As the cationic component of natural polymer polyionic complexes, polysaccharide chitosan or polypeptide polylysine, and as the anionic component, polysaccharide carboxymethyl cellulose (=CMC) or polypeptide poly(glutamic acid) (PGA) were used. The degree of chitosan/CMC or polylysine/PGA complex film swelling was minimum, when the amounts of the cationic and anionic groups in these polymers were balanced at 1 : 1 ratio. On the other hand, the degree of chitosan/PGA film swelling was minimum at the ratio with excess of PGA which is a linear flexible chain. Comparing the enthalpies estimated by DSC analysis, it was suggested that the larger quantity of water in the form of nonfreezable bound water was absorbed in the complex films with polysaccharide which is a chain of ring structure. Further, the compressive moduli of swollen complex films were high, when the complex consisted of polysaccharide components.  相似文献

For the past few years, hybrid generation systems including solar panel and gas cogeneration have been used for residential houses. Solar panels can generate electronic power at daytime but not at night. But the power consumption of residential houses usually peaks in the evening. The gas engine cogeneration system can generate electronic power without such a restriction, and it also can generate heat power to warm up a house or to produce hot water. In this paper we propose a solar panel and gas engine cogeneration hybrid system with an energy storage device, combined by a DC bus. If a blackout occurs, the system still can supply electronic power for special house loads. We propose a control scheme for the system related to the charging level of the energy storage device and the voltage of the utility grid, which can be applied to both grid‐connected and standalone operation. A method for calculating the single‐step response of five‐phase hybrid stepping motors is obtained. The effects of winding connecting methods—(1) star‐bipolar SB, (2) pentagon PD, and (3) new pentagon NP—on the single‐step response are investigated. By application of the above calculation method, the continuous rotating characteristics are studied. It is found that the maximum slewing pulse rate of NP is higher than that of SB and PD connections, and that the variation of the rotor speed at high speed with the SB and PD connections is larger than that with the NP connection. Takashi Nakai and Toshinari Tanaka of the Forestry and Forest Products Research Institute in Japan present the results of tests, using the easy handling and reasonable cost technique of fundamental vibration frequency measurement, on larger than usual cross-sections. Modulus of elasticity measurements showed this method to have high possibilities for stress grading, overcoming the size limitations of existing commercial equipment at the saw mill or lumber yard. The four hydrogenase isozymes; hydrogenase 1 (Hyd-1), hydrogenase 2 (Hyd-2), hydrogenase 3 (Hyd-3) and hydrogenase 4 (Hyd-4) of Escherichia coli have been reported for their crucial functions in the hydrogen metabolism; however, their distinctive roles could not be completely understood. In this study, four ideal hydrogenase operon mutants, Δhyb hyc hyf, Δhya hyc hyf, Δhya hyb hyf, and Δhya hyb hyc, in which only a single hydrogenase is intact in the genome, were constructed as well as one quadruple mutant (Δhya hyb hyc hyf) that all four hydrogenase operons were deleted. First, single operon mutants and single-gene mutants for each hydrogenase showed different hydrogen productivity and growth in the anaerobic fermentation, indicating that bacterial phenotype regarding the hydrogen metabolism via the deletion of each operon is different with that of each single gene. Then, 4 triple hydrogenase operon mutants and one quadruple mutant were investigated to evaluate the hydrogen metabolism (hydrogen production and uptake) using glucose or glycerol as a substrate of hydrogen fermentation. With both the carbon sources, only Hyd-2 and Hyd-3 were able to produce hydrogen. Furthermore, all the hydrogenases showed hydrogen uptake activity. In addition, no hydrogen production and hydrogen uptake were detected in the quadruple mutant which does not have all 4 hydrogenases. Hydrogen production from Hyd-2 and Hyd-3 was further confirmed by complementing their operons in the cloning vector pBR322.  相似文献

Cell binding assays on antibody arrays permit the rapid immunophenotyping of living cells. The throughput of the analysis, however, is still limited due to our inability to perform parallel and quantitative detection of cells captured on the array. To address this limitation, we employed here an imaging technique based on surface plasmon resonance (SPR). SPR has been frequently used to monitor capture of proteins on antibody microarrays, while few cases were reported for capture of cells. Antibody arrays were prepared through the photopatterning of an alkanethiol monolayer on a gold-evaporated glass plate and the subsequent immobilization of various antibodies onto 4-9 separate spots created by photopatterning. A glass slip was mounted onto the array with a thin spacer to construct a parallel-plate chamber. Leukemia cells were injected into the chamber to conduct a binding assay, while refractive index changes at the vicinity of the array surface were monitored by SPR imaging. We observed that SPR signals were intensified on specific antibody spots but not on nonspecific spots. Confocal laser scanning microscopy revealed that the observed SPR signals were attributed to cell deformations caused by multivalent interactions with immobilized antibody, which effectively elevated the refractive index of a medium phase within an evanescent field. This effect could be suitably utilized to monitor quantitatively cell binding to multiple spots from a heterogeneous cell population.  相似文献

Water trees are the most hazardous factor affecting the life of XLPE distribution cables and the major cause of insulation failure. It is well known that insulation failure causes stoppages in electrical service and requires extensive repair work. Up to now, deterioration of cable insulation has been diagnosed mainly by the dc leakage current method. However, application of this method for diagnostic measurements requires interruption of electrical service. Several types of hot‐line diagnostic methods (including the dc component current and dc superposition methods) were developed to detect water tree deterioration. However, these methods have some shortcomings, such as being subject to effects of stray currents and the accuracy of measurements not being sufficiently high. Therefore, we have attempted to develop a new hot‐line diagnostic method. We investigated whether the signals produced by superposition of voltages of various frequencies to the cover layer of energized cables were correlated with deterioration by the water treeing. As a result, we found that a deterioration signal of 1 Hz was observed when we superposed an ac voltage (commercial frequency × 2 +1 Hz) on the cover layer of cable where the water tree had occurred in the insulation. © 1999 Scripta Technica, Electr Eng Jpn, 130(2): 49–58, 2000  相似文献

This paper investigates a bubble, placed so that it bridges the cross section of a horizontal narrow divergent passage under Earth gravity (1-G). In a narrow passage, inertial forces are known to be small in comparison with viscous forces. Gravitational force is not dominant in bubble behavior in a horizontal narrow passage under the 1-G condition. In this sense, the bubble behavior in the passage is similar to that under a low-gravity condition. Understanding bubble behavior is important in relation to separating a gas from a two-phase gas-liquid flow and controlling a gas-liquid interface under low-gravity conditions. Thus, a single bubble is placed under the 1-G condition. The bubble geometry and its behavior are studied experimentally for gap sizes ranging from 0.5 mm to 2 mm and divergent angles from 1° to 5° using ethyl alcohol as the working fluid. The following results were obtained: (1) the bubble was found to move to the greater cross-sectional area; (2) the gas-liquid interface geometry in a top view can be expressed as a contact circle model that takes the maximum radius in the passage; and (3) the effects of gap size and the projected bubble area on bubble behavior are clarified. © 1998 Scripta Technica, Heat Trans Jpn Res, 27(6): 447–461, 1998  相似文献

Strain-specific PCR primers for Lactococcus lactis subsp. cremoris FC were developed using the randomly amplified polymorphic DNA (RAPD) technique. RAPD was used to generate strain-specific markers. A 1164-bp RAPD marker found to be strain-specific was sequenced, and a primer pair specific for L. lactis subsp. cremoris FC was designed. The specificity of this primer pair was tested with 23 L. lactis subsp. cremoris strains and 20 intestinal bacterial species, and was found to be strain-specific. Subsequently, this primer pair was subjected to the quantification of L. lactis subsp. cremoris FC in the feces of subjects fed fermented milk containing this strain. After administration, L. lactis subsp. cremoris FC was detected in the feces of all 7 subjects, with the maximum number being between 10(5) and 10(9) cells g(-1) of feces. Furthermore, this strain was detected in only one feces sample 2 weeks after administration was stopped. These results suggest that L. lactis subsp. cremoris FC can survive passage through the gastrointestinal tract.  相似文献
