I discuss three-period crossover designs for an efficient comparison of two test treatments with special application to clinical trials which often have many practical limitations. In this paper I specify a subset of three-period crossover designs so that the investigators are not left with the problematic two-period two-sequence design, should the trials be terminated after the second period. I show that there is a dramatic reduction in variability for estimating the direct and residual treatment effects in three-period designs compared to two-period designs. I also show that the universally optimal design with ABB and BAA sequences is unsuitable when a complex form of residual effects is suspected, such as the second-order residual effects or treatment by period interactions. The design with ABB, BAA, AAB, and BBA sequences is relatively robust to these uncertain model assumptions. I also discuss missing data problems and conclude that, even with a large proportion of missing values, the three-period design is far more efficient than the two-period design.  相似文献

The steel/steel boundary friction properties of soybean oil (SBO) and high-oleic soybean oil (HOSBO) are compared. HOSBO is significantly more saturated than SBO and more oxidatively stable. Changes in degree of unsaturation affect lateral interactions of adsorbate molecules, which in turn affects their adsorption and, hence, their boundary lubrication properties. To investigate this possibility, the free energies of adsorption (ΔG ads) of SBO, HOSBO, and methyl laurate (ML) were determined from the analysis of friction-derived adsorption isotherms using the Langmuir and Temkin adsorption models. The results showed a stronger adsorption for the vegetable oils than for ML, an indication of multiple interactions between the ester groups of the triglycerides and the steel surface. The result also showed no difference in the ΔG ads values of SBO and HOSBO obtained using either the Langmuir or Temkin models. This was interpreted as an indication of the lack of appreciable net lateral interaction between triglyceride adsorbates.  相似文献

Sensory Analysis of Whole Wheat/Soy Flour Breads   总被引:1,自引:0,他引:1  
ABSTRACT: Breads containing 0% to 40% defatted soy flour, 35% to 100% whole wheat flour, and 0% to 35% white bread flour were prepared. Beany or bitter flavor ratings for the yeast-leavened breads containing up to 30% soy flour were not significantly different from the control whole wheat bread. A high-quality yeast was the most important factor in reducing the beany taste of the bread. Addition of ascorbic acid significantly increased yeasty flavor, and there was a strong inverse correlation between beany and sweet flavors. These studies suggest that an appealing, nutritious bread containing up to 30% to 40% soy flour can be prepared in an easy and economical manner using equipment available in home or institutional kitchens.  相似文献

A foam rheometer has been developed based on the impulse theory of linear viscoelasticity. The rheometer is a large-volume parallel-plate device which operates under gap-loaded conditions, and is designed as an add-on fixture for the Materials Testing System Model 312. The rheometer provides characterization data of the pre-gel (via the zero-shear viscosity) or the post-gel (via the equilibrium modulus) properties of a foaming cellular polymer with a precision of better than ± 15%.  相似文献

Sample size sensitivity of thermal gravimetric analysis (TGA) presents a serious handicap in the obtaining of reliable thermal stability data for high-temperature applications. For example, predictions of the apparent time for degradation during an isothermal experiment based on the results obtained using a 10 μm thick specimen can be off by an order of magnitude when applied to a product with a thickness of 10 cm. To address this effect, TGA experiments studying the thermal degradation of poly(methyl methacrylate) were conducted. Analysis of the experimental data resulted in the development of a relationship between the apparent time scale of the thermal degradation and the specimen thickness. Origins of the new dependence were traced to the change of the diffusivity resulting from material volatilization. Implications of the coupling between these two events for the analysis of thermal stability for new polymeric materials are discussed, and required changes in the current methodology are outlined.  相似文献

The effect of starch volume fraction on starch‐filled poly(hydroxy ester ether) (PHEE) biodegradable composites was analyzed using Mooney, Thomas, Maron & Pierce, and Frankel & Acrivos equations. Corn starch/PHEE materials were extruded using a twin‐screw extruder with starch volume fractions from 0.27 to 0.66. The starch was dried to a moisture content of 0.7% to minimize moisture effect on composite rheology. Dynamic frequency sweep measurements were carried out at 120°C and 1% strain. The equations of Maron & Pierce, Thomas, and Frankel & Acrivos can be used to describe the experimental data. It is found that the relative viscosity at the same shear stress increases with increasing shear stress. The possible existence of yield stress at high starch concentrations was examined using a Casson plot.  相似文献

The Saccharomyces cerevisiae Lanosterol 14 alpha-demethylase (14DM) gene was overexpressed in S. cerevisiae using promoter sequences of the highly expressed S. cerevisiae glyceraldehyde-3-phosphate dehydrogenase (TDH3) gene. To investigate factors affecting 14DM overproduction, the levels of 14DM-specific RNAs, apoprotein, and heme protein, respectively, were determined and the 14DM-specific RNA levels compared with the RNA levels originating from the endogenous TDH gene(s). The quantitative measurements revealed that the 14DM steady-state RNA levels reached were some three- to five-fold below the theoretically expected values. With a view towards further improving expression of the 14DM gene, the spacing between the TDH3 promoter and the AUG was adjusted precisely and to rule out possible toxic effects exerted by the 14DM protein, the TDH3 promoter was placed under galactose regulation by introducing an UASG segment. Furthermore, the effects of the gene copy number on 14DM overproduction were investigated. From the analysis of the improved expression constructs five conclusions could be reached: (1) expression from the native 14DM gene is comparable to the expression driven by the TDH3 promoter-14DM fusion construct on single copy plasmid vectors; (2) expression from the TDH3 promoter-14DM construct on single-copy vectors is nearly as efficient as expression from the corresponding endogenous TDH3 gene; (3) the gene copy number has an effect on the relative expression levels of the TDH3 promoter-14DM constructs; (4) the steady-state amounts of protein produced are very nearly proportional to gene dosage; and (5) protein toxicity does not have a major impact on 14DM production. The maximum yield of 14DM was in the order of 7% of the total yeast protein and the maximum production of functional 14DM heme protein appears to be limited by the availability of heme.  相似文献

The rheological behavior of multilayer polyjeric structures has been investigated Measurements of the bulk viscoelastic properties via small-amplitude oscilatory rheometry indicated that the shear viscosity is independent of both the numbe of layers (83 vs. 165) and the composition (30/70, 50/50, and 70/30 PC/PMMA by weight) within the limits of the data obtained. It is also apparent tha tthe shear viscosity is influenced strongly by the skin layer material. In additon, a model has been developed tha tcan be used to proedict the shear viscosity and shear stress ofa multilayer structure experiencing shear flow. The model predicts tha thte shear viscosity of a multilayer structure should be independent of the number of layers and strongly dependent on the material in the skin layer. These predictions are in agreement with experimental data.  相似文献
