A total synthesis of the unnamed cembranolide 35 is described starting from enal XV , an intermediate derived from geraniol. Key transformations include asymmetric reduction of stannyl ketone 1 with (R)-(+)-BINAL-H to give the hydroxy stannane 2 of high ee, and cyclization of the derived MOM ether 6 to the cis-(Z) hydroxy enol ether 7 in high yield. Further elaboration of 7 to the natural product was achieved by OH-directed hydroalanation-iodination of the derived diol 17 , methylation of iodide 17 with methyl cuprate and selective protection, oxidation, lactonization, and α-methylenation. The resulting product was identified by comparison of 1H NMR, 13C NMR, IR, and optical rotation with an authentic sample. The synthesis defines the absolute configuration of the natural cembranolide.  相似文献

Kaempferol is a natural flavonoid. Previous studies have reported that kaempferol has anti‐proliferation activities and induces apoptosis in many cancer cell lines. However, there are no reports on human osteosarcoma. In this study, we investigate the anti‐cancer effects and molecular mechanisms of kaempferol in human osteosarcoma cells. Our results demonstrate that kaempferol significantly reduces cell viabilities of U‐2 OS, HOB and 143B cells, especially U‐2 OS cells in a dose‐dependent manner, but exerts low cytotoxicity on human fetal osteoblast progenitor hFOB cells. Comet assay, DAPI staining and DNA gel electrophoresis confirm the effects of DNA damage and apoptosis in U‐2 OS cells. Flow cytometry detects the increase of cytoplasmic Ca2+ levels and the decrease of mitochondria membrane potential. Western blotting and fluorogenic enzymatic assay show that kaempferol treatment influences the time‐dependent expression of proteins involved in the endoplasmic reticulum stress pathway and mitochondrial signaling pathway. In addition, pretreating cells with caspase inhibitors, BAPTA or calpeptin before exposure to kaempferol increases cell viabilities. The anti‐cancer effects of kaempferol in vivo are evaluated in BALB/cnu/nu mice inoculated with U‐2 OS cells, and the results indicate inhibition of tumor growth. In conclusion, kaempferol inhibits human osteosarcoma cells in vivo and in vitro.  相似文献

Several cyclic ammonium-based ionic liquids (ILs) with allyl substituent are synthesized, these allyl substituent ILs have high ionic conductivity (up to 4.72 mS cm−1 at 30 °C) and wide electrochemical window of 5 V. The electrochemical behaviors of two organometallic redox couples Fc/Fc+ (ferrocene/ferrocenium) and Cc/Cc+ (cobaltocene/cobaltocenium) have been studied in these ILs, the calculated Stokes–Einstein product ( T−1) of ferrocene in ILs is larger than that of cobaltocenium in ILs. The self-diffusion coefficients of cation and anion in these ILs are studied using pulsed gradient spin-echo NMR technique. There are very few reports where electrochemically derived diffusion coefficients and self diffusion coefficients are available for comparison, so a new key concept in electrochemistry could be developed in this paper.  相似文献

Complex dielectric response measurements have been made on a C60/C70 mixture pellet. The frequency and temperature ranges cover from 20 Hz to 1 MHz and from 30 to 300 K. Results show that the real part of the dielectric constant is 5 with a weak thermally activated polarization contribution. This thermal polarization is believed to be related to reorientation of C60 molecules recently observed by many techniques such as NMR, sound velocity, thermal conductivity, and others. Our data yield relaxation frequencies for reorientation, indicating that the presence of an additional electric dipole moment of the orientationally ordered C60 molecules at low temperature contributes 0.2–0.3 to the static dielectric constant.We wish to thank William Jenks and Chris Rey for their assistance and advice in performing the measurements. Anthraquinone compounds have been shown to induce apoptosis in different cancer cell types. Effects of chrysophanol, an anthraquinone compound, on cancer cell death have not been well studied. The goal of this study was to examine if chrysophanol had cytotoxic effects and if such effects involved apoptosis or necrosis in J5 human liver cancer cells. Chrysophanol induced necrosis in J5 cells in a dose‐ and time‐dependent manner. Non‐apoptotic cell death was induced by chrysophanol in J5 cells and was characterized by caspase independence, delayed externalization of phosphatidylserine and plasma membrane disruption. Blockage of apoptotic induction by a general caspase inhibitor (z‐VAD‐fmk) failed to protect cells against chrysophanol‐induced cell death. The levels of reactive oxygen species production and loss of mitochondrial membrane potential (ΔΨm) were also determined to assess the effects of chrysophanol. However, reductions in adenosine triphosphate levels and increases in lactate dehydrogenase activity indicated that chrysophanol stimulated necrotic cell death. In summary, human liver cancer cells treated with chrysophanol exhibited a cellular pattern associated with necrosis and not apoptosis.  相似文献

Several hydroxyl- and ether-functionalized binary task specific ionic liquids (ILs) are prepared, ether-functionalized ILs exhibit higher conductivity and lower viscosity than those of hydroxyl-functionalized ILs, whereas hydroxyl-functionalized ILs show wider potential window than those of ether-functionalized ILs. The correlation between ionic conductivity and viscosity is based on the classical Walden rule; a relatively large deviation of the plots from the ideal Walden line is observed for the ILs without considering the ion size, whereas the deviation decreases significantly when the adjusted Walden plot is adopted. The α values of ILs calculated from the slopes of the Walden plots are compared to those calculated from the ratio of activation energies for viscosity and molar conductivity (Ea,Λ/Ea,η). There are very few reports where electrochemically derived activation energy from conductivity and voltammetric characterization are available for comparison, so a key concept of activation energy in electrochemistry could be developed in this paper.  相似文献

BaTiO3/polyaniline and BaFe12O19/polyaniline composites were synthesized by in situ polymerization and introduced into epoxy resin and polyethylene to be microwave and infrared absorber. The spectroscopic characterizations of the formation processes of BaTiO3/polyaniline and BaFe12O19/polyaniline composites were examined using Fourier transform infrared, ultraviolet–visible spectrophotometer, X-ray diffraction, scanning electron microscopy, transmission electron microscopy and electron spin resonance. Microwave absorbing properties were investigated by measuring reflection loss in the 2–18 and 18–40 GHz microwave frequency range using the free space method. The thermal extinction measurements in the 3–5 and 8–12 μm were done to evaluate the shielding affectivity of infrared. The results showed that the BaTiO3/polyaniline and BaFe12O19/polyaniline composites have good compatible dielectric and magnetic properties and hence the microwave absorbency show broad frequencies absorbing properties. Moreover, the infrared thermal image testing that the detecting ability of infrared thermal imaging was decreased when the BaFe12O19 and BaTiO3 was coating with polyaniline.  相似文献

Epoxy–melamine hybrid resins were obtained from in situ polymerization of siliconized (SE500) and phosphorylated (PE690) epoxy resins with hexakis(methoxymethyl)melamine (HMMM). The hybrid resins having HMMM contents less than 15 wt % exhibited high transparency and homogeneity. The compatibilities between SE500 and melamine as well as that between PE690 and melamine were poor than the compatibility between general bisphenol‐A epoxy and melamine. Incorporation of HMMM altered the degradation mechanisms and enhanced the thermal stability of the epoxy resins, especially for PE690 based resins. Excellent flame retardant property was observed with the hybrid resins because of the Si? N and P? N synergisms of flame retardation. © 2006 Wiley Periodicals, Inc.
