An object-based approach to generating shrub cover change maps of potential use for monitoring shrubland habitat reserves was developed and tested. A high fidelity, bi-temporal airborne image data set was generated through frame-based image acquisition, precise image-to-image registration, radiometric normalization, and selection of near-anniversary image acquisition dates with similar precipitation conditions prior to both image acquisitions. Image segmentation and classification processes were applied to the bi-temporal layer stack of very high spatial resolution visible and near infrared (V/NIR) image data, such that shrub change objects were delineated and identified directly.Image segments derived from the bi-temporal V/NIR image data set having 1 m spatial resolution delineated most shrub change features in a qualitatively realistic manner. A Standard Nearest Neighbor classifier with segment mean and standard deviation measures of Red, NIR, and normalized difference vegetation index (NDVI) image features yielded the shrub change map that agreed more closely with reference data than the classifier based on fuzzy membership functions. The overall accuracy and kappa statistics for the optimal shrub change map were 0.83 and 0.64, respectively, with the predominant error being associated with “over-classification” of no-change objects as some type of shrub change. No statistical difference in accuracies of three- and five-class maps was found, suggesting that changes in true shrubs and sub-shrubs within coastal sage scrub vegetation communities can be differentiated reliably. A net 5% loss of shrub cover was determined for the 1998–2005 period from the shrub change map of the study area. The greatest decrease and net loss of shrub cover occurred within the urban edge zone and within flat-lying areas. Patterns of shrub loss appear to be more related to anthropogenic disturbance than effects of the severe seven-year drought that occurred between image acquisition dates.  相似文献

Gene therapy is an attractive therapeutic method for the treatment of genetic disorders for which the efficient delivery of nucleic acids into a target cell is critical. The present study is aimed at evaluating the potential of copolymers based on linear polyglycidol to act as carriers of nucleic acids. Functional copolymers with linear polyglycidol as a non-ionic hydrophilic block and a second block bearing amine hydrochloride pendant groups were prepared using previously synthesized poly(allyl glycidyl ether)-b-polyglycidol block copolymers as precursors. The amine functionalities were introduced via highly efficient radical addition of 2-aminoethanethiol hydrochloride to the alkene side groups. The modified copolymers formed loose aggregates with strongly positive surface charge in aqueous media, stabilized by the presence of dodecyl residues at the end of the copolymer structures and the hydrogen-bonding interactions in polyglycidol segments. The copolymer aggregates were able to condense DNA into stable and compact nanosized polyplex particles through electrostatic interactions. The copolymers and the corresponding polyplexes showed low to moderate cytotoxicity on a panel of human cancer cell lines. The cell internalization evaluation demonstrated the capability of the polyplexes to successfully deliver DNA into the cancer cells.  相似文献

The monoterpenic composition of Alvarinho, Arinto, Avesso, Azal, Batoca, Loureiro and Trajadura Vitis vinifera cultivars from the Vinhos Verdes region (Portugal), in respect to free volatile compounds as well as glycosidically bound aroma precursors, was determined by GC‐MS after adsorption on XAD‐2 resin. Eighteen monoterpenic compounds were identified and quantified in the free form and twenty‐two compounds in the glycosidically bound form. In the free fraction, the Loureiro variety could be easily differentiated from the other six varieties by the important levels of linalool, above the odor perception threshold. This was in contrast to Alvarinho, Avesso, Arinto, Azal and Trajadura, where geraniol prevailed. The Batoca variety showed a very poor monoterpenic profile in the free form. On the other hand, Alvarinho was the richest variety in respect to the glycosidically bound form, followed by Loureiro. In this fraction, linalool and 3,7‐dimethylocta‐1,5‐dien‐3,7‐diol were the most abundant compounds in the Loureiro grape cultivar; additionally, the isomer (E)‐8‐hydroxylinalool was in a higher concentration than (Z)‐8‐hydroxylinalool (sub‐region of Sousa), while for the rest of the varieties the (Z) isomer prevailed.  相似文献

Wine aroma is formed by volatile compounds of different chemical natures and origins. Betanzos is the most northern viticole geographic area from Galicia (NW Spain). The aromatic profiles of wines produced from two white varieties (Blanco lexítimo and Agudelo) and one red variety (Serradelo) from Betanzos, harvested in 2006 and 2007, have been established by gas chromatography (GC/FID). A total of 35 varietal and fermentative aroma compounds were identified and quantified in free volatile form, which include terpenols, C13‐norisoprenoids, alcohols, esters, volatile fatty acids and volatile phenols. To ascertain the compounds with major impact odours, the Odour Activity Value (OAV) was calculated. Twelve compounds showed concentrations above their perception threshold (OAV>1). The results obtained suggest that ethyl octanoate (apple), isoamyl acetate (banana), ethyl hexanoate (fruity) and β‐damascenone (floral) were the most powerful odorants for the white wines Blanco lexítimo and Agudelo from Betanzos. Ethyl octanoate and β‐damascenone (fruity and floral aroma respectively) were the most odorant for the red wine Serradelo.  相似文献

A competitive immunoassay for staphylococcal enterotoxin A (SEA) detection in milk was developed, using immobilised antibody onto magnetic nanoparticles (MNPs). MNPs were prepared and then modified to introduce amino groups on them. The morphology and size of the obtained both unmodified and modified MNPs were characterized using TEM analyses. Monoclonal anti-SEA antibody was immobilised onto the modified MNPs (MNP-Ab). Staphylococcal enterotoxin A was conjugated with fluorescent dye ATTO620NHS. The characteristics of fluorescence conjugate were examined. The amount of MNP-Ab and concentration of the fluorescent conjugate used for competitive immunoassay were optimized: 0.25 mg and 53 μg mL−1, respectively. The detection limit of developed immunoassay was determined – 0.23 ng mL−1 SEA in spiked milk samples. The immunoassay takes only 30 min, the magnetic separation is fast (<10 s) and the volume of the sample for analysis is very small (200 μL).  相似文献

The pineal gland regulates the aging process via the hormone melatonin. The present report aims to evaluate the effect of pinealectomy (pin) on behavioral and oxidative stress-induced alterations in cholesterol and sphingomyelin (SM) levels in young adult, mature and aging rats. Sham and pin rats aged 3, 14 and 18 months were tested in behavioral tests for motor activity, anxiety, and depression. The ELISA test explored oxidative stress parameters and SM in the hippocampus, while total cholesterol was measured in serum via a commercial autoanalyzer. Mature and aged sham rats showed low motor activity and increased anxiety compared to the youngest rats. Pinealectomy affected emotional responses, induced depressive-like behavior, and elevated cholesterol levels in the youngest rats. However, removal of the pineal gland enhanced oxidative stress by diminishing antioxidant capacity and increasing the MDA level, and decreased SM level in the hippocampus of 14-month-old rats. Our findings suggest that young adult rats are vulnerable to emotional disturbance and changes in cholesterol levels resulting from melatonin deficiency. In contrast, mature rats with pinealectomy are exposed to an oxidative stress-induced decrease in SM levels in the hippocampus.  相似文献

One of the pathological hallmarks of Alzheimer’s disease (AD) associated with its progression that contributes to β-amyloid (Aβ) generation is oxidative stress (OS). Clinical data suggest that melatonin is a potent antioxidant that might be effective in the adjunctive therapy of this neurodegenerative disease. The present study aimed to explore the role of melatonin on behavioral changes and markers of OS in three rat models, namely, pinealectomy (pin) model of melatonin deficit, intracerebroventricular (icv)Aβ1-42 model of AD, and combination of both pin and Aβ1-42 model (pin+icvAβ1-42). The chronic injection with vehicle/melatonin (50 mg/kg, i.p. for 40 days) started on the same day of sham/pin and icv vehicle/Aβ1-42 infusion procedures. Anxiety in the open field and the elevated plus-maze test and cognitive responses in the object recognition test were tested between the 30th–35th day after the surgical procedures. Markers of OS in the frontal cortex (FC) and hippocampus were detected by the ELISA method. Melatonin treatment corrected the exacerbated anxiety response only in the pin+icvAβ1-42 model while it alleviated the cognitive impairment in the three models. Pinealectomy disturbed the antioxidant system via enhanced SOD activity and decreased GSH levels both in the FC and hippocampus. The Aβ1-42 model decreased the SOD activity in the FC and elevated the MDA level in the two brain structures. The pin+icvAβ1-42 model impaired the antioxidant system and elevated lipid peroxidation. Melatonin supplementation restored only the elevated MDA level of icvAβ1-42 and pin+icvAβ1-42 model in the hippocampus. In conclusion, our study reveals that the pin+icvAβ1-42 rat model triggers more pronounced anxiety and alterations in markers of OS that may be associated with melatonin deficit concomitant to icvAβ1-42-induced AD pathology.  相似文献

White wine was produced with Saccharomyces cerevisiae cells immobilized on grape pomace, by natural adsorption. The support, the main solid waste from the wine industry, consisted of the skins, seeds and stems. Immobilization was tested using different media, namely complex culture medium, raw grape must and diluted grape must. Grape pomace was revealed to be an appropriate support for yeast cell immobilization. Moreover, grape must was shown to be the most suitable medium as immobilized cells became adapted to the conditions in the subsequent alcoholic fermentation in the wine‐making process. The wines produced, either with immobilized cells or with free cells, were subjected to chemical analysis by HPLC (ethanol, glycerol, sugars and organic acids) and by gas chromatography (major and minor volatile compounds); additionally, colour (CIELab) and sensory analysis were performed. The use of immobilized systems to conduct alcoholic fermentation in white wine production proved to be a more rapid and a more efficient process, especially when large amounts of SO2 were present in the must. Furthermore, the final wines obtained with immobilized cells demonstrated improved sensory properties related to the larger amounts of ethanol and volatile compounds produced. The more intense colour of these wines could be a drawback, which could be hindered by the reutilization of the biocatalyst in successive fermentations. MgH2 75 at.%–C 25 at.% composites are synthesized by ball milling using different kinds of carbon additives: carbon black (CB), nanodiamonds (ND) and amorphous carbon soot (AC). X-ray diffraction analysis showed that the MgH2 phase in the as-obtained composite powders is nanocrystalline (80–100 nm). The SEM observations revealed that the samples consist of 5–15 μm MgH2 particles, surrounded and in some cases coated by carbon flakes. The composite containing nanodiamonds revealed strong decrease of the MgH2 decomposition temperature with more than 100 °C, compared to ball-milled pure MgH2. Important issue of the present study is also the low temperature hydriding of the ball-milled Mg–C nanocomposites, investigated by high-pressure DSC. The process starts at about 200 °C for all materials studied, but the hydriding mechanism looks different for the composites with different kinds of carbon additives. Whereas for Mg–carbon black it takes place in a relatively narrow temperature range, expressed by a single exothermic peak (200–300 °C) for the other two composites the hydriding is a multi-step process, featured by two overlapped exothermic peaks for Mg-nanodiamonds and by two well separated exothermic effects (at about 300 °C and 400 °C) for Mg-amorphous carbon soot. The observed difference in the hydriding behavior of the Mg–C composites is attributed to the different kind of carbon component, which is supposed to play a catalytic role as well as protects magnesium from oxidation. The incorporation of carbon into the MgH2 particles results in the formation of high density of defects (dislocations and grain boundaries), which is supposed to be among the most possible reasons for the decreased hydride decomposition temperature. The Mg–C nanocomposites show reproducible hydriding/dehydriding behavior (thermodynamics and kinetics) during multiple cycling. Among the composites in the present study “Mg–carbon black” reveals the best hydriding characteristics – low temperature of hydriding in a relatively narrow temperature range by a single-step reaction and relatively fast hydriding kinetics.  相似文献
