PURPOSE: The activity of Al3+, Ga3+, and Be2+ ions in the presence of NaF to directly activate G-proteins was investigated by their potentiative effect on forskolin (FSK)-activated adenylyl cyclase in rabbit ciliary process membranes and their effects on aqueous humor dynamics in vivo. METHODS: Adenylyl cyclase (AC) was determined by radiometric conversion of ATP to cAMP by the particulate fraction of rabbit ciliary processes. Intravitreal injections of sterile solutions of analytical grade salts were made into the center of the vitreous in a volume of 20 microliters. Intraocular pressure, aqueous humor flow, and uveoscleral outflow measurements were made by pneumatonometry, fluorophotometry, and fluorescein-dextran method, respectively. Outflow facility was determined by tonography in the intact eyes and by two-level constant pressure perfusion in cannulated eyes. RESULTS: Both Al3+ (EC50, 40 mumol/l) and Be2+ (EC50, 11 mumol/l) in the presence of 0.5-2 mM NaF activated the stimulatory G-protein Gs. Ga3+ was ineffective and did not antagonize the activation by Al3+. Intravitreal injections of Al3+ (1 mumol/eye) or Be2+ (0.5 or 1 mumol/eye) had no significant intraocular pressure (IOP) effect, nor did 1.5 or 3 mumol/eye of NaF, but when either cation was injected together with NaF, IOP decreased by up to 40% for up to 140 hr. At the time of maximum IOP effect (72 hr) aqueous humor flow determined by fluorophotometry was decreased in BeCl2+ NaF-treated eyes by 40% relative to BeCl2-treated eyes; however, tonographic facility of outflow was unaffected. Uveoscleral flow was also decreased by 38% in BeCl2+ NaF treated eyes. CONCLUSIONS: These findings support the hypothesis that Gs activation of ciliary process adenylyl cyclase decreases aqueous humor formation rate in rabbit eyes, and that activation of G-proteins mediates contraction of ciliary muscles causing a decrease of aqueous humor outflow via the uveoscleral route. The results suggest that G-proteins putatively involved in trabecular facility changes are less sensitive to activation by BeF3- than are other parameters of aqueous humor dynamics.  相似文献

Image cytometry is an important technique in affordable healthcare and cellular research. Some efforts toward establishing a personal, low-cost cytometer have been described in the literature. However, a self-assembled fluorescence microscope requires software for cytometric analysis. There are some open-source image-based software analysis applications available. However, for a quantitative analysis of images, software that can generate data comparable to those of previously evaluated cytometric analyses programs is required. Hence, the aim of this study is to compare results of a commercially available image cytometry program to data obtained using the open-source software CellProfiler (CP). Leukocytes and fluorescent bead images obtained using a Laser Scanning Cytometer were analyzed by CP and the results compared with those of conventional cytometric analyses' programs. Algorithms were developed enabling the analysis of leukocytes and beads by CP. CP provided similar results to those obtained by the cytometer software. Hallmark parameters, including cell count and fluorescence intensity, revealed a high correlation in the analysis of both programs. Therefore, CP is appropriate for cellular analysis on a self-assembled microscope, thereby enabling affordable cytometry.  相似文献

The ATWS transient “Loss of main feed water supply” in a generic four-loop PWR at the nominal power of 3750 MW was analyzed using the coupled code system DYN3D/ATHLET. A variation of the MOX-fuel-assembly portion in the core has an effect on the reactivity coefficients of the fuel temperature and the moderator density. These two parameters mainly influence the behaviour of the coolant pressure, which is safety-relevant. It has been demonstrated that the pressure maximum decreases with an increasing portion of MOX. For all core loadings considered, both primary-circuit mechanical integrity and sufficient core cooling are guaranteed.  相似文献

Measurements carried out in an original-size VVER-1000 mock-up (V-1000 facility, Kurchatov Institute, Moscow) were used for the validation of three-dimensional neutron-kinetic codes, designed for VVER safety calculations. The significant neutron flux tilt measured in the V-1000 core, which is caused only by radial-reflector asymmetries, was successfully modeled. A good agreement between calculated and measured steady-state powers has been achieved, for relative assembly powers and inner-assembly pin power distributions. Calculated effective multiplication factors exceed unity in all cases. The time behaviour of local powers, measured during two transients that were initiated by control rod moving in a slightly super-critical core, has been well simulated by the neutron-kinetic codes.  相似文献

Super-homogenisation (SPH) factors were generated by a modified version of the code DYN3D for PWR fuel assemblies in hot-zero-power states defined in the OECD MOX/UO2 Benchmark. SPH factors averaged for each pin-material type and factors for each individual pin position were produced. The application of the SPH factors improves the accuracy of DYN3D calculations, especially for configurations with control rods inserted.  相似文献

The angiotensin receptor/neprilysin inhibitor Sacubitril/Valsartan (Sac/Val) has been shown to be beneficial in patients suffering from heart failure with reduced ejection fraction (HFrEF). However, the impact of Sac/Val in patients presenting with heart failure with preserved ejection fraction (HFpEF) is not yet clearly resolved. The present study aimed to reveal the influence of the drug on the functionality of the myocardium, the skeletal muscle, and the vasculature in a rat model of HFpEF. Female obese ZSF-1 rats received Sac/Val as a daily oral gavage for 12 weeks. Left ventricle (LV) function was assessed every four weeks using echocardiography. Prior to organ removal, invasive hemodynamic measurements were performed in both ventricles. Vascular function of the carotid artery and skeletal muscle function were monitored. Sac/Val treatment reduced E/é ratios, left ventricular end diastolic pressure (LVEDP) and myocardial stiffness as well as myocardial fibrosis and heart weight compared to the obese control group. Sac/Val slightly improved endothelial function in the carotid artery but had no impact on skeletal muscle function. Our results demonstrate striking effects of Sac/Val on the myocardial structure and function in a rat model of HFpEF. While vasodilation was slightly improved, functionality of the skeletal muscle remained unaffected.  相似文献

Bifacial solar cells and modules are a promising approach to increase the energy output of photovoltaic systems, and therefore decrease levelized cost of electricity (LCOE). This work discusses the bifacial silicon solar cell concepts PERT (passivated emitter, rear totally diffused) and BOSCO (both sides collecting and contacted) in terms of expected module cost and LCOE based on in‐depth numerical device simulation and advanced cost modelling. As references, Al‐BSF (aluminium back‐surface field) and PERC (passivated emitter and rear) cells with local rear‐side contacts are considered. In order to exploit their bifacial potential, PERT structures (representing cells with single‐sided emitter) are shown to require bulk diffusion lengths of more than three times the cell thickness. For the BOSCO concept (representing cells with double‐sided emitter), diffusion lengths of half the cell thickness are sufficient to leverage its bifacial potential. In terms of nominal LCOE, BOSCO cells are shown to be cost‐competitive under monofacial operation compared with an 18% efficient (≙ pMPP = 18 mW/cm2) multicrystalline silicon (mc‐Si) Al‐BSF cell and a 19% mc‐Si PERC cell for maximum output power densities of pMPP ≥ 17.3 mW/cm2 and pMPP ≥ 18.1 mW/cm2, respectively. These values assume the use of $10/kg silicon feedstock for the BOSCO and $20/kg for the Al‐BSF and PERC cells. For the PERT cell, corresponding values are pMPP ≥ 21.7 mW/cm2 and pMPP ≥ 22.7 mW/cm2, respectively, assuming the current price offset (≈50%, at the time of October 2014) of n‐type Czochralski‐grown silicon (Cz‐Si) compared with mc‐Si wafers. The material price offset of n‐type to p‐type Cz‐Si wafers (≈15%, October 2014) currently accounts for approximately 1 mW/cm2, which correlates to a conversion efficiency difference of 1%abs for monofacial illumination with 1 sun. From p‐type mc‐Si to p‐type Cz‐Si (≈30% wafer price offset, October 2014), this offset is approximately 2.5 mW/cm2 for a PERT cell. When utilizing bifacial operation, these required maximum output power densities can be transformed into required minimum rear‐side illumination intensities for arbitrary front‐side efficiencies ηfront by means of the performed numerical simulations. For a BOSCO cell with ηfront = 18%, minimum rear‐side illumination intensities of ≤ 0.02 suns are required to match a 19% PERC cell in terms of nominal LCOE. For an n‐type Cz‐Si PERT cell with ηfront = 21%, corresponding values are ≤ 0.11 suns with 0.05 suns being the n‐type to p‐type material price offset.
