A low-cost and simple on-site technique for genotyping single nucleotide polymorphisms (SNPs) was developed. The technique is based on allele-specific primer PCR and the recently developed bead arrays in a single tip technique. We have established a quantitative method for evaluation of ocular surface lesions on the basis of lactate dehydrogenase (LDH) activity, malate dehydrogenase (MDH) activity, and albumin levels in rabbit tears. Lesions were produced with solutions of 0.005-0.02% benzalkonium chloride (BAK), 0.01-0.03% chlorhexidine digluconate (CHX), and 0.01-0.03% polyhexamethylene biguanide hydrochloride (PHMB), all of which are widely used in topical ophthalmic preparations. Two drops of test solution were instilled 15 times into rabbit eyes at 5 minute intervals. Sixty minutes after 0.02% BAK instillation, tear LDH activity increased from 1,840 U/L (without instillation) to 26,100 U/L, and concomitantly tear albumin levels rose from 0.11 mg/mL (without instillation) to 9.48 mg/mL. Instillation of 0.03% CHX and 0.03% PHMB caused smaller increases in LDH and MDH activity and albumin tear levels. LDH activity and albumin levels in tears were significantly correlated with the degree of total ocular surface lesions in both cornea and conjunctiva as observed by slit lamp biomicroscopy quantified using a modified Draize score. Based on the results of this study, we believe that LDH activity and albumin level in tears can be used as objective indicators for the quantitative evaluation of ocular surface lesions on both cornea and conjunctiva following application of topical ophthalmic preparations containing cytotoxic preservatives in animals and man.  相似文献

A single-nucleotide-polymorphism-typing method using a novel three-dimensional DNA microarray, Bio-Strand, is promising because it is rapid, inexpensive and easily automated. It has been developed with the intent to overcome the drawbacks of conventional DNA microarrays, which use flat surfaces and impermeable materials such as glass slides; Bio-Strand as a novel DNA microarray, with its permeability, has a significantly improved stability compared with conventional DNA microarrays that use impermeable materials. In this study, we have developed a simple method of pretreating a polyamide monofilament to increase its surface area and to make it permeable, which makes Bio-Strand more sensitive and stable, allowing it to be adapted for clinical diagnostic applications. The fluorescence signal obtained with a nylon 6 monofilament pretreated under optimal conditions (hydrolysis by 5 M HCl/ethanol followed by washing with 50% ethanol and 100% ethanol) was significantly stronger than that obtained with an untreated monofilament.  相似文献

A fuzzy self-tuning parallel genetic algorithm for optimization   总被引:1,自引:0,他引:1  
The genetic algorithm (GA) is now a very popular tool for solving optimization problems. Each operator has its special approach route to a solution. For example, a GA using crossover as its major operator arrives at solutions depending on its initial conditions. In other words, a GA with multiple operators should be more robust in global search. However, a multiple operator GA needs a large population size thus taking a huge time for evaluation. We therefore apply fuzzy reasoning to give effective operators more opportunity to search while keeping the overall population size constant. We propose a fuzzy self-tuning parallel genetic algorithm (FPGA) for optimization problems. In our test case FPGA there are four operators—crossover, mutation, sub-exchange, and sub-copy. These operators are modified using the eugenic concept under the assumption that the individuals with higher fitness values have a higher probability of breeding new better individuals. All operators are executed in each generation through parallel processing, but the populations of these operators are decided by fuzzy reasoning. The fuzzy reasoning senses the contributions of these operators, and then decides their population sizes. The contribution of each operator is defined as an accumulative increment of fitness value due to each operator's success in searching. We make the assumption that the operators that give higher contribution are more suitable for the typical optimization problem. The fuzzy reasoning is built under this concept and adjusts the population sizes in each generation. As a test case, a FPGA is applied to the optimization of the fuzzy rule set for a model reference adaptive control system. The simulation results show that the FPGA is better at finding optimal solutions than a traditional GA.  相似文献

The observation of alterations in crystal structure at ultra-low temperatures by X-ray diffraction, made possible by the realization of synchrotron radiation use after a quarter of this century, plays an important role in obtaining an understanding of the base structure of solids. For that, two types of3He-4He dilution refrigerators with a modified version of the top-loading facilities were installed at the BL-3C2 and 6C1 stations of the Photon Factory. In BL-3C2, the behaviors of lattice defects in solid helium have been studied by X-ray topography. In this note, the migrations of sub-boundaries in3He and4He single-crystals are reported as being the result of an annealing effect. After annealing hcp4He single crystals for 80min at 0.5K, no change in crystallographic orientations could be easily observed from white SR X-ray topographs. In the same type topographs of bcc3He single crystals after annealing for several hours at 03K, migration of sub-boundaries were conspicuously discerned.  相似文献

The process of corneal endothelial wound healing was studied using laser and tandem scanning confocal microscopy (LSCM and TSCM). Following transcorneal freeze (TCF) injury, rabbit corneas were observed using ex vivo LSCM and in vivo TSCM. LSCM revealed the intracellular actin filament organization which, stained with phalloidin-FITC, in migrating endothelial cells, transformed fibroblast-like cells, stroma keratocytes, and epithelial cells during wound healing in corneal tissue. The TSCM provided sequential spatial observation of morphologic changes from endothelium to epithelium of the cornea during in vivo cellular repair of wound healing noninvasively on the same cornea without animal sacrifice. Ex vivo LSCM supported the morphologic analysis of the in vivo TSCM observations.  相似文献

Catastrophic corrosion of iron by oxychlorination in hydrogen chloride gas containing 0–75 vol. % oxygen at 300–800°C has been investigated mainly by means of measurements of weight loss and the amount of sublimates. In pure hydrogen chloride gas, the corrosion behaviour was determined by the formation and sublimation of ferrous chloride. Addition of oxygen to hydrogen chloride gas greatly accelerated corrosion of iron due to the formation and sublimation of a low melting point volatile ferric chloride by oxychlorination reactions.  相似文献

Sample cables having a few types of defects (such as voids, contaminants, and protrusions) with more volume than usual, were prepared intentionally to clarify how these defects affect separately insulation characteristics under the presence/nonpresence of moisture. In the case of void inclusion, insulation characteristics are even more susceptible to ac voltage than impulse voltage, and the distribution of lifetimes is classified into a wornout failure type due to discharge deterioration. While protrusions and some other contaminants can greatly deteriorate original dielectric strength, these defects have a relatively slight effect on long-term endurance in the atmosphere. Therefore, the distribution of lifetimes is classified into an early-failure type. With reference to the influence of moisture, in a strong electric field (exceeding 15 kV/mm) and in a short-term region, there is an increase in harmfulness around defects with water absorbed, thereby inducing electrical trees and resulting in a breakdown. In the light electric field (no more than 15 kV/mm) and in a long-term region, harmfulness of defects will not deepen to a considerable extent, and the growth of water trees very possibly is influential.  相似文献
