Mitochondria are complex organelles that play critical roles in diverse aspects of cellular function. Heart disease and a number of other pathologies are associated with perturbations in the molecular machinery of the mitochondria. Therefore, comprehensive, unbiased examination of the mitochondrial proteome represents a powerful approach toward system-level insights into disease mechanisms. A crucial aspect in proteomics studies is design of bioanalytical strategies that maximize coverage of the complex repertoire of mitochondrial proteins. In this study, we evaluated the performance of gel-based and gel-free multidimensional platforms for profiling of the proteome in subsarcolemmal mitochondria harvested from rat heart. We compared three different multidimensional proteome fractionation platforms: polymeric reversed-phase liquid chromatography at high pH (PLRP), sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), and isoelectric focusing (IEF) separations combined with liquid chromatography-mass spectrometry/mass spectrometry (LC-MS/MS), and bioinformatics for protein identification. Across all three platforms, a total of 1043 proteins were identified. Among the three bioanalytical strategies, SDS-PAGE followed by LC-MS/MS provided the best coverage of the mitochondrial proteome. With this platform, 890 proteins with diverse physicochemical characteristics were identified; the mitochondrial protein panel encompassed proteins with various functional roles including bioenergetics, protein import, and mitochondrial fusion. Taken together, results of this study provide a large-scale view of the proteome in subsarcolemmal mitochondria from the rat heart, and aid in the selection of optimal bioanalytical platforms for differential protein expression profiling of mitochondria in health and disease.  相似文献

The so-called ''tribase'' acquaintance model of the agent's behavior is presented in this paper. This represents an extension of the twin-base model (Cao et al., 1997). Based on practical experience, the new model tries to cope with parallel processing, precedence constraints, and sparse resources. The idea of substituting the interagent negotiation processes by the periodical internal planning activity of the agents is stressed. A multiagent system, ProPlanT, as an application of the tribase model for the project-oriented production planning developed for TESLA TV company is described in detail. Three types of agents production planning agent (PPA), production management agent (PMA), and production agent (PA) are distinguished. The corresponding tribase models and potential role of metaagents are discussed.  相似文献

A compact, internally heated, catalytic reactor is demonstrated for the low-temperature oxidation of carbon monoxide. Carbon nanofibres were grown on carbon felt and used as a support material for Au/TiO2 catalysts. The carbon composite plays two roles; as a support material for the catalyst and for providing heat to the reaction by the Joule effect. The internal heating offers a stable reactor system with quick temperature response at relatively low energy input. Comparison between external and internal heating shows higher conversion of CO in the low-temperature range when using internal heating. The Au/TiO2 catalyst supported on the carbon-carbon composite shows good stability at 250°C.  相似文献

OBJECTIVE: To explore potential long term consequences of second trimester amniocentesis. DESIGN: Children exposed to second trimester amniocentesis have been followed prospectively from birth to school age and compared with children whose mothers declined the test. SETTING: The Hospital for Sick Children, Toronto. SAMPLE: Eighty-six children exposed to amniocentesis and 44 children whose mothers declined the test. INTERVENTIONS: Second trimester amniocentesis for the late maternal age indication. MAIN OUTCOME MEASURES: Standardised tests of intelligence, academic achievement, speech articulation, visual-perceptual-motor ability and fine motor coordination were administered. Parents reported on child behaviour using standardised measures of behaviour problems, social competence, hyperactivity and temperament. Physical growth was also measured, and parents reported on child health. RESULTS: Group differences were not observed. CONCLUSIONS: Second trimester amniocentesis does not appear to compromise child development, behaviour, growth or health.  相似文献

Metasurfaces are engineered nanostructured interfaces that extend the photonic behavior of natural materials, and they spur many breakthroughs in multiple fields, including quantum optics, optoelectronics, and biosensing. Recent advances in metasurface nanofabrication enable precise manipulation of light–matter interactions at subwavelength scales. However, current fabrication methods are costly and time-consuming and have a small active area with low reproducibility due to limitations in lithography, where sensing nanosized rare biotargets requires a wide active surface area for efficient binding and detection. Here, a plastic-templated tunable metasurface with a large active area and periodic metal–dielectric layers to excite plasmonic Fano resonance transitions providing multimodal and multiplex sensing of small biotargets, such as proteins and viruses, is introduced. The tunable Fano resonance feature of the metasurface is enabled via chemical etching steps to manage nanoperiodicity of the plastic template decorated with plasmonic layers and surrounding dielectric medium. This metasurface integrated with microfluidics further enhances the light–matter interactions over a wide sensing area, extending data collection from 3D to 4D by tracking real-time biomolecular binding events. Overall, this work resolves cost- and complexity-related large-scale fabrication challenges and improves multilayer sensitivity of detection in biosensing applications.  相似文献

This study aimed to better understand and quantify the influence of ventilation strategies on occupant‐related indoor air chemistry. The oxidation of human skin oil constituents was studied in a continuously ventilated climate chamber at two air exchange rates (1 h?1 and 3 h?1) and two initial ozone mixing ratios (30 and 60 ppb). Additional measurements were performed to investigate the effect of intermittent ventilation (“off” followed by “on”). Soiled t‐shirts were used to simulate the presence of occupants. A time‐of‐flight‐chemical ionization mass spectrometer (ToF‐CIMS) in positive mode using protonated water clusters was used to measure the oxygenated reaction products geranyl acetone, 6‐methyl‐5‐hepten‐2‐one (6‐MHO) and 4‐oxopentanal (4‐OPA). The measurement data were used in a series of mass balance models accounting for formation and removal processes. Reactions of ozone with squalene occurring on the surface of the t‐shirts are mass transport limited; ventilation rate has only a small effect on this surface chemistry. Ozone‐squalene reactions on the t‐shirts produced gas‐phase geranyl acetone, which was subsequently removed almost equally by ventilation and further reaction with ozone. About 70% of gas‐phase 6‐MHO was produced in surface reactions on the t‐shirts, the remainder in secondary gas‐phase reactions of ozone with geranyl acetone. 6‐MHO was primarily removed by ventilation, while further reaction with ozone was responsible for about a third of its removal. 4‐OPA was formed primarily on the surfaces of the shirts (~60%); gas‐phase reactions of ozone with geranyl acetone and 6‐MHO accounted for ~30% and ~10%, respectively. 4‐OPA was removed entirely by ventilation. The results from the intermittent ventilation scenarios showed delayed formation of the reaction products and lower product concentrations compared to continuous ventilation.  相似文献

Neither the European standard nor the US standard for classification of intermediate class filters comprises testing of filter performance with respect to ultrafine particles (UFPs) or particles of the most penetrating size (MPPS). This could turn out to be a major lack in classification standards since UFPs have been pointed out as a serious health hazard. In this study, fractional efficiencies of eight new full-scale bag filters and twenty-three new filter medium samples were determined. The influence of air velocity and aerosol type was investigated, and correlations between efficiencies for UFPs (EFUFPs), MPPS-sized particles (EFMPPS) and 0.4 μm-sized particles (EF0.4μm) were established. The tested bag filters were challenged by four aerosol types: a neutralized atomized oil aerosol, the same oil aerosol but non-neutralized, a non-neutralized thermally generated oil smoke, and a “natural” indoor aerosol. The tests were carried out at different air velocities through the filter medium, ranging between 0.08 m/s and 0.22 m/s. The relationships that were observed between EFUFPs, EFMPPS, and EF0.4μm appeared to be linear within the observed filtration efficiency ranges. These relationships were similar regardless of the test aerosol type used, but somewhat different for glass fiber filters than for charged synthetic filters. Generally, EFMPPS was 10–20% lower than EF0.4μm. The influence of air velocity variations on the size resolved efficiency was determined. The glass fiber filters showed practically the same fractional efficiencies regardless of whether the test aerosol was neutralized or not. However, the charged synthetic filters showed substantially lower efficiencies when tested with the non-neutralized aerosol compared to the case when the aerosol was neutralized.

We describe a comprehensive method for the analysis of complex proteomes, multiple gel-based technology (MGT), which combines in-gel IEF-LC-MS/MS and SDS-PAGE-LC-MS/MS strategies. MGT was applied to the analysis of the proteome of human pituitary tissue. A total of 1449 proteins were uniquely identified. To our knowledge, this is the most comprehensive proteomic analysis of human pituitary tissue to date. Categorization of the identified proteins revealed that MGT provides an excellent and largely unbiased access to proteins with diverse characteristics, including low-abundance proteins, membrane proteins, and proteins with extremes in pI and MW.  相似文献
