Summary Linear discriminant analysis of GLC-aromagram data is used to separate several classes of wine distillates, namely cognac, armagnac and brandy. In this paper we demonstrate how this method was used to detect adulteration. The adulteration was confirmed by the presence of ethyl heptanoate and limonene, which are well-known flavour compounds. They were analysed by means of a Purge and Trap headspace technique. Mass spectrometry was used to identify these flavours.

Zusammenfassung Die Linear-Discriminanz-Analyse von GLC-Aromagramm-Daten wird zur Klassifizierung von Weindestillaten wie Cognac, Armagnac und Brandy verwendet. In dieser Arbeit werden mit dieser Methode Verfälschungen nachgewiesen, die durch die Gegenwart der Ethylester der Heptansäure und des Limonen belegt werden konnten. Diese Substanzen wurden mit der Purge- und Trap-Methode erfaßt und massenspektrometrisch identifiziert.

Information technology is becoming increasingly important for entrepreneurs. Protecting their technical infrastructure and stored data is, therefore, also becoming more vital. Nevertheless, research into the safety and security of entrepreneurs in general, and online threats targeted at entrepreneurs in particular, is still limited. This study investigates to what extent self-employed entrepreneurs protect themselves against online threats, and what motivates them to do so. Based on secondary analyses on data collected from 1622 Dutch entrepreneurs, we observe that the majority implement technical and personal coping measures. By adopting protection motivation theory as a theoretical basis for our study, we found that coping appraisal explains the adoption of protective measures. Entrepreneurs are likely to implement protective measures when they believe a measure is effective, when they are capable of using internet technology, when their attitude towards online protection is positive and when they believe they are responsible for their own online security. Although the secondary analysis provides some limitations, this study offers new insights into the usage of protective measures and the motivation for taking them. These insights can help to improve information security campaigns.  相似文献

Using data from the first two waves of the Netherlands Kinship Panel Study for 338 women and 262 men, we examine the consequences of making the transition to parenthood for life satisfaction, loneliness, positive affect, negative affect, and partnership satisfaction. We extend previous work by taking transitions in partner status and work hours into account. Results show a moderate impact of becoming a parent on well-being. In so far as effects of making the transition to parenthood emerge, they are attributable to changes in partner status and work hours. First, the decrease in negative affect upon making the transition to motherhood is attributable to the group of women who increase their working hours. Second, the detrimental impact of making the transition to motherhood on partnership satisfaction is attributable to the group of new mothers who quit their job. Third, the detrimental impact of making the transition to fatherhood on loneliness is attributable to the group of new fathers who become married. There is one exception to this pattern of partner status and work hours as mechanisms for changes in well-being. Men who become fathers remain less satisfied with their partnership, even when transitions in partner status and work hours have been taken into account. In the discussion-section, we consider the possible underestimation of negative effects because of the focus on the continuously partnered. We also reflect on our results in the light of the high incidence of part-time work in the Netherlands and Dutch policies aimed at supporting new parents. The present study investigated the relationships between adolescents' online communication and compulsive Internet use, depression, and loneliness. The study had a 2-wave longitudinal design with an interval of 6 months. The sample consisted of 663 students, 318 male and 345 female, ages 12 to 15 years. Questionnaires were administered in a classroom setting. The results showed that instant messenger use and chatting in chat rooms were positively related to compulsive Internet use 6 months later. Moreover, in agreement with the well-known HomeNet study (R. Kraut et al., 1998), instant messenger use was positively associated with depression 6 months later. Azo dye reduction results from a combination of biotic and abiotic processes during the anaerobic treatment of dye containing effluents. Biotic processes are due to enzymatic reactions whereas the chemical reaction is due to sulfide. In this research, the relative impact of the different azo dye reduction mechanisms was determined by investigating the reduction of Acid Orange 7 (AO7) and Reactive Red 2 (RR2) under different conditions. Reduction rates of two azo dyes were compared in batch assays over a range of sulphide concentrations in the presence of living or inactivated anaerobic granular sludge. Biological dye reduction followed zero order kinetics and chemical dye reduction followed second-order rate kinetics as a function of sulfide and dye concentration. Chemical reduction of the dyes was greatly stimulated in the presence of autoclaved sludge: whereas chemical dye reduction was not affected by living or gamma-irradiated-sludge. Presumably redox-mediating enzyme cofactors released by cell lysis contributed to the stimulatory effect. This hypothesis was confirmed in assays evaluating the chemical reduction of AO7 utilizing riboflavin, representative of the heat stable redox-mediating moieties of common occurring flavin enzyme cofactors. Sulfate influenced dye reduction in accordance to biogenic sulfide formation from sulfate reduction. In assays lacking sulfur compounds, dye reduction only readily occurred in the presence of living granular sludge, demonstrating the importance of enzymatic mechanisms. Both chemical and biological mechanisms of dye reduction were greatly stimulated by the addition of the redox-mediating compound, anthraquinone-disulfonate. Based on an analysis of the kinetics and demonstration in lab-scale upward-flow anaerobic sludge bed reactors, the relative importance of chemical dye reduction mechanisms in high rate anaerobic bioreactors was shown to be small due to the high biomass levels in the reactors.  相似文献

The progression of anchorage-dependent epithelial cells to anchorage-independent growth represents a critical hallmark of malignant transformation. Using an in vitro model of human papillomavirus (HPV)-induced transformation, we previously showed that acquisition of anchorage-independent growth is associated with marked (epi)genetic changes, including altered expression of microRNAs. However, the laborious nature of the conventional growth method in soft agar to measure this phenotype hampers a high-throughput analysis. We developed alternative functional screening methods using 96- and 384-well ultra-low attachment plates to systematically investigate microRNAs regulating anchorage-independent growth. SiHa cervical cancer cells were transfected with a microRNA mimic library (n = 2019) and evaluated for cell viability. We identified 84 microRNAs that consistently suppressed growth in three independent experiments. Further validation in three cell lines and comparison of growth in adherent and ultra-low attachment plates yielded 40 microRNAs that specifically reduced anchorage-independent growth. In conclusion, ultra-low attachment plates are a promising alternative for soft-agar assays to study anchorage-independent growth and are suitable for high-throughput functional screening. Anchorage independence suppressing microRNAs identified through our screen were successfully validated in three cell lines. These microRNAs may provide specific biomarkers for detecting and treating HPV-induced precancerous lesions progressing to invasive cancer, the most critical stage during cervical cancer development.  相似文献
