The mechanism of gas permeation in Nafion membranes for polymer electrolyte fuel cells has been investigated from the viewpoint of free volume. Three different samples, a membrane with ionic exchange capacity (IEC) = 0.92 meq/g, and recast samples with IEC = 0.92 and 1.00 meq/g were used after drying. Free volume was quantified using the positron annihilation lifetime (PAL) technique and gas permeabilities were measured for O2 and H2 as functions of temperature and relative humidity. Good linear correlations between the logarithm of the permeabilities at different temperatures and reciprocal free volume indicate that gas permeation in dry Nafion is governed by the free volume. Nevertheless permeabilities are much smaller than the corresponding flexible chain polymer with a similar free volume size due to stiff chains of the perfluoroethylene backbone. In highly hydrated Nafion above 60% relative humidity, where the O2 permeability varies oppositely to the free volume, gas permeation proved to be controlled by the gradual increase in overall flexibility of the Nafion–water system.  相似文献

A 32-year-old female was admitted due to splenomegaly and leukocytosis in September, 1993. The leukocyte count was 26,900/microliter with 29% monocytes (7,800/microliter). A diagnosis of the chronic phase of chronic myelomonocytic leukemia was made. On November 19, 1993, splenic arterial embolization was performed. After the embolization, the leukocyte count rapidly increased, and acute respiratory failure developed. The respiratory condition was improved by methylprednisolone (m-PRED) pulse therapy. Subsequently, the effectiveness of chemotherapy gradually decreased, and there was an increase in the leukocyte count. Respiratory failure developed again but was successfully treated with m-PRED pulse therapy in addition to aclarubicin. On July 4, 1995, splenectomy was performed. The leukocyte count rapidly increased, and acute respiratory failure again developed. She did not respond to m-PRED pulse therapy, but the respiratory condition was markedly improved by leukoplasmapheresis. The respiratory failure in this patient may be associated with capillary leak syndrome due to neutrophilia. In addition, stasis of increased monocytes in the pulmonary capillaries and their infiltration into the pulmonary parenchyma and alveoli was thought to have occurred.  相似文献

A phytoplankton model, which includes a carbon cycle combined with a constant current obtained by the computation of tidal currents, was applied to the coastal region (Hiroshima Bay, Japan). The transformation rate constants for the carbon cycle were experimentally evaluated.The seasonal variations of particulate and dissolved organic matter by the simulation model were reasonably consistent with the field data. Carbon concentration increased by primary production in the upper layer was approx. 10 times that augmented by loading of carbon from land at the shore in summer. Sinking plays an important role in the elimination of particulate organic matter. On the other hand, horizontal advection and turbulent diffusion are important factors for the diminution of dissolved organic matter.  相似文献

We have previously identified and cloned a human gene, D40, that is preferentially expressed in testis among normal organs, while it is widely expressed in various human tumor cell lines and primary tumors derived from different organs. In this report, we have examined the expression and localization of this protein in human testis with an antibody specific to D40 protein. In Western analyses, the anti-D40 antibody recognized a major band with a molecular mass of 300 kDa and a minor band of 250 kDa. These bands were not observed in the testis lysates from patients with Sertoli-cell-only syndrome and with Kleinfelter syndrome, who lack germ cells of the testis, indicating that D40 protein is expressed in the germ cells of normal testis. Immunohistochemical studies have revealed that D40 protein is highly expressed in spermatocytes and in the pre-acrosome of round spermatids. In the acrosome, D40 protein expression is observed not inside but outside the acrosome membrane. This is consistent with the finding that the amino-acid sequence at the amino terminal of the D40 protein lacks a hydrophobic signal peptide that is required for proteins to translocate to the membrane. Expression of D40 protein is observed in the acrosome of ejaculated spermatozoa as well, although the level is low compared with that in the pre-acrosome of spermatids. These results suggest that D40 protein plays important roles in spermatogenesis, especially in the formation and maintenance of the acrosome.  相似文献

We have investigated a new approach to efficiently find a novel inhibitor against a serine protease (i.e. an activated coagulation factor X, FXa) by using de novo design programs and the X-ray crystal structure of the target enzyme. FXa is a coagulant enzyme that generates thrombin (a serine protease) and participates in both intrinsic and extrinsic coagulation pathways. We adopted multiple copy simultaneous search (MCSS) and CAVEAT linker search techniques, which disclosed a novel FXa inhibitor (T01312) consisting of two binding moieties (the benzamidinyl and adamantyl groups) and a linker unit (the carboxybenzylamine group). The inhibitory activity of T01312 against FXa was determined to be a small K(i)-value of 48nM, which is two orders of magnitude smaller than that against thrombin. An X-ray crystal analysis of T01312 complexed with trypsin (an analogue of FXa) and docking studies of T01312 with trypsin and FXa showed that: (i) the benzamidinyl group is a predominant binding moiety in the anionic pocket (S1 site) with an asparatic acid residue; (ii) a hydrophobic pocket (S4 site) is the binding site of the adamantyl group; (iii) the carboxylate group of the linker contributes to the selectivity for FXa against thrombin. Thus, the combination of the knowledge of the X-ray crystal structure of the target molecule with MCSS and CAVEAT linker search techniques proved to be an effective hit-finding method that does not require the screening of huge compound libraries.  相似文献

The dynamics of filamentations in broad-area semiconductor lasers with short optical feedback were numerically studied. Regular pulse packages similar to those observed in narrow-stripe edge-emitting semiconductor lasers were found as the feedback reflectivity is increased. However, the regular pulse packages consisted of a periodic envelope of the external cavity frequency and fine pulse trains of periodic filaments, which is different from the regular pulse packages observed in narrow-stripe edge-emitting semiconductor lasers. From the analysis for spatio-temporal near-field patterns, we found periodic undulations of the spatial and temporal filament sizes as the external mirror reflectivity was changed. We also investigated filtered optical feedback, where a part of the emitted beam is fed back into the laser cavity. The time-averaged near-field pattern was found to have a strong dependence on the feedback level in the case of spatial filtered optical feedback.  相似文献

The relationships between morphology and proton conduction for Nafion membranes and hydrocarbon-type proton exchange membranes, namely, sulfonated poly(arylene ether ether ketone) (S-PEEK) and sulfonated poly(arylene ether sulfone) (S-PES), were investigated by small-angle X-ray scattering (SAXS) and atomic force microscopy (AFM). The direct simultaneous observation of surface morphology and active regions of proton conduction on membranes by combined high-resolution AFM phase imaging and an electrochemical technique at controlled humidity provided significant morphological information, particularly for the hydrocarbon-type membranes that exhibit few or no features on SAXS profiles. For the Nafion membranes, the active proton paths became denser and congregated with each other at over 60% RH, resulting in the formation of well-connected networks. For the hydrocarbon-type membranes, however, only the relatively small and dispersed proton paths were observed, which showed no significant change even as water content increased. We have demonstrated that the differences in microscopic morphology between the Nafion and hydrocarbon-type membranes are associated with the differences between their macroscopic proton conductivities.  相似文献

Reconstructions of the intrathoracic trachea in 24 dogs were done with the use of 50 mm long collagen-conjugated tracheal prostheses. Omental wrapping was also done in 14 of the dogs (omentopexy group) to evaluate the efficacy of this option in comparison with results in the other 10 dogs (control group). All 24 dogs had uneventful postoperative courses and were killed at 4 weeks or 3, 6, or 12 months after the operation. Better epithelialization and fewer complications, such as mesh exposure and luminal stenosis, were observed in the omentopexy group than in the control group. Angiography and analysis of regenerated blood vessels revealed that vessel ingrowth had started within 4 weeks and that vessel formation reached its maximal point within 6 to 12 months in the omentopexy group. In contrast, revascularization of the subepithelial region in the control group was poor even after 3 months, and vessel formation continued for as long as 12 months. The differences between the two groups were considered to be mainly a result of the speed of blood vessel ingrowth into the regenerated mucosa. We conclude that our prosthesis can be used safely for intrathoracic tracheal reconstruction and that omental wrapping is a useful supplementary method that reduces the occurrence of complications.  相似文献
