Cell therapy of the post-infarcted myocardium is still far from clinical use. Poor survival of transplanted cells, insufficient regeneration, and replacement of the damaged tissue limit the potential of currently available cell-based techniques. In this study, we generated a multilayered construct from adipose-derived mesenchymal stromal cells (MSCs) modified to secrete stem cell factor, SCF. In a rat model of myocardium infarction, we show that transplantation of SCF producing cell sheet induced activation of the epicardium and promoted the accumulation of c-kit positive cells in ischemic muscle. Morphometry showed the reduction of infarct size (16%) and a left ventricle expansion index (0.12) in the treatment group compared to controls (24–28%; 0.17–0.32). The ratio of viable myocardium was more than 1.5-fold higher, reaching 49% compared to the control (28%) or unmodified cell sheet group (30%). Finally, by day 30 after myocardium infarction, SCF-producing cell sheet transplantation increased left ventricle ejection fraction from 37% in the control sham-operated group to 53%. Our results suggest that, combining the genetic modification of MSCs and their assembly into a multilayered construct, we can provide prolonged pleiotropic effects to the damaged heart, induce endogenous regenerative processes, and improve cardiac function.  相似文献

2-Hydroxy-1,4-napthoquinone(lawsone) natural red-orange dye was extracted from fresh henna(Lawsonia inermis) leaves in an alkaline media.The lawsone-surfactant solubilization constants(K_(LS))were calculated for the first time by using cationic cetyltrimethylammonium bromide(CTAB) and anionic sodium dodecyl sulphate(SDS).The standard free energy,concentration of solubilized lawsone and number of lawsone molecules solubilized into micelles were calculated and discussed.Surface excess,minimum surface area per molecule,surface pressure,free energy(adsorption and aggregation) and equilibrium constants of different states were determined from tensiometry.Different metal ions(Ag~+,Co~(2+),Cu~(2+),Ni~(2+),Fe~(3+),Zn~(2+) and Al~(3+)) were used to determine the complex forming ability with lawsone.Out of these,Ag~+ ions have strong binding capacity with lawsone.The adsorption of lawsone on the surface of glass with silver ions in presence of CTAB was also observed at pH 9.0.The pseudo-first, secondorder kinetic equation,intraparticles diffusion and Elovich models were used to determine the kinetics of lawsone adsorption onto the surface of glass and a probable mechanism has been discussed.Lawsone adsorption followed second-order kinetic equation(k_2=0.019 g·mg~(-1)·min~(-1)).  相似文献

A growing trend within nanomedicine has been the fabrication of self‐delivering supramolecular nanomedicines containing a high and fixed drug content ensuring eco‐friendly conditions. This study reports on green synthesis of silica nanoparticles (Si‐NPs) using Azadirachta indica leaves extract as an effective chelating agent. X‐ray diffraction analysis and Fourier transform‐infra‐red spectroscopic examination were studied. Scanning electron microscopy analysis revealed that the average size of particles formed via plant extract as reducing agent without any surfactant is in the range of 100–170 nm while addition of cetyltrimethyl ammonium bromide were more uniform with 200 nm in size. Streptomycin as model drug was successfully loaded to green synthesised Si‐NPs, sustain release of the drug from this conjugate unit were examined. To grapple with multidrug resistant bacterial infections, implementations of antibacterial nanomedicines have gained prime attention of the researchers across the globe. Nowadays, zinc oxide (ZnO) at nano‐scale has emerged as a promising antibacterial therapeutic agent. Keeping this in view, ZnO nanostructures (ZnO‐NS) have been synthesised through reduction by P. aphylla aqueous extract without the utilisation of any acid or base. Structural examinations via scanning electron microscopy (SEM) and X‐ray diffraction have revealed pure phase morphology with highly homogenised average particle size of 18 nm. SEM findings were further supplemented by transmission electron microscopy examinations. The characteristic Zn–O peak has been observed around 363 nm using ultra‐violet–visible spectroscopy. Fourier‐transform infrared spectroscopy examination has also confirmed the formation of ZnO‐NS through detection of Zn–O bond vibration frequencies. To check the superior antibacterial activity of ZnO‐NS, the authors'' team has performed disc diffusion assay and colony forming unit testing against multidrug resistant E. coli, S. marcescens and E. cloacae. In this study, barium-doped lanthanum manganite, La0.8Ba0.2MnO3, was synthesized via a mechano-thermal route employing high energy ball milling and subsequent heat treatment. The structural evolution, morphology and thermal behaviour of the powders were evaluated using XRD, FESEM, and DTA/TGA, respectively. DTA/TGA results showed that the calcination temperature of the carbonates significantly decreased by increasing the milling time. The results revealed that single phase perovskite was formed at 900 °C in a milled sample for 2 h and this temperature decreased to 600 °C by increasing the milling time to 30 h. The mean crystallite size also decreased from 32 to 20 nm by increasing the milling time from 2 to 30 h. The reaction sequence of La0.8Ba0.2MnO3 formation via the mechano-thermal route is proposed using XRD and DTA/TGA results. FESEM micrographs showed that the mean particle size of the perovskite phase is increased slightly from 30 to 40 nm by increasing the heat treatment temperature from 600 to 900 °C.  相似文献

This paper links the industrial policy and national systems of innovation literature to the investigation of learning capabilities of suppliers in the context of the automotive parts industry of Pakistan. Drawing data from 50 Pakistani autoparts suppliers, the findings suggest that industrial policy has been helpful in creating a local parts supply base and facilitating the entry of Japanese assemblers in the market. However, the implementation of the policy has been weak, and it is an arduous journey for the local suppliers to develop ambidextrous (exploratory and exploitative) learning capabilities. The findings also indicate that where local training and support from R&D institutions are weak, networking alone with foreign multinationals is not sufficient on its own to develop exploratory learning capabilities of local suppliers. This paper shows the importance of creating national–provincial institutions offering learning and skills development aimed towards innovation.  相似文献

Food induced viscosity can delay disintegration and subsequent release of API from solid dosage form which may lead to severe reduction in the bioavailability of BCS type III compounds. Formulations of such tablets need to be optimized in view of this postprandial viscosity factor. In this study, three super disintegrants, croscarmellose sodium (CCS), cross-linked polyvinylpolypyrrolidone (CPD), and sodium starch glycolate (SSG) were assessed for their efficiency under simulated fed state. Tablets containing these disintegrants were compressed at 10 and 30?KN, while taking lactose as a soluble filler. In addition to other compendial tests, disintegration force of these formulations was measured by texture analysis. Comparison of parameters derived from force – time curves revealed a direct relation of maximum disintegration force (Fmax) and disintegration force development rate (DFDR) with compressional force in fasted state, whereas an inverse relationship of Fmax and DFDR with compressional force was observed in fed state. The gelling tendency of disintegrants influenced the rate of release of API in simulated fed and fasted states when compressional force was changed. These observations recommend the evaluation of formulations in simulated fed state, in the development stage, with an objective of minimizing the negative impact of food induced viscosity on disintegration. Use of disintegrants that act without gelling or can counteract the effect of gelling is recommended for tablet formulations with reduced disintegration time (DT) and mean dissolution time (MDT) in fed state, respectively.  相似文献

It is a common practice to model multi‐storey tall buildings as frame structures where the loads for structural design are supported by beams and columns. Intrinsically, the structural strength provided by the walls and slabs are neglected. As the building height increases, the effect of lateral loads on multi‐storey structures increases considerably. The consideration of walls and slabs in addition to the frame structure modelling shall theoretically lead to improved lateral stiffness. Thus, a more economic structural design of multi‐storey buildings can be achieved. In this research, modelling and structural analysis of a 61‐storey building have been performed to investigate the effect of considering the walls, slabs and wall openings in addition to frame structure modelling. Sophisticated finite element approach has been adopted to configure the models, and various analyses have been performed. Parameters, such as maximum roof displacement and natural frequencies, are chosen to evaluate the structural performance. It has been observed that the consideration of slabs alone with the frame modelling may have negligible improvement on structural performance. However, when the slabs are combined with walls in addition to frame modelling, significant improvement in structural performance can be achieved. Copyright © 2012 John Wiley & Sons, Ltd.  相似文献

Recently, the drawbacks arising from the overuse of antibiotics have drawn growing public attention. Among them, drug-resistance (DR) and even multidrug-resistance (MDR) pose significant challenges in clinical practice. As a representative of a DR or MDR pathogen, Staphylococcus aureus can cause diversity of infections related to different organs, and can survive or adapt to the diverse hostile environments by switching into other phenotypes, including biofilm and small colony variants (SCVs), with altered physiologic or metabolic characteristics. In this review, we briefly describe the development of the DR/MDR as well as the classical mechanisms (accumulation of the resistant genes). Moreover, we use multidimensional scaling analysis to evaluate the MDR relevant hotspots in the recent published reports. Furthermore, we mainly focus on the possible non-classical resistance mechanisms triggered by the two important alternative phenotypes of the S. aureus, biofilm and SCVs, which are fundamentally caused by the different global regulation of the S. aureus population, such as the main quorum-sensing (QS) and agr system and its coordinated regulated factors, such as the SarA family proteins and the alternative sigma factor σB (SigB). Both the biofilm and the SCVs are able to escape from the host immune response, and resist the therapeutic effects of antibiotics through the physical or the biological barriers, and become less sensitive to some antibiotics by the dormant state with the limited metabolisms.  相似文献

Abstract

A series of Cu-K/Al2O3 catalysts were synthesized by wet impregnation technique. The reduced catalysts were further used for conversion of carbon dioxide to methane and carbon monoxide. Moreover, the fresh and used catalysts were characterized to investigate the changes in the surface morphology, metal dispersion, surface area, crystalline phases, and functional groups of studied catalysts. The SEM analysis of fresh and spent catalysts showed no remarkable difference in surface morphology with irregular shaped agglomerated particles. Furthermore, TEM micrographs presented the well distribution of metal catalyst over alumina support. The decrease in surface area from 115 to 77?m2/g for Cu1.62-K0.5/Al2O3 after reaction was related to sintering and oxidation of catalyst during reaction. XRD revealed the disappearance of some minor peaks which can be associated with the sintering of spent catalyst. FTIR also presented some new peak for spent catalyst which can be linked with metal oxides. Moreover, various reaction conditions of temperature (230, 400, and 600?°C), pressure (1 and 7?bar), and feed molar ratio of H2/CO2 (2:1 and 4:1) were investigated using different Cu loading (0, 1, 1.25, 1.62, and 4 weight percent). A maximum CO2 conversion of 63% with 39% CH4 selectivity was achieved by using Cu1.62-K0.5/Al2O3 at 600?°C, molar ratio of H2/CO2 4 under 7?bar. The presence of K on the surface of synthesized catalyst increased the CO2 conversion from 48% (Cu1/Al2O3) to 55% (Cu1-K0.5/Al2O3) at above mentioned reaction conditions which suggested the promoter effect of K during conversion of carbon dioxide.  相似文献
