Ovarian steroids such as estrogen and progesterone have been reported to influence knee laxity. The effect of testosterone, however, remains unknown. This study investigated the effect of testosterone on the knee range of motion (ROM) and the molecular mechanisms that might involve changes in the expression of relaxin receptor isoforms, Rxfp1 and Rxfp2 in the patella tendon and lateral collateral ligament of the female rat knee. Ovariectomized adult female Wistar rats received three days treatment with peanut oil (control), testosterone (125 and 250 μg/kg) and testosterone (125 and 250 μg/kg) plus flutamide, an androgen receptor blocker or finasteride, a 5α-reductase inhibitor. Duplicate groups received similar treatment however in the presence of relaxin (25 ng/kg). A day after the last drug injection, knee passive ROM was measured by using a digital miniature goniometer. Both tendon and ligament were harvested and then analysed for protein and mRNA expression for Rxfp1 and Rxfp2 respectively. Knee passive ROM, Rxfp1 and Rxfp2 expression were significantly reduced following treatment with testosterone. Flutamide or finasteride administration antagonized the testosterone effect. Concomitant administration of testosterone and relaxin did not result in a significant change in knee ROM as compared to testosterone only treatment; however this was significantly increased following flutamide or finasteride addition. Testosterone effect on knee passive ROM is likely mediated via dihydro-testosterone (DHT), and involves downregulation of Rxfp1 and Rxfp2 expression, which may provide the mechanism underlying testosterone-induced decrease in female knee laxity.  相似文献

Highly porous activated carbon (HPAC) was used as carrier matrix for immobilization of acid protease (AP). Immobilization of acid protease on mesoporous activated carbon (AP-HPAC) performs as best enzyme carrier. At pH 6.0, 250 mg acid protease g−1 HPAC was immobilized. The optimum temperature for both free and immobilized AP activities were 50 °C. After incubation at 50 °C, the immobilized AP maintained about 50% of its initial activity, while the free enzyme was completely inactivated. When testing the reusability of AP-HPAC combination immobilized system, a significant catalytic efficiency was maintained along more than five consecutive reaction cycles. The highly porous nature of the carbon permits significant higher loadings of enzyme, which results in a higher enzyme-support strength and increased stability. The changes in the AP, HPAC and AP-HPAC were confirmed by Fourier Transform Infrared spectroscopy (FT-IR). Furthermore, scanning electron microscopy (SEM) allowed us to observe that the morphology of the surface of HPAC and the AP-HPAC.  相似文献

The high concentration of trivalent chromium along with organic/inorganic compounds in tannery sludge causes severe ground water contamination in the case of land disposal and chronic air pollution during incineration. In the present investigation, the sludge was subjected to flow-through column test to evaluate the concentration of leachable organics (tannin, COD and TOC) and heavy metal ions (Cr(3+), Fe(2+)) present in it. The dried sludge was incinerated at 800 degrees C in an incinerator under starved oxygen supply (starved-air combustion) to prevent the conversion of Cr(3+) to Cr(6+). The efficiency of starved air combustion was studied under different loading rates of sludge. The calcined sludge was solidified/stabilized using fly ash and Portland cement/gypsum. The solidified bricks were tested for unconfined compressive strength and heavy metal leaching. Unconfined compressive strength of the blocks was in the range of 83-156 kg/cm(2). The stabilization of chromium (III) in the cement gel matrix was confirmed with scanning electron microscopy (SEM) and X-ray energy dispersive spectroscopy (EDX). Leachability studies on solidified bricks were carried out to determine the metal fixation and dissolved organic (as COD) concentration in the leachate.  相似文献

Strontium added NiAl(2)O(4) composites prepared by sol-gel technique was utilized for the detection of methanol vapors. X-ray diffraction, scanning electron microscopy (SEM), FT-IR spectroscopy and nitrogen adsorption/desorption isotherm at 77K was employed respectively to identify the structural phases, surface morphology, vibrational stretching frequencies and BET surface area of the composites. The composites were prepared with the molar ratios of Ni:Sr as (1.0:0.0, 0.8:0.2, 0.6:0.4, 0.4:0.6, 0.2:0.8, 0.0:1.0) keeping the aluminum molar ratio as constant for all the compositions and were labeled as NiSA1, NiSA2, NiSA3, NiSA4, NiSA5 and NiSA6, respectively. The samples sintered at 900 degrees C for 5 h were subjected to dc resistance measurements in the temperature range of 30-250 degrees C to study the methanol vapor detection characteristics. The results revealed that the sensitivity in detecting methanol vapor increased with increase in temperature up to 175 degrees C for the composites NiSA1 and NiSA6 while for the other composites up to 150 degrees C and thereafter decreased. The sensitivity increased with increase in methanol concentration from 100 to 5000 ppm at 150 degrees C. Among the different composites NiSA5 showed the best sensitivity to methanol detection at an operating temperature of 150 degrees C.  相似文献

Seaweed extracts are considered effective therapeutic alternatives to synthetic anticancer, antioxidant, and antimicrobial agents, owing to their availability, low cost, greater efficacy, eco-friendliness, and non-toxic nature. Since the bioactive constituents of seaweed, in particular, phytosterols, possess plenty of medicinal benefits over other conventional pharmaceutical agents, they have been extensively evaluated for many years. Fortunately, recent advances in phytosterol-based research have begun to unravel the evidence concerning these important processes and to endow the field with the understanding and identification of the potential contributions of seaweed-steroidal molecules that can be used as chemotherapeutic drugs. Despite the myriad of research interests in phytosterols, there is an immense need to fill the void with an up-to-date literature survey elucidating their biosynthesis, pharmacological effects, and other biomedical applications. Hence, in the present review, we summarize studies dealing with several types of seaweed to provide a comprehensive overview of the structural determination of several phytosterol molecules, their properties, biosynthetic pathways, and mechanisms of action, along with their health benefits, which could significantly contribute to the development of novel drugs and functional foods.  相似文献

In this present investigation, physical and chemical characteristics of salt-laden solid waste (SLSW) generated from leather industry during soaking process of raw skins/hides and reverse osmosis reject stream were determined. The disposal of SLSW onto landfill was banned due to the possibility of ground water contamination by leachate, and its treatment process becomes tedious and cost intensive. The detailed characterisation of SLSW was evaluated for the development of effective methods for the treatment and/or disposal of SLSW. In the present study, sodium chloride was recovered to reduce the impact of the disposal of SLSW. The recovery of sodium chloride was achieved by selective precipitation using hydrogen chloride gas as a precipitating agent. The optimum conditions for the precipitation of sodium chloride were concentration of SLSW, 60 % (w/w); HCl gas purging, time 3 min (for a batch volume of 100 mL); pH, 8.0; and temperature, 30–40 °C. The scanning electron microscopy images of SLSW and the recovered sodium chloride were captured to observe the surface morphology. The recovered salt showed purity equivalent to that of standard reference salt. The recovered salt can be reused within leather sector or to the other process industries that meet their specification.  相似文献

The aim of this study was to investigate the association of single nucleotide polymorphisms (SNPs) and haplotypes of potassium voltage-gated channel, KQT-like subfamily, member 1 (KCNQ1) with type 2 diabetes (T2D) in Malaysian Chinese subjects. The KCNQ1 SNPs rs2237892, rs2283228 and rs2237895 were genotyped in 300 T2D patients and 230 control subjects without diabetes and metabolic syndrome. Two logistic regression models of analysis were applied, the first adjusted for age and gender while the second adjusted for age, gender and body mass index. The additive genetic analysis showed that adjusting for body mass index (BMI) even strengthened association of rs2237892, rs2283228 and rs2237895 with T2D (OR = 2.0, P = 5.1 × 10(-5); OR = 1.9, P = 5.2 × 10(-5); OR = 1.9, P = 7.8 × 10(-5), respectively). The haplotype TCA containing the allele of rs2237892 (T), rs2283228 (C) and rs2237895 (A) was highly protective against T2D (Second model; OR = 0.17, P = 3.7 × 10(-11)). The KCNQ1 rs2237892 (TT), and the protective haplotype (TCA) were associated with higher beta-cell function (HOMA-B) in normal subjects (P = 0.0002; 0.014, respectively). This study found that KCNQ1 SNPs was associated with T2D susceptibility in Malaysian Chinese subjects. In addition, certain KCNQ1 haplotypes were strongly associated with T2D.  相似文献
