The potential of electrical field-flow fractionation (ElFFF) for characterization of metal nanoparticles was investigated in this study. Parameters affecting separation and retention such as applied DC voltage and flow rate were examined. Nanoparticles with different types of stabilizers, including citrate and tannic acid, were investigated. Changes to the applied voltage showed a significant influence on separation in ElFFF, and varying flow rate was used to improve plate heights in the experiments. For nanoparticles of a fixed size, the separation was based primarily on electrophoretic mobility. Particles with low electrophoretic mobility elute earlier. Therefore, citrate stabilized gold nanoparticles (-2.72 × 10(-4) cm(2) V(-1) s(-1)) eluted earlier than tannic acid stabilized gold nanoparticles (-4.54 × 10(-4) cm(2) V(-1) s(-1)) of the same size. In addition, ElFFF can be used for characterization of gold nanoparticles with different particle sizes including 10, 20, and 40 nm with a fixed stabilizing agent. For a specific separation condition, the separation of 10, 20, and 40 nm gold nanoparticles was clearly based on the particle size as opposed to the electrophoretic mobility, as the elution order was in order of decreasing mobility for 10 (-4.54 × 10(-4) cm(2) V(-1) s(-1)), 20 (-3.97 × 10(-4) cm(2) V(-1) s(-1)), and 40 (-3.76 × 10(-4) cm(2) V(-1) s(-1)) nm particles, respectively.  相似文献

This study aims to investigate the influence of heat treatment temperatures on the mechanical properties and chemical solubility (CS) of lithium disilicate-fluorcanasite glass-ceramics and to develop new dental materials. The glasses and glass-ceramics were prepared using CaF2-SiO2-CaO-K2O-Na2O-Li2O-Al2O3-P2O5-based glass system using a conventional melt quenching method followed by a two-stage crystallization process. This two-stage method involves two heating temperature steps: first at a constant temperature (TS1) of 600°C and second step at varying temperatures (TS2) of 650, 700, 750, and 800°C. The crystallization behavior, phase formation, microstructure, translucency characteristic, density, hardness, fracture strength, and CS were investigated. It was found that the lithium disilicate crystal acted as the main crystalline phase, and the crystalline phase of fluorcanasite occurred at the heat treatment temperatures of 750 and 800°C. In addition, it was found that density, hardness, fracture strength, and CS increased while the translucency values decreased with increasing heat treatment temperatures. Furthermore, the CS increased dramatically when the fluorcanasite phases occurred in the glass-ceramic samples. The maximum density values, Vickers hardness, fracture toughness, and flexural strength are 2.56 g/cm3, 6.73 GPa, 3.38 MPa.m1/2, and 259 MPa, respectively. These results may offer a possibility to design a new material for dental applications based on lithium disilicate-fluorcanasite glass-ceramics.  相似文献

Functional mesoporous carbon has been built using 1,10-phenanthroline as the fundamental building block, resulting in a nanoporous, high surface area sorbent capable of selectively binding transition metal ions. This material had a specific surface area of 870 m2/g, an average pore size of about 30 Å, and contained as much as 8.2 wt% N. Under acidic conditions, where the 1,10-phenanthroline ligand is protonated, this material was found to be an effective anion exchange material for transition metal anions like PdCl42- and H2VO41-. 1,10-Phenanthroline functionalized mesoporous carbon (“Phen-FMC”) was found to have a high affinity for Cu(II), even down to a pH of 1. At pHs above 5, Phen-FMC was found to bind a variety of transition metal cations (e.g. Co(II), Ni(II), Zn(II), etc.) from filtered ground water, river water and seawater. Phen-FMC displayed rapid sorption kinetics with Co(II) in filtered river water, reaching equilibrium in less than an hour, and easily lowering the [Co(II)] to sub-ppb levels. Phen-FMC was found to be more effective for transition metal ion capture than ion-exchange resin or activated carbon.  相似文献

2-Hydroxy-ethyl methacrylate (HEMA) is a major monomer released from resin-base dental restorative materials. HEMA is cytotoxic to pulp cells and leads to apoptosis. This study examined the effect of Translationally Controlled Tumor Protein (TCTP) against apoptosis from HEMA. TCTP from banana prawn (Penaeus merguiensis) was cloned and the protein was purified. It significantly increased the number of viable of HEMA-treated cells compared to HEMA-treated cells alone. Flow cytometry indicated the addition of TCTP at 10 μg/ml to 8 and 10 mM HEMA decreased the apoptotic cells from 20 to 10%. The proliferative property and anti-apoptotic activity against HEMA was concentration dependent. It was interesting that the added TCTP was not detected inside the cells and the native human TCTP was decreased after treated with HEMA and TCTP (20 μg/ml) + HEMA(10 mM) for 24 h. These results provided preliminary information, which may contribute to the development of less toxic dental materials.  相似文献

Surface-functionalized nanoporous silica, often referred to as self-assembled monolayers on mesoporous supports (SAMMS), has previously demonstrated the ability to serve as very effective heavy metal sorbents in a range of aquatic and environmental systems, suggesting that they may be advantageously utilized for biomedical applications such as chelation therapy. Herein we evaluate surface chemistries for heavy metal capture from biological fluids, various facets of the materials' biocompatibility, and the suitability of these materials as potential therapeutics. Of the materials tested, thiol-functionalized SAMMS proved most capable of removing selected heavy metals from biological solutions (i.e., blood, urine, etc.) Consequentially, thiol-functionalized SAMMS was further analyzed to assess the material's performance under a number of different biologically relevant conditions (i.e., variable pH and ionic strength) to gauge any potentially negative effects resulting from interaction with the sorbent, such as cellular toxicity or the removal of essential minerals. Additionally, cellular uptake studies demonstrated no cell membrane permeation by the silica-based materials generally highlighting their ability to remain cellularly inert and thus nontoxic. The results show that organic ligand functionalized nanoporous silica could be a valuable material for a range of detoxification therapies and potentially other biomedical applications.  相似文献

White spot syndrome virus (WSSV) is a major cause of high mortality in cultured shrimp all over the world. VP26 is one of the structural proteins of WSSV that is assumed to assist in recognizing its host and assists the viral nucleocapsid to move toward the nucleus of the host cell. The objective of this work was to produce a polyclonal antibody against VP26 and use it as a biosensor. The recombinant VP26 protein (rVP26) was produced in E. coli (BL21), purified and used for immunizing rabbits to obtain a polyclonal antibody. Western blot analysis confirmed that the antiserum had a specific immunoreactivity to the VP26 of WSSV. This VP26 antiserum was immobilized onto a gold electrode for use as the sensing surface to detect WSSV under a flow injection system. The impedance change in the presence of VP26 was monitored in real time. The sensitivity of the biosensor was in the linear range of 160–160000 copies of WSSV, indicating that it is good and sensitive for analysis of WSSV. The specificity of the biosensor was supported by the observation that no impedance change was detected even at high concentrations when using Yellow Head Virus (YHV). This biosensor may be applied to monitor the amount of WSSV in water during shrimp cultivation.  相似文献

Dental materials that can promote cell proliferation and function is required for regenerative pulp therapy. Resin modified glass ionomer cement (RMGIC), a broadly used liner or restorative material, can cause apoptosis to pulp cells mainly due to HEMA (2-hydroxyethyl methacrylate), the released residual monomer. Recent studies found that chitosan and albumin could promote release of protein in GIC while translationally controlled tumor protein (TCTP) has an anti-apoptotic activity against HEMA. The aim of this study was to examine the effect of chitosan and albumin modified RMGIC (Exp-RMGIC) supplemented with TCTP on pulp cell viability and mineralization. Exp-RMGIC+TCTP was composed of RMGIC powder incorporated with 15 % of chitosan, 5 % albumin and supplemented with TCTP mixed with the same liquid components of RMGIC. The effect of each specimen on pulp cells was examined using the Transwell plate. From the MTT assay, Exp-RMGIC+TCTP had the highest percentages of viable cells (P < 0.05) at both 24 and 74 h. Flow cytometry revealed that, after 24 h, Exp-RMGIC+TCTP gave the lowest percentages of apoptotic cells compared to other groups. There was no difference in alkaline phosphatase (ALP) activity among different formula of the specimens, while cells cultured in media with TCTP had higher ALP activity. Von Kossa staining revealed that RMGIC+TCTP, and Exp-RMGIC+TCTP had higher percentages of calcium deposit area compared to those without TCTP. It was concluded that Exp-RMGIC supplemented with TCTP had less cytotoxicity than RMGIC and can protect cells from apoptosis better than RMGIC supplemented with TCTP.  相似文献

An efficient non-viral gene delivery for varieties of cells has been considered essential for gene therapy and tissue engineering. This study evaluated transfection efficiency of chitosan (HW) with molecular weights (Mw) at 470 and degree of deacetylation (DDA) 80% and its depolymerization product (LW) with Mw at 16 kDa and DDA 54%, as well as epidermal growth factor (EGF) conjugated to chitosan–DNA microparticles of both HW and LW by using either disulfide linkage or NHS-PEO4-Maleimide as a cross linker. The results revealed that the depolymerized LW at chitosan/DNA charge ratio 56:1 and pH 6.9 gave high transfection efficiency in both KB, a cancer cell line, and fibroblast cells at about the same level of LipofectamineTM, but the EGF-conjugated chitosan–DNA polyplexes from these methods did not improve transfection efficiency, which may come from the aggregation and fusing of the complexes as shown in scanning electron microscopy. However, this depolymerized LW chitosan showed the potential for further development as a safe and cost-effective non-viral gene delivery vehicle.  相似文献

Mangosteen shell‐derived activated carbons (ACs) were prepared by potassium hydroxide (KOH) activation for refining crude biodiesel derived from the transesterification of waste cooking oil and methanol via an alkali catalyst. Effects of the KOH/char (w/w) ratio (0.50–1.50), activation temperature (500–900 °C), adsorption time (15–180 min) and dosage of activated carbon (3–11 g/L) on the efficiency of the adsorptive refining of biodiesel were explored. The AC prepared at a KOH/char (w/w) ratio of 1.25 at 800 °C exhibited the best ability to remove impurities contained in the crude biodiesel, and enhanced the purity of the biodiesel by up to 97.18 wt% at a dosage of 9 g/L and 120 min adsorption time. This was better than that refined by the conventional wet washing method (96.03 wt%). Equilibrium adsorption data fitted well with Freundlich isotherms, and the adsorption mechanism was controlled by the transport of impurities from the bulk solution to the surface of the AC.  相似文献
