The effects of partially hydrolyzed, nonviscous, guar gum (PHGG) on cholesterol metabolism and digestive balance have been compared with those of native guar gum (GUAR) in rats adapted to 0.4% cholesterol diets. Both types of guar gum elicited acidic fermentations in the large intestine, but only GUAR effectively lowered plasma cholesterol (P<0.001), chiefly in the triglyceride-rich lipoprotein fraction. The biliary bile acid excretion was significantly enhanced in rats fed GUAR (P<0.05), as well as the intestinal and cecal bile acid pool (P<0.001). In rats fed GUAR and to a lesser extent in those fed PHGG, the fecal excretion of bile acids and neutral sterol was higher than in controls (P<0.01). The digestive balance (cholesterol intake-steroid excretion) was positive in control rats (+47 μmol/d), whereas it was negative in rats fed GUAR (−20 μmol/d), which could involve a higher rate of endogenous cholesterol synthesis. In rats fed PHGG, the steroid balance remained slightly positive. Liver 3-hydroxy-3-methylglutaryl-CoA (HMG-CoA) reductase activity was very low (22 pmol/min/mg protein), owing to cholesterol supplementation, in control rats or in rats fed PHGG, whereas it was markedly higher (+463%) in rats fed GUAR. In conclusion, even if PHGG does alter some parameters of the enterohepatic cycle of cholesterol and bile acids, its effects are not sufficient to elicit a significant cholesterol-lowering effect. The intestinal (ileal or cecal) reabsorption of bile acids was not reduced, but rather increased, by GUAR; nevertheless the intestinal capacities of reabsorption were overwhelmed by the enlargement of the digestive pool of bile acids. In the present model, induction of HMG-CoA reductase probably takes place in the presence of elevated portal bile acid concentrations.  相似文献

Implementation of a rule-based transformation engine consists of several tasks with various abstraction levels. We present a new tool called mtom for the efficient implementation of rule-based transformations. This engine should help to bridge the gap between rewriting implementations and practical applications. It aims at implementing well-identified parts of complex applications where the use of rewriting is natural or crucial. These parts are specified using rewrite rules and integrated with the rest of the application, which is kept in a classical imperative language such as C, C++ or Java. Our tool, which can be viewed as a Yacc-like pre-processor, does not depend on a given term representation, rather it accepts implementation of terms (or term like data-types) of yet existing applications and it permits to define and execute rewrite rules upon those types. From our experiences, this system is well-suited for industrial use as well as for implementations of rule-based languages. The paper introduces several features supported by mtom.  相似文献

Poly(oxyethylene 20 sorbitan) monolaurate (Tween® 20) methacrylates were synthesized by coupling methacryloyl chloride (MeOCl) to Tween 20 in the presence of 4-(N,N-dimethylamino) pyridine, using THF as a solvent, in order to investigate their suitability as precursors for photopolymerizable hydrogels in tissue engineering applications. The degree of substitution could be controlled by adjusting the molar ratio of MeOCl and Tween 20, giving three different monomers: Tween 20 monomethacrylate, Tween 20 dimethacrylate and Tween 20 trimethacrylate. Combined 1 H NMR and MALDI-TOF MS confirmed these monomers to be of high purity and to have polydispersities less than 1.3. It was shown that aqueous solutions of the monomers were photoactive, all the methacrylate groups reacting within 30 minutes exposure to a UV light intensity of 145 mW/cm2. Aqueous Tween 20 trimethacrylate was then combined with N-vinyl-2-pyrrolidone (NVP), giving tough copolymer hydrogels on photopolymerization, whose swelling ratios and swelling rates could be tuned by varying the Tween 20 trimethacrylate content. The use of a flexible spacer with a multifunctional monomer gives a permanent three-dimensional network, whilst maintaining degrees of swelling of between 60 and 85%, with potential for a wide range of biological and non-biological applications.  相似文献

A role for GH in the pathogenesis of diabetic retinopathy has long been postulated. Previous clinical studies, however, have been confounded by hyperglycemia. We have identified 2 cases of retinopathy associated with exogenous GH therapy in nondiabetic patients. Cases were identified through the MedWatch drug surveillance system of the U.S. Food and Drug Administration. Causality by concomitant medications was excluded by a search of the literature and the FDA data base. The first patient, an obese, 31-yr-old male with traumatic hypothalamic injury, presented with nonproliferative retinopathy and macular edema, resulting in decreased visual acuity (OD 20/40-1; OS count fingers), which required laser surgery. Human GH had been initiated at 0.009 mg/ kg.day, 14 months earlier, and titrated to 0.017 mg/kg.day. The second patient, a nonobese, 11-yr-old girl receiving GH for the management of short stature in Turner's Syndrome, presented with neovascularization. GH doses were 0.033 mg/kg.day for the first 17 months and 0.043 mg/ kg.day for the following 5 months. Cumulative laboratory and clinical observations suggest that GH and related peptides have a role in retinal pathology independent of the degree of glucose tolerance.  相似文献

The protective role of selenium (Se), given as a Se-rich yeast, selenomethionine or selenomethionine+vitamin E supplement, toward changes in lipid, peroxide, and fatty acid distribution in tissues of streptozotocin-induced diabetic rats, was investigated, after 24 wk of disease. Diabetes increased liver thiobarbituric acid-reactive substances and conjugated dienes; Se supplement completely corrected these changes. In kidney, as in heart, the peroxide levels were not significantly changed by diabetes. In diabetic rat liver, a significant drop in triglycerides and phospholipids (P<0.05) was observed; this was modulated by Se+vitamin F supplementation. Se+vitamin E supplementation also inhibited the decrease in 18∶2n-6 and the increase in 22∶6n-3 observed in liver of diabetic rats, changes which reflect altered glycemic control. In kidney, heart, and aorta, diabetes produced some changes in lipid content and fatty acid distribution, especially an increase in heart triglycerides which was also corrected by the Se supplement. Se supplementation to diabetic rats also increased 18∶0 etherlinked alcohol, 20∶4 n-6, and 22∶5 n-3 in cardiac lipids. In aorta, Se + vitamin E significanlty increased 20∶5 n-3. These polyunsaturated fatty acids are precursors, in situ, of prostaglandin l2 (PGl2) and PGl3 which may protect against cardiovascular dysfunction. In kidney, converrely, Se decreased 20∶4 n-6, the precursor of thromboxane A1 implicated in diabetic glomerular injury. thus Se, and more efficiently Se + vitamin E supplementation, in experimental diabetes could play a role in controlling oxidative status and altered lipid metabolism in liver, thereby maintaining favorable fatty acid distribution in the major tissues affected by diabetic complications.  相似文献

BACKGROUND: Soybean seeds are rich in natural protein and are favorable environments for targeted protein expression. Soybeans represent an ideal platform for the production of novel vaccines that, in theory, do not require a cold chain. This study investigated the stability of a soybean‐derived antigen following long‐term storage, formulation, and shipment overseas in the absence of refrigeration. RESULTS: Transgenic seeds can be stored for more than 4 years at ambient temperature with no detectable loss of FanC antigen stability. Conventional processing methods utilizing heat, mechanical extraction and solvent extraction resulted in practical formulations that could be lyophilized, stored as dried milk powder and rehydrated in water without loss of FanC antigen stability. Overseas shipment of transgenic seed powder and soymilk formulations in the absence of a cold chain had no adverse effects on formulations or the FanC antigen. CONCLUSION: The feasibility of long‐term storage, processing and shipment of transgenic soy products in the absence of a cold chain was demonstrated. Soybeans represent a practical platform for development of novel vaccines to potentially address the worldwide need for vaccines that are cost‐effective, easy to formulate and can be manufactured, stored and transported without refrigeration.
