YM-32890 A and B, new types of macrolide antibiotics produced by Cytophaga sp

Two new types of macrolide antibiotics, YM-32890 A and B, have been isolated from the fermentation broth of cytophaga sp. YL-02905S. In this paper, the taxonomy of the producing strain, fermentation, isolation, structure elucidation, and biological activity of the antibiotics are reported. YM-32890 A inhibits the growth of staphylococci including a macrolide-resistant strain, but shows no antimicrobial activity against other Gram-positive, Gram-negative bacteria and yeast.     

Pulse-width tunable, self-frequency conversion of short opticalpulses over 200 nm based on supercontinuum generation

Self-frequency conversion of ultrashort optical pulses into arbitrary multiwavelength components without the use of a pump is achieved by using supercontinuum generation. Frequency conversion of a randomly-modulated 1.535-1.560 μm, 2.5 ps pulse stream at 6.3 Gbit/s into 1.41-1.64 μm, 0.29-1.0 ps pulse streams is demonstrated for the first time. The method features ultrawide tunability of over 200 nm, and multiwavelength, pulsewidth-tunable output for flexible WDM distribution networks     

Brillouin-Scattering Study of the Elastic Constants of ErVO4

The room-temperature elastic constants of ErVO₄ were considerably smaller than those of isostructural silicate and phosphate analogs. The generally "less-rigid" crystalline lattice and weaker metal-oxygen bond-strength in the RVO₄ (R = rare earth elements) phases indicates that these materials are of interest for potential applications as an interphase component in toughened oxide ceramic composites.     

Hydrophobic effects on protein/nucleic acid interaction: enhancement of substrate binding by mutating tyrosine 45 to tryptophan in ribonuclease Tl

Hydrophobic effects on binding of ribonuclease Tl to guaninebases of several ribonucleotides have been proved by mutatinga hydrophobic residue at the recognition site and by measuringthe effect on binding. Mutation of a hydrophobic surface residueto a more hydrophobic residue (Tyr45 – Trp) enhances thebinding to ribonucleotides, including mononucleotide inhibitorand product, and a synthetic substrate-analog trinudeotide aswell as the binding to dinucleotide substrates and RNA. Enhancementson binding to non-substrate ribonucleotides by the mutationhave been observed with free energy changes ranging from –2.2 to – 3 .9 kJ/mol. These changes are in good agreementwith that of substrate binding, –2.3 kJ/mol, which iscalculated from Michaelis constants obtained from kinetic studies.It is shown, by comparing the observed and calculated changesin binding free energy with differences in the observed transferfree energy changes of the amino acid side chains from organicsolvents to water, that the enhancement observed on guaninebinding comes from the difference in the hydrophobic effectsof the side chains of tyrosine and tryptophan. Furthermore,a linear relationship between nucleolytic activities and hydrophobicityof the residues (Ala, Phe, Tyr, Trp) at position 45 is observed.The mutation could not change substantially the base specificityof RNase Tl, which exhibits a prime requirement for guaninebases of substrates.     

Higher order structures and mechanical properties of bacterial homo poly(3-hydroxybutyrate) fibers prepared by cold-drawing and annealing processes

Pure bacterial homo poly(3-hydroxybutyrate) (PHB) fibers were prepared by melt spinning, followed by cold-drawing in an amorphous state at a temperature just above its glass transition temperature. Cold drawn fibers obtained were further drawn at higher temperatures, followed by annealing at various temperatures under tension. Relations among the processing conditions, higher order structures and mechanical properties were investigated using wide- and small-angle X-ray diffractions (WAXD and SAXD, respectively), birefringence, differential scanning calorimetry (DSC), and tensile measurements. PHB has two different crystalline forms, 2₁ helix conformation (α-form) and planar zigzag conformation (β-form). A single broad reflection of β-form was detected even in a PHB fiber drawn once at a temperature just above its T_g immediately after quenching and it tended to be stronger after 2nd drawing at higher temperatures. Annealing under low temperature and high tension facilitates the occurrence of β-form. It is suggested that the β-form crystal is formed not only from the tie chains between α-form lamella, but also from completely free amorphous chains. Changes in the amount of two types of crystals were analyzed using the WAXD integrated intensity. Birefringence of these fibers shows negative and positive values, depending on process conditions. Changes in higher order structure on the mechanical properties are also discussed.     

SleepExplorer: a visualization tool to make sense of correlations between personal sleep data and contextual factors

Getting enough quality sleep is a key part of a healthy lifestyle. Many people are tracking their sleep through mobile and wearable technology, together with contextual information that may influence sleep quality, like exercise, diet, and stress. However, there is limited support to help people make sense of this wealth of data, i.e., to explore the relationship between sleep data and contextual data. We strive to bridge this gap between sleep-tracking and sense-making through the design of SleepExplorer, a web-based tool that helps individuals understand sleep quality through multi-dimensional sleep structure and explore correlations between sleep data and contextual information. Based on a two-week field study with 12 participants, this paper offers a rich understanding on how technology can support sense-making on personal sleep data: SleepExplorer organizes a flux of sleep data into sleep structure, guides sleep-tracking activities, highlights connections between sleep and contributing factors, and supports individuals in taking actions. We discuss challenges and opportunities to inform the work of researchers and designers creating data-driven health and well-being applications.     

Peroxiredoxin 2 as a chemotherapy responsiveness biomarker candidate in osteosarcoma revealed by proteomics

Purpose : We aimed to identify novel chemotherapy responsiveness biomarkers for osteosarcoma (OS) by investigating the global protein expression profile of 12 biopsy samples from OS patients. Experimental design : Six patients were classified as good responders and six as poor responders, according to the Huvos grading system. The protein expression profiles obtained by 2‐D DIGE consisted of 2250 protein spots. Results : Among them, we identified 55 protein spots whose intensity was significantly different (Bonferroni adjusted p‐value<0.01) between the two patient groups. Mass spectrometric protein identification demonstrated that the 55 spots corresponded to 38 distinct gene products including peroxiredoxin 2 (PRDX 2). Use of a specific antibody against PRDX 2 confirmed the differential expression of PRDX 2 between good and poor responders, while PRDX 2 levels as measured by Western blotting correlated highly with their corresponding 2‐D DIGE values. The predictive value of PRDX 2 expression was further confirmed by examining an additional four OS cases using Western blotting. Conclusions and clinical relevance : These results establish PRDX 2 as a candidate for chemotherapy responsiveness marker in OS. Measuring PRDX 2 in biopsy samples before treatment may contribute to more effective management of OS.     

13 years’ operational experience of SPring-8 vacuum system

The first electron beam was stored in SPring-8 storage ring in March 1997. An overview of the 13 years’ operational experience concerning the vacuum system of the SPring-8 storage ring is presented. The evolution of the vacuum system and their performance are summarized. Moreover, the main vacuum failures and their impact on accelerator operation are described in detail. We analyze the relationship between the difference types of vacuum failure and downtime of user experiment.
