We investigated the effect of platelet-activating factor (PAF) and of the PAF specific antagonist CV-6209 on plasma lipid metabolism, and particularly on post-heparin plasma lipolytic activity in male Wistar rats. Lipoprotein lipase (LPL) activity was enhanced by intravenous injection of PAF before intravenous injection of heparin when the PAF dose was low (0.2 μg/kg). PAF activated hepatic triacylglycerol lipase (HTGL) activity dose-dependently. Plasma triacylglycerols (TG) significantly decreased with the activation of LPL and/or HTGL. Plasma total cholesterol (TC) and phospholipid (PL) levels decreased at a low dose of PAF (0.2 μg/kg), but increased when higher doses were used. The PAF antagonist CV-6209 partially reversed the PAF induced effects on HTGL, TC and PL. α-Tocopherol was reacted with methyl linoleateperoxyl radicals at 37°C. The peroxyl radicals were generated by the reaction of methyl linoleate with a free radical initiator, 2,2′-azobis(2,4-dimethylvaleronitrile). The primary products of α-tocopherol with methyl linoleate-peroxyl radicals were isolated by reversephase and normal-phase high performance liquid chromatography (HPLC), and their structures were characterized by ultraviolet (UV), infrared (IR),1H and13C nuclear magnetic resonance (NMR) and mass spectrometry (MS). There were four stereoisomers of methyl 13-(8a-peroxy-α-tocopherone)-9(Z),11(E)-octadecadienoate and four stereoisomers of methyl9-(8a-peroxy-α-tocopherone)-10(E),12(Z)-octadecadienoate.  相似文献

The optimum conditions for the preparation of protein concentrate from cowpea were determined using response surface methodology. A central composite rotatable design, consisting of eight experimental points and five replications at the centre point, was used to investigate the effects of two independent variables, namely pH and NaCl concentration on four responses: protein content (%,), protein yield (%), reactive lysine (g/16 g N) and zinc content (mg/100 g). A second-order polynomial model was used for predicting the responses. Regression analysis indicated that more than 80% of the variation was explained by the fitted models. Experimental results showed that under optimum conditions (pH and NaCl concentration of 9.91 and 0.15 M, respectively) the protein content was 84%, protein yield 87%, reactive lysine 1.175 g/16 g N and zinc content 7.75 mg/100 g. These results were in agreement with those predicted, hence indicating the suitability of the model used.  相似文献

Effect of cabinet-drying, vacuum-drying, and freeze-drying methods on the surface hydrophobicity, secondary structure, emulsifying, and foaming properties of protein concentrates prepared from different cultivars of cowpea and Bambara bean was investigated. The vacuum-drying method reduced hydrophobicity, while freeze-dried concentrates presented high hydrophobicity. The concentrates prepared by freeze drying presented more β-sheet (40–43%) and less β-turn (19–24%) structures. Bambara bean protein concentrates prepared by freeze-drying presented higher emulsifying activity (56–59%) compared to those by vacuum-drying and cabinet-drying, while emulsifying activity varied significantly among cultivars of cowpea (46–61%). Protein concentrates prepared by cabinet-drying showed the highest foaming ability.  相似文献

α-Tocopherol wss reacted with an alkylperoxyl radical at 37°C in ethanol. 2,2′-Azobis(2,4-dimethylvaleronitrile) was used to generate the alkylperoxyl radicals. They were 8aS-hydroperoxy-α-tocopherone, 8aR-hydroperoxy-α-tocopherone, a mixure of 7,8-epoxy-8aS-ethoxy-α-tocopherone, and 7,8-epoxy-8aR-ethoxy-α-tocopherone, 8aS-(1R-cyano-1,3-dimethyl)butylperoxy-α-tocopherone, 8aS-(1S-cyano-1,3-dimethyl)butylperoxy-α-tocopherone, a mixture of 8aR-(1S-cyano-1,3-dimethyl)butylperoxy-α-tocopherone and 8aR-(1S-cyano-1,3-dimethyl)butylperoxy-α-tocopherone, 4aR,5S-epoxy-8aS-ethoxy-α-tocopherone, 4aS,5R-epoxy-8aS-ethoxy-α-tocopherone and 4aR,5S-epoxy-8aR-ethoxy-α-tocopherone.  相似文献

Based on urinary excretion studies the secretion of the cortisol derivatives, 18-oxocortisol and 18-hydroxycortisol are believed to be regulated by ACTH and to a lesser degree by the renin-angiotensin system. Plasma concentrations of 18-oxocortisol and 18-hydroxycortisol were measured during the simultaneous activation of the renin-angiotensin system and inhibition of ACTH secretion. Five healthy male subjects consuming a sodium diet ad libitum were studied. Blood was drawn at 0800 h after 1 h in the supine position. In the first set of experiments, the subjects remained in the supine position from 0800 to 1000 h with or without the oral administration of 2 mg dexamethasone at 0800 h. In the second set of experiments the subjects were placed in the upright position after drawing the 0800 h sample. The subjects were studied with and without dexamethasone administered at 0800 h. Blood was drawn again at 1000 h. Plasma levels of 18-oxocortisol, 18-hydroxycortisol, ACTH, plasma renin activity (PRA), cortisol, aldosterone and 18-hydroxycorticosterone were measured by radioimmunoassay. None of these parameters changed during the 2 h in the supine position. 18-Oxocortisol, 18-hydroxycortisol, aldosterone, 18-hydroxycorticosterone and PRA increased, but ACTH and cortisol did not change when the subjects were placed in the upright position. After dexamethasone administration, 18-oxocortisol, 18-hydroxycortisol, cortisol, aldosterone and 18-hydroxycorticosterone decreased in the supine position and no increase occurred in 18-oxocortisol, 18-hydroxycortisol and 18-hydroxycorticosterone in the upright position. PRA and aldosterone increased and ACTH and cortisol decreased in these subjects. 18-Oxocortisol and 18-hydroxycortisol were more dependent on ACTH regulation and less on the renin-angiotensin system than aldosterone.  相似文献
