Samples of chicken meat from spent laying hens were obtained by four different methods: (1) manual deboning of whole carcasses; (2) manual deboning of skinned carcasses; (3) mechanical deboning of whole carcasses; and (4) mechanical deboning of skinned carcasses. The meat was packaged, frozen and stored at ?18 °C for up to 3 months. Functional properties studied were pH, emulsifying capacity (EC) and water‐holding capacity (WHC) and, in addition, pigment concentration was investigated. Sensory properties included aroma, colour, texture and overall acceptability, as judged by a trained panel using a nine‐point hedonic scale. Removal of the skin prior to deboning by either method gave significantly higher values (P < 0.05) for EC, while WHC values were not significantly different among the experimental treatments, despite higher pH values for mechanically‐deboned meat, with and without skin. Pigment concentrations were highest in meat from Treatment 4 and lowest for Treatment 1, but values for both methods of deboning were affected by the presence of skin. There were no significant differences between treatments for any of the sensory properties studied. Changes occurring in the meat during frozen storage are described.  相似文献

Recent legislation in several states has called for the removal of methyl tert-butyl ether (MTBE) from gasoline. In order to comply with Federal Clean Air Act requirements for carbon monoxide and ozone attainment, ethanol is being considered as a replacement for MTBE. The objective of this study is to evaluate the potential impact of ethanol on benzene plume lengths in subsurface environments following accidental spills of ethanol-blended gasoline. Two types of studies were conducted here. First, laboratory studies were performed using a pure culture indigenous to a gasoline-contaminated aquifer to evaluate the effect of ethanol on the rate of benzene biodegradation under aerobic conditions. Results from microbial studies showed that the biodegradation of 25 mg/L benzene was severely inhibited in the presence of 25 mg/L ethanol. While the enzymes responsible for benzene biodegradation by the culture were inducible, ethanol degradation appeared to be constitutive. Second, a two-dimensional model was developed to quantify the impact of ethanol on benzene plume lengths using weighted-average aerobic and anaerobic biodegradation rates for benzene in the presence and absence of ethanol. Model simulations indicated that benzene plume lengths are likely to increase by 16–34% in the presence of ethanol.  相似文献

Wettability alteration mechanism is still believed to be the main contributor to low salinity water injection (LSWI) effect on oil recovery from carbonate rocks. The authors investigated the main mechanism and contributor to the LSWI effect on oil recovery from carbonate rocks through history matching recently published corefloods using the UTCHEM simulator. The history match results suggest that wettability alteration has negligible effect on water relative permeability, which could be related to the double electric layer expansion mechanism. This paper provides a basis to develop a LSWI model to predict oil recovery from carbonate rocks.  相似文献

NUMB is an endocytic adaptor protein that contains four isoforms (p65, p66, p71 and p72) due to alternative splicing regulation. Here, we show that NUMB exon12 (E12)-skipping isoforms p65/p66 promote epithelial to mesenchymal transition (EMT) and cancer cell migration in vitro, and facilitate cancer metastasis in mice, whereas E12-included p71/p72 isoforms attenuate these effects. Mechanistically, p65/p66 isoforms significantly increase the sorting of Notch1 through early endosomes (EEs) for enhanced Notch1 activity. In contrast, p71/p72 isoforms act as negative regulators of Notch1 by ubiquitylating the Notch1 intracellular domain (N1ICD) and promoting its degradation. Moreover, we observed that the interaction between N1ICD and SMAD3 is important for their own stabilization, and for NUMB-mediated EMT response and cell migration. Either N1ICD or SMAD3 overexpression could significantly recuse the migration reduction seen in the p65/p66 knockdown, and Notch1 or SMAD3 knockdown rescued the migration advantage seen in the overexpression of p66. Taken all together, our study provides mechanistic insights into the opposite regulation of Notch1-SMAD3 crosstalk by NUMB isoforms and identifies them as critical regulators of EMT and cancer cell migration.  相似文献

RNA molecules can adopt specific RNA triplex structures to execute critical biological functions. Human adenoviruses (HAdVs) are abundant pathogens encoding the essential, noncoding virus-associated RNA I (VA RNAI). Here, we employ a triplex-specific probing assay, based on the intercalating and cleaving agent benzoquinoquinoxaline 1, 10-phenanthroline (BQQ–OP), to unravel a potential RNA triplex formation in VA RNAI. The BQQ–OP cleavage of the pathogenic HAdV type 4 (HAdV-4) VA RNAI indicates that a potential triplex is formed involving the highly conserved stem 4 of the central domain and side stem 7. Further, the integrity of the HAdV-4 VA RNAI side stem 7 contributes to a potential triplex formation in vitro and virus growth in vivo. Collectively, we propose that the HAdV-4 VA RNAI can potentially form a biologically relevant triplex structure.  相似文献

Expansion of GAA triplet repeats in intron 1 of the FXN gene reduces frataxin expression and causes Friedreich's ataxia. (GAA)n repeats form non‐B‐DNA structures, including triple helix H‐DNA and higher‐order structures (sticky DNA). In the proposed mechanisms of frataxin gene silencing, central unanswered questions involve the characterization of non‐B‐DNA structure(s) that are strongly suggested to play a role in frataxin expression. Here we examined (GAA)n binding by triplex‐stabilizing benzoquinoquinoxaline (BQQ) and the corresponding triplex‐DNA‐cleaving BQQ‐1,10‐phenanthroline (BQQ‐OP) compounds. We also examined the ability of these compounds to act as structural probes for H‐DNA formation within higher‐order structures at pathological frataxin sequences in plasmids. DNA‐complex‐formation analyses with a gel‐mobility‐shift assay and sequence‐specific probing of H‐DNA‐forming (GAA)n sequences by single‐strand oligonucleotides and triplex‐directed cleavage demonstrated that a parallel pyrimidine (rather than purine) triplex is the more stable motif formed at (GAA)n repeats under physiologically relevant conditions.  相似文献

Low-salinity water injection is an emerging improved oil recovery technique due to its simplicity compared to other water based improved oil recovery methods. In this study, the wettability alteration option in the authors’ in-house simulator is used to history match and provide some insights in different seawater dilution cycles based on recently published corefloods. Two newly proposed methodologies to model dilution cycles are employed. The authors successfully modeled the experiments enhancing the wettability alteration model in the simulator using two different scaling factors. The study also revealed that the process is more sensitive to oil relative permeability compared to that of the water phase. A linear interpolation model for residual oil saturation (Sor) was proposed.  相似文献
