model reaches state-of-the-art performance using AUROC scores on the established MaSIF dataset. We furthermore introduce a new dataset with more diverse protein interactions and show that our model generalizes well to this new data. These generalization capabilities allow our model to predict the PPIs relevant for targeted protein degradation, which we show by demonstrating the high accuracy of our model for PPI prediction on the available ternary complex data. Our results suggest that PPI prediction models can be a valuable tool for screening protein pairs while developing new drugs for targeted protein degradation. 相似文献

ABSTRACT Northwest Arkansas planning policies, like a number of communities across the country, have identified goals working toward more sustainable, livable, and subsequently denser development patterns. However, the understanding of residents’ perceptions of such living arrangements is limited. This study provides a more nuanced understanding of Northwest Arkansas residents’ spatial preferences through a survey of residents’ preferences for private amenities and their trade-off with various spatial densities and patterns in support of sustainability. Results of the survey indicate a preference for, and experience with, single-family residential living arrangements and amenities, with the preponderance (80%) of survey respondents currently living in single-family housing. There is a preference for low-density neighborhoods even if it means sacrificing other amenities. This study is in alignment with previous research suggesting that people may learn to prefer where they live. Additionally, while the majority of survey respondents indicated a preference for communal greenspaces, renters are more likely to prefer communal greenspaces when compared to homeowners. This study indicates that attached, multi-family development and renter development in Northwest Arkansas should consider the provision of communal green spaces, walkable access to transit, and walkable access to services as desired amenities for those residents. 相似文献

Iron and iron oxide nanoparticles (NPs) are finding wide applications for the remediation of various toxic chloro-organic compounds (such as trichloroethylene, TCE), via reductive and oxidative processes. In this study, Fe NPs (30-50 nm) are synthesized by reduction from ferric ions immobilized (by ion exchange) on a platform (two types of sulfonated silica particles), in order to prevent the NP agglomeration. Next, the Fe NPs are oxidized and their effectiveness for the oxidative dechlorination of TCE via the heterogeneous decomposition of hydrogen peroxide to OH? on the surface of the iron oxide NPs was demonstrated. For the reductive approach, the use of ascorbic acid as a "green" reducing agent in conjunction with a secondary metal (Pd) inhibits NP oxidation and agglomeration through surface adsorbed species. Although several measures of obsessive-compulsive (OC) symptoms exist, most are limited in that they are not consistent with the most recent empirical findings on the nature and dimensional structure of obsessions and compulsions. In the present research, the authors developed and evaluated a measure called the Dimensional Obsessive-Compulsive Scale (DOCS) to address limitations of existing OC symptom measures. The DOCS is a 20-item measure that assesses the four dimensions of OC symptoms most reliably replicated in previous structural research. Factorial validity of the DOCS was supported by exploratory and confirmatory factor analyses of 3 samples, including individuals with OC disorder, those with other anxiety disorders, and nonclinical individuals. Scores on the DOCS displayed good performance on indices of reliability and validity, as well as sensitivity to treatment and diagnostic sensitivity, and hold promise as a measure of OC symptoms in clinical and research settings.
