The family of NAD(P)H-dependent short-chain dehydrogenases/reductases (SDRs) comprises numerous biocatalysts capable of C=O or C=C reduction. The highly homologous noroxomaritidine reductase (NR) from Narcissus sp. aff. pseudonarcissus and Zt_SDR from Zephyranthes treatiae, however, are SDRs with an extended imine substrate scope. Comparison with a similar SDR from Asparagus officinalis (Ao_SDR) exhibiting keto-reducing activity, yet negligible imine-reducing capability, and mining the Short-Chain Dehydrogenase/Reductase Engineering Database indicated that NR and Zt_SDR possess a unique active-site composition among SDRs. Adapting the active site of Ao_SDR accordingly improved its imine-reducing capability. By applying the same strategy, an unrelated SDR from Methylobacterium sp. 77 (M77_SDR) with distinct keto-reducing activity was engineered into a promiscuous enzyme with imine-reducing activity, thereby confirming that the ability to reduce imines can be rationally introduced into members of the “classical” SDR enzyme family. Thus, members of the SDR family could be a promising starting point for protein approaches to generate new imine-reducing enzymes.  相似文献

Ergothioneine has emerged as a crucial cytoprotectant in the pathogenic lifestyle of Mycobacterium tuberculosis. Production of this antioxidant from primary metabolites may be regulated by phosphorylation of Thr213 in the active site of the methyltransferase EgtD. The structure of mycobacterial EgtD suggests that this post-translational modification would require a large-scale change in conformation to make the active-site residue accessible to a protein kinase. In this report, we show that, under in vitro conditions, EgtD is not a substrate of protein kinase PknD.  相似文献

A thick-layer light-sensitive material consisting of glycerin-containing bichromated gelatin has been prepared and used to record real-time volume holograms with 0.44 μm helium-cadmium laser radiation. The thickness of the layers is 400–600 μm. The holographic sensitivity of the material is 1 J/cm2. Wear resistance measurements have been made with U8A steel bearing protective coatings based on the carbides of titanium, vanadium, and chromium or iron borides, for various forms of wear. The carbide and boride coatings raise the wear resistance considerably (by factors of 6-30 in accordance with the type of wear). Measurements have been made on the microhardness, microbrittleness, and adhesion of the coating to the substrate, which are effective characteristics that quite fully reflect the effects of the microhardness and state of stress, structure, and defectiveness on the wear resistance.  相似文献

The corrosion properties of TiCN, TiCN – AlN, and (TiCN – AlN) – (Fe – Cr) ceramics as well as those of the individual components TiN and TiC in a 3% NaCl solution have been investigated. The kinetics and the mechanism of anode dissolution of metals and oxidation of specimens have been studied by using polarization curves, chemical and x-ray phase analyses, Auger electron spectroscopy, and scanning electron microscopy (SEM). TiCN and TiCN – AlN composites have been found to be the most corrosion-resistant. The presence of a metallic binder in the titanium carbon nitride somewhat decreases the corrosion resistance of the ceramics. On the whole, however, the ceramics developed boast a significantly higher corrosion resistance than that of structural steel.  相似文献

A new diffuser capable of scattering light in one direction (one-dimensional diffuser) has been developed and experimentally tested. The diffuser is formed in a thick layer of dichromated gelatin. The scattering indicatrices of the diffuser measured in two mutually perpendicular directions are presented. It is demonstrated that the indicatrix width in one direction is significantly greater than that in the perpendicular direc-tion. However, no zero-order diffraction has been observed. The proposed one-dimensional diffuser exhibits close scattering indicatrices for reading at a wavelength of 0.44 and 0.63 μm.  相似文献

Übersicht Um die Betriebssicherheit großer supraleitender Magnetsysteme für die Kernfusionsforschung zu gewährleisten, muß die im Magnetsystem gespeicherte Energie schnell entladen werden können. Durch die veränderlichen großvolumigen Magnetfelder werden in benachbarten leitfähigen Strukturen durch Induktion hohe Wirbelströme hervorgerufen. Durch sie treten dort elektromagnetische Kräfte. auf, die einzelne Komponenten mechanisch überlasten können. Zur Berechnung dieser Kräfte wurde eine einfache numerischanalytische Methode entwickelt, die im folgenden vorgestellt und mit Finite-Elemente-Verfahren verglichen wird.
Calculation of eddy current densities and forces around large superconducting coils for nuclear fusion
Contents The reliable operation of the superconducting magnet systems used for the magnetic confinement in nuclear fusion research requires the ability of a fast discharge of the stored magnetic energy. The corresponding magnetic field variation gives rise to high eddy currents by inductive coupling with surrounding conductive components. The eddy current forces can lead to a mechanical overload of some of these components. A simple numerical-analytical method was developed in order to calculate these forces. This method is presented and compared with the Finite Element Method.

Lp(a), one of the most atherogenic lipoproteins, is believed to contribute significantly to vascular diseases in non-insulin-dependent diabetic (NIDDM) patients. Contradictive data have been published on these patients concerning plasma concentrations of Lp(a) and their relation to renal function. Since apo(a) fragments appear in urine, we measured urinary apo(a) in 134 NIDDM patients and 100 matched controls and related urinary apo(a) concentrations to plasma Lp(a) levels and kidney function. Plasma Lp(a) values were found to be significantly higher in NIDDM patients. NIDDM patients also secreted significantly more apo(a) into their urine as compared to control subjects. There was no correlation between creatinine clearance or albumin excretion and urinary apo(a) concentrations. Patients with macroalbuminuria exhibited a twofold higher apparent fractional excretion of apo(a) in comparison to patients with normal renal function. Urinary apo(a) values in both patients and control subjects were highly correlated to plasma Lp(a), yet no correlation was found with HbA1c or serum lipoproteins. It is concluded that urinary apo(a) excretion is correlated to plasma Lp(a) levels but not to creatinine clearance in patients suffering from NIDDM.  相似文献
