A protein-free microemulsion (LDE) with a lipid composition resembling that of low-density lipoprotein (LDL) was used in metabolic studies in rats to compare LDE with the native lipoprotein. LDE labeled with radioactive lipids was injected into the bloodstream of male Wistar rats, and plasma kinetics of the labeled lipids were followed on plasma samples collected at regular intervals for 12 h after injection. The 24-h LDE uptake by different tissues was also measured in tissue samples excised after the animals had been sacrificed. We found that LDE plasma kinetics were similar to those described for native LDL [fractional clearance rate (FCR) of cholesteryl ester, 0.42±0.11 h−1]. The major site for LDE uptake was the liver, and the tissue distribution of the LDE injected radioactivity was as one would expect for LDL. To test whether LDE was taken up by the specific LDL receptors, the LDE emulsion was injected into rats treated with 17α-ethinylestradiol, which is known to increase the activity of these receptors; as expected, removal of LDE from the bloodstream increased (FCR=0.90±0.35 h−1). On the other hand, saturation of the receptors that remove remnants by prior infusion of massive amounts of lymph chylomicrons did not change LDE plasma kinetics. These results indicate that LDE is cleared from plasma by B,E receptors and not by the E receptors that remove remnants. Incorporation of free cholesterol into LDE increased LDE plasma clearance. Incubation studies also showed that LDE incorporates a variety of apolipoproteins, including apo E, a ligand for recognition of lipoproteins by specific receptors. Our data suggest that LDE can be a useful tool to test LDL metabolism and B,E receptor function.  相似文献

In this paper, we present OpenCOPI (Open COntext Platform Integration), a Service-Oriented Architecture-based middleware platform that supports the integration of services provided by distinct sources, ranging from services offered by simple systems to more complex services provided by context-provision middleware. OpenCOPI offers selection and composition mechanisms to, respectively, select and compose services provided by different sources, considering applications of both Quality of Service and Quality of Context requirements. It also offers an adaptation mechanism that enables to adapt the application execution due to service failures, service quality fluctuation and user mobility. OpenCOPI allows the definition of applications in a higher abstraction level by the specification of a semantic workflow that contains abstract activities. This paper illustrates the use of OpenCOPI in an application from the Gas & Oil Industry and it also shows the evaluation of the main mechanisms of OpenCOPI: the service selection, composition, adaptation and workflow execution.  相似文献

This work describes the preparation, the swelling properties and the potassium diclofenac (KDF) release profile of hydrogels of gum arabic (GA), N′,N′‐dimethylacrylamide, and methacrylic acid. In order to convert GA into a hydrogel, the polysaccharide was vinyl‐modified with glycidyl methacrylate. The hydrogels showed pH‐responsive swelling changes, which were more expressive in the basic environment. Release data of KDF were adjusted to a diffusion‐based kinetic model that provides an important insight on affinity of the drug for hydrogel and solvent, which may be the leading parameter for release of guest molecules from polymers. The KDF release from the hydrogels into simulated intestinal fluid decreases when the amount of modified GA increases. This was demonstrated to be due to the higher affinity of KDF for GA‐richer hydrogel, which makes the anti‐inflammatory release less favorable. Fluorescence depolarization and quenching measurements of N‐vinyl carbazole/vinyl tert‐butyl‐benzoate copolymers in dilute solution of several fluid solvents and in a rigid poly(methyl methacrylate) matrix were performed to study the effect of copolymer composition on the intramolecular energy transfer (IET) between carbazole groups (Cz) along the polymer chain. IET efficiency, as the amount of Cz? Cz intramolecular excimer formation, monotonically increases with Cz content up to a number average sequence length of carbazole of ca 12 (Cz molar content ≥ 90%). After this value, IET efficiency remains almost constant. Molecular dynamics simulations on isotactic and syndiotactic copolymer fragments of several monomer compositions were used for obtaining different parameters related to the efficiency for the IET process between Cz units along the polymer chain. Measurements of degradation in frying oils are mainly based on physico-chemical properties. Total polar compounds (TPC) and free fatty acids (FFA) content in frying oils are used as a guide for discarding used oils. The purpose of this study was to evaluate the efficacy of a sensory method in detecting degradation in soybean oils used in potato chips deep frying. The sensory evaluation of oil samples was determined by a trained panel; after rigorous selection and training steps. Free fatty acid, TPC and Rancimat induction period (IP) were quantified in the same samples. The proposed sensory method was sensitive to small differences in rancidity. The selected and trained sensory panel discarded oil samples with 0.175% FFA as oleic acid, 18.92% TPC, and 0.20 h IP. According to the results achieved in this research sensorial trained panel response is sensitive and accurate in refusing deteriorated frying oils. Besides this, soybean oil can be used for deep frying procedures and safely discarded according to the panel response, although presenting up to 7% linolenic acid.  相似文献

Artemisia verlotorum, popularly ‘infalivina’, is a medicinal herb used for respiratory, circulatory and digestive treatments. Antiproliferative and genotoxic effects of A. verlotorum aqueous extracts in three different concentrations (6, 32 and 48 g/L) were evaluated on the Allium cepa cell cycle. Results clearly showed a significant difference using the χ2 test (p = 0.05) among treatments with a decrease in the mitotic index as the concentration of the extract increased. Extracts of A. verlotorum proved to be genotoxic and showed antiproliferative potential on the A. cepa cell cycle. This study suggests the use of infusions of A. verloturum for medicinal purposes only after selecting suitable concentrations.  相似文献

The dynamic behavior of a droplet on a solid surface is simulated by the lattice Boltzmann method (LBM) for two-phase fluids with large density differences; the wetting boundary condition on solid walls is incorporated in this simulation. By using the method, the dynamic behavior of a droplet impinging on a horizontal wall is investigated in terms of various Weber numbers. The dynamic contact angle, the contact line velocity, and the wet length are calculated, and found to be in good agreement with available experimental data. In addition, the method is applied to simulations of the collision of a falling droplet with a stationary droplet on a solid surface. The behavior of the droplets and the mixing process during their collision are simulated in terms of various impact velocities and several static contact angles on the solid surface. It is seen that mixing occurs around the rim of the coalescent droplet due to the circular flows. Also, the relationship between the mixing rate of the primary coalescent droplet and Weber number is investigated.  相似文献

This paper presents a mechanism for dynamic reconfiguration of component-based applications and its fault tolerance strategy. The mechanism, named generic connector, allows composing a component-based application as a set of services with no previous knowledge about the specific components that will provide some services. The components will be selected at runtime. The objective of this work is to offer a mechanism that tries to satisfy every invocation under its responsibility and makes failures as transparent as possible. It is an important tool to compose applications through the reuse of existing components because it frees the programmer from the task of searching components in order to use them and, also, from solving some types of failures.  相似文献
