Monocytes expressing the inflammation suppressing active CD11b, a beta2 integrin, may regulate neuroinflammation and modify clinical outcomes in amyotrophic lateral sclerosis (ALS). In this single site, retrospective study, peripheral blood mononuclear cells from 38 individuals living with ALS and 20 non-neurological controls (NNC) were investigated using flow cytometry to study active CD11b integrin classical (CM), intermediate (IM) and non-classical (NCM) monocytes during ALS progression. Seventeen ALS participants were sampled at the baseline (V1) and at two additional time points (V2 and V3) for longitudinal analysis. Active CD11b+ CM frequencies increased steeply between the baseline and V3 (ANOVA repeated measurement, p < 0.001), and the V2/V1 ratio negatively correlated with the disease progression rate, similar to higher frequencies of active CD11b+ NCM at the baseline (R = −0.6567; p = 0.0031 and R = 0.3862; p = 0.0168, respectively). CD11b NCM, clinical covariates and neurofilament light-chain plasma concentration at the baseline predicted shorter survival in a multivariable and univariate analysis (CD11b NCM—HR: 1.05, CI: 1.01–1.11, p = 0.013. Log rank: above median: 43 months and below median: 21.22 months; p = 0.0022). Blood samples with the highest frequencies of active CD11b+ IM and NCM contained the lowest concentrations of soluble CD11b. Our preliminary data suggest that the levels of active CD11b+ monocytes and NCM in the blood predict different clinical outcomes in ALS. 相似文献

Influence of diverse botanical sources (wheat, maize, waxy maize, cassava, potato, rice or waxy rice) on in vitro native starch digestibility has been investigated. Physicochemical properties (chemical composition, particles size and shape, surface features) of starch granules were determined with a view to explaining digestibility differences between samples. Rapidly digestible starch (RDS), slowly digestible starch (SDS) and resistant starch (RS) contents were measured according to Englyst method. Potato starch was shown to be composed of large rounded granules having smooth surfaces, which explains its slow enzymatic breakdown. Potato starch displayed the highest RS (86%) content and the lowest RDS content (9.9%). Since RS positively influences health and SDS may result in cell, tissue and/or organ damages, potato starch is an ideal starch nutrient. Conversely, waxy rice starch was rich in amylopectin and displayed small diameters and angular shapes, which are both known to facilitate enzymatic starch hydrolysis. It exhibited a near-zero RS content (0.9%) and a high RDS fraction (60%). According to this study, potato starch exhibited the best nutrient profile, followed up in this order by cassava, waxy maize, wheat, maize and waxy rice starches. 相似文献

This paper describes the characterization of ciders (both “natural” and sparkling cider) from the Principality of Asturias (northwest region of Spain) through the analysis of their protein content, based on their hydrophobic properties, and their foam characteristics. A reversed-phase high performance liquid chromatography (RP-HPLC) was applied to the protein analysis, and the foam parameters were measured with Bikerman’s method. Multivariate techniques allowed the authors to differentiate ciders on the basis of the press and foam taking technologies, and foam sensory quality. Feasible and robust models were constructed for classifying purposes. Higher than 95% correct classifications were obtained for differentiating ciders on the basis of the factors studied (cider making technology and foam sensory quality). The multivariate regression model computed allowed the authors to predict (correlation coefficients higher than 0.8) the foam parameters related to foam stability and bubble average lifetime in “natural” cider. 相似文献

Environmental benefits associated with reduced rates of nitrogen (N) application, while maintaining economically optimum yields
have economic and social benefits. Although N is an indispensable plant nutrient, residual soil N could leach out to contaminate
groundwater and surface water resources, particularly in sandy soils. A 2-year field study was conducted in an established
bermudagrass (Cynodon dactylon) pasture in the Lower Suwannee Watershed, Florida, to evaluate N application rates on forage yield, forage quality, and nitrate
(NO3-N) leaching in rapidly permeable upland sandy soils. Four N application rates (30, 50, 70, and 90 kg N ha−1 harvest−1) corresponding to 0.33, 0.55, 0.77 and IX, respectively, of recommended N rate (90 kg N ha−1 harvest−1) for bermudagrass hay production in Florida were evaluated vis-à-vis an unfertilized (0 N) control. Suction cups were installed
near the center of each plot at two depths (30 and 100 cm) to monitor NO3-N leaching. The grass was harvested at 28 days intervals to determine dry matter yield, N uptake, and herbage nutritive value.
Nitrogen application at the recommended rate produced the greatest total dry matter yield (~18.4 Mg ha−1 year−1), but a modeled economically optimum N rate of ~57 kg N ha−1 harvest−1 (~60% of the recommended N rate) projected an average dry matter yield of ~17.3 Mg ha−1 year−1, which represents >90% of the observed maximum yield. Nitrogen application increased nutritive quality of the grass, but
increases in N application rate above 30 kg N ha−1 did not result in significant increases in in vitro digestible organic matter concentration, and tissue crude protein was
not significant above 50 kg N ha−1. Across the sampling period, treatments with N rates ≤50 kg N ha−1 harvest−1 had leachate NO3-N concentration below the maximum contaminant limit of <10 mg l−1. Conversely, applying N at rates ≥70 kg N ha−1 harvest−1 resulted in leachate N concentration that exceeded the maximum contaminant limit, and suggest high risk of impacting groundwater
quality, if such rates are applied to soils with coarse (sand) textures. The study demonstrates that recommendation of a single
N application rate may not be appropriate under all agro-climatic conditions and, thus, a site-specific evaluation of best

Protein supplements have received increasing attention by consumers over the last few decades. However, hundreds of them have recently exhibited irregularities including lower quantities of proteins than disclosed values. This study aimed to evaluate the nutritional quality of six commercial protein supplement powders (calcium caseinate, milk protein concentrate, egg white, pea protein isolate, whey protein concentrate and soy protein isolate). The chemical composition, amino acid content and in vitro digestibility were examined. Thus, calculate the amino acid scores corrected for amino acid and protein digestibility. In vitro digestion was also conducted and protein hydrolysis was monitored by SDS-page. Calcium caseinate powder and whey protein concentrate were only composed of proteinogenic amino acids and exhibited the highest essential amino acid content. As regards in vitro digestibility, these two supplements perfectly meet the quality expectations of the Food and Agricultural Organization (FAO). Conversely, the other four supplements exhibited a lower quality than the FAO reference protein. This was due to low digestibility (for egg white and milk concentrate) and/or lack of a specific essential amino acid (for milk concentrate and pea isolate). 相似文献
