Although both B7 and its counter-receptor CD28 are expressed in the thymus, the role of B7 in thymic selection is not clear. We investigated the role of B7 in intrathymic deletion of antigen-specific T cells using a TCR transgenic model specific for antigen ovalbumin (OVA) and H-2Ad. Intraperitoneal injection of OVA induced apoptosis of thymocytes and drastic reduction of thymocyte numbers. This was significantly inhibited by co-injection of CTLA-4-Ig which blocks B7 co-stimulation. Deletion of T cells in the thymus following i.p. injection of OVA was associated with T cell pre-activation as demonstrated by T cell proliferation and cytokine production. Injection of CTLA-4-Ig blocked all these activation events and rescued thymocytes from activation-induced cell death. These results demonstrate that B7 is required for the activation-induced cell death of MHC class II-restricted thymocytes in vivo.  相似文献

The nonlinear response of superconducting thin film bridges and transmission line sections at microwave frequencies has been investigated. A number of mixer prototypes have been designed and characterized. A model, based on analysis of experimental pulsedI-V characteristics, predicting results of mixing experiments, has been introduced and good agreement with the model has been obtained.  相似文献

A facile method of preparation of stabilized and functionalized nano‐ and microparticles of magnetite by successive application of oppositely charged polymers with a regular structure of macromolecular chains (chitosan and maleic acid copolymers) onto Fe3O4 core are developed. This approach makes it possible to create two types of magnetite interpolyelectrolyte shells, containing carboxylic or amino groups in outer layer of shell. Composition and magnetic properties of composite particles depend on nature of the copolymer of maleic acid, reaction conditions and size of obtained particles. The carboxylic groups of copolymer in outer layer of interpolyelectrolyte shell were converted into reactive anhydride groups by heating. Thermal treatment also leads to covalent crosslinking of shell and improves stability of composites both in acidic and alkaline media. Horseradish peroxidase was successfully immobilized onto covalently crosslinked and activated microparticles of Fe3O4 in aqueous medium without of condensing agents. The proposed reproducible and low‐cost technique does not use toxic reagents or solvents at all stages, including preparation of Fe3O4, formation, activation and crosslinking, of magnetite shell, modification of activated surface of composite particles. © 2013 Wiley Periodicals, Inc. There is an urgent need for identification of new prognostic markers and therapeutic targets for non-small cell lung cancer (NSCLC). In this study, we evaluated immune cells markers in 100 NSCLC specimens. Immunohistochemical analysis revealed no prognostic value for the markers studied, except CD163 and CD206. At the same time, macrophage markers iNOS and CHID1 were found to be expressed in tumor cells and associated with prognosis. We showed that high iNOS expression is a marker of favorable prognosis for squamous cell lung carcinoma (SCC), and NSCLC in general. Similarly, high CHID1 expression is a marker of good prognosis in adenocarcinoma and in NSCLC in general. Analysis of prognostic significance of a high CHID1/iNOS expression combination showed favorable prognosis with 20 months overall survival of patients from the low CHID1/iNOS expression group. For the first time, we demonstrated that CHID1 can be expressed by NSCLC cells and its high expression is a marker of good prognosis for adenocarcinoma and NSCLC in general. At the same time, high expression of iNOS in tumor cells is a marker of good prognosis in SCC. When used in combination, CHID1 and iNOS show a very good prognostic capacity for NSCLC. We suggest that in the case of lung cancer, tumor-associated macrophages are likely ineffective as a therapeutic target. At the same time, macrophage markers expressed by tumor cells may be considered as targets for anti-tumor therapy or, as in the case of CHID1, as potential anti-tumor agents.  相似文献

Conclusions The effect of metallizing conditions and Ni content on the structural and mechanical properties of Nitron fibre has been investigated.It has been shown that the metallization temperature-time regime leads to changes both in the amorphous and also in the crystalline regions of PAN fibre; these are connected with a compaction in polymer structure.The Ni content of the fibre significantly affects its structure and thermomechanical properties.
